CATECHOLAMINES probably influence cell proliferation by delaying cells in the premitotic phase1,2. Bullough and Laurence found that crude skin extracts contained a tissue-specific protein (chalone) which inhibited epidermal cell proliferation and that the action of this extract was augmented by adrenaline3. They later found that adrenaline alone (0.00025 µg/ml.) reduced epidermal mitotic activity in mouse ears by about 50% in vitro4.  相似文献

In Vietnam a green strain of tobacco mosaic virus was isolated having TIP 89°C (10 min) and causing systemic necrosis in tobacoo ‘Xanthi-nc’ and sometimes also inDatura stramonium. In symptomless tomato plants an elongated virus belonging apparently to the Carlavirus group (NL 630 nm) was found. In papaya trees showing severe symptoms of mosaic and/or ringspot elongated virus particles (NL 730 nm) were observed; this virus being apparently a member of the Potyvirus group, resembled as far as its symptoms in papaya are concerned, the papaya ringspot or the distortion ringspot. In Cambodia some young rubber trees showed malformed leaves (esp. edges and veins) with yellow discolorations along the veins. Such leaves contained elongated virus-like particles (rigid or slightly flexible) of various length (60 to 880 nm), so that their normal length (NL) could not be established precisely. Particles 120 to 150 nm long occurred very frequently.  相似文献

Prostaglandin biosynthesis from eicosa-8,11,14-trienoic acid in microsomes from the bovine vesicular gland is inhibited by the antioxidants α-naphthol, guaiacol, NDGA and propyl gallate. Prostaglandin biosynthesis in this system is not inhibited by the antioxidants BHT, DL-α-tocopherol and Trolox C. Arachidonic acid induced platelet aggregation is inhibited specifically by α-naphthol, guaiacol, NDGA and propyl gallate. Both arachidonic acid induced platelet aggregation and ADP induced platelet aggregation are inhibited non-specifically by the antioxidants BHT, DL-α-tocopherol and Trolox C. All antioxidants tested in this study inhibit soybean lipoxidase. Thus α-naphthol, NDGA and propyl gallate are non-specific inhibitors of both prostaglandin synthetase and soybean lipoxidase while BHT, DL-α-tocopherol and Trolox C are specific inhibitors of soybean lipoxidase alone.  相似文献

Properties of cholesteryl esters in pure and mixed monolayers   总被引:1,自引:0,他引:1  
The surface properties of cholesteryl palmitate, stearate, linoleate, linolenate, arachidonate, and acetate were investigated. Long-chain esters were not surface-active and force-area (pi-A) isotherms were not obtained. Unsaturated cholesteryl esters were oxidized at the air-water interface and these oxidized lipids gave expanded pi-A isotherms. Cholesteryl acetate had an equilibrium spreading pressure of 14.0 dynes/cm and formed a stable monolayer indistinguishable from cholesterol below that surface pressure. Cholesteryl linoleate formed mixed monolayers with surface-active lipids, and the amount of cholesteryl linoleate in the monolayer depended both on its solubility in the other lipid and on the surface pressure. Even at moderate surface pressures cholesteryl linoleate was extruded from the monolayer into a bulk phase. Cholesteryl acetate exhibited the well-known condensing effect of cholesterol in mixed monolayers with egg lecithin.  相似文献

Prostaglandin biosynthesis from eicosa-8,11,14-trienoic acid in microsomes from bovine seminal vesicles is inhibited by acetylenic acids. Octadeca-6,9,12-triynoic acid and eicosa-8,11,14-triynoic acid are the most potent inhibitors. These acids both contain an ω-8 methylene group. Within the 20-carbon acetylenic acid series, inhibition decreases in the sequence eicosa-8,11,14-triynoic acid > eicosa-7,10,13-triynoic acid > eicosa-5,8,11-triynoic acid. Furthermore, eicosa-8,11,14-triynoic acid is a more potent inhibitor of arachidonic acid induced platelet aggregation than either eicosa-7,10,13-triynoic acid or eicosa-5,8,11-triynoic acid. The ω-8 methylene group is not the only determinent of inhibitory potency since docosa-10,13,16-triynoic acid is less potent than its 18 and 20 carbon analogs and all of these acids have an ω-8 methylene group.  相似文献

Antisera were raised against degrading amyloid fibrils isolated from the heart of a patient with senile cardiac amyloidosis (SCA), and from a medullary carcinoma of the thyroid (MCT). The antisera were absorbed and used in indirect immunofluorescence to identify an amyloid fibril protein (ASCA) in heart tissue from patients with senile cardiac amyloidosis and to identify the amyloid fibril protein (AMCT) found in association with medullary carcinomas of the thyroid. Absorbed anti-ASCA antiserum did not react with normal tissue such as heart, liver, spleen, and striated muscle, or with amyloid tissue known to contain amyloid fibril proteins AA, AlambdaI, AlambdaIV, AlambdaV, AMCT or with pancreatic tissue containing islet amyloid deposits. The reactions with senile amyloid he,rt tissue could be blocked completely by degraded amyloid fibrils extracted from senile amyloid heart tissue or by amyloid fibril protein ASCA isolated from such fibrils. The anti-AMCT antiserum showed a similar specific reaction restricted to amyloid associated with MCT. In addition, antisera specific for amyloid fibril proteins AA, AlambdaI, AlambdaIV, and AlambdaV failed to react with senile cardiac amyloid, pancreatic islet amyloid, or medullary thyroid amyloid.  相似文献

The effect of methional on prostaglandin biosynthesis from 5,8,11,14-eicosatetraenoic acid was studied with microsomes from both bovine vesicular glands (BVG) and sheep vesicular glands (SVG). Ethylene was identified when methional was added to the fatty acid-microsome incubation systems showing that oxygen centered radicals such as hydroxyl radical were generated during incubation. A low methional level, 1 mM, enhanced the rate of prostaglandin biosynthesis in both BVG and SVG. A high methional level, 10 mM, inhibited prostaglandin biosynthesis in both BVG alone and SVG solubilized with 1% Tween 20. The inhibitory effect of 10 mM methional was reversed by lyophilization. These data suggest that oxygen centered radicals are used in prostaglandin biosynthesis even though they inactivate the enzyme complex.  相似文献

Summary Human-foreskin fibroblast (HF) and guinea-pig aorta smooth-muscle (SM) cultures were treated with several saturated and unsaturated fatty acids. Relative plating efficiencies were used to determine the proliferative response to each treatment. At low concentrations (16 to 18 μm), proliferation in HF cultures was inhibited by 8,11,14-eicosatrienoic acid (20:3), and stimulated by both 5,8,11,14-eicosatetraenoic acid (20:4) and 9-octadecenoic acid (18:1). At these levels, proliferation in SM cultures was unchanged by 20:3, inhibited by 20:4, and enhanced by 18:1. At higher concentrations (80 to 90 μm), HF cultures were inhibited by all three unsaturated fatty acids. At these same concentrations, proliferation in SM cultures was inhibited by 20:3 and 20:4, whereas 18:1 continued to stimulate proliferation. Thus proliferative response was a specific effect of the fatty acid used, its concentration, and the cell line involved. Further treatment of SM cultures by tetradecanoic acid (14:0), hexadecanoic acid (16:0), and octadecanoic acid (18:0) showed that their relative abilities to inhibit cell proliferation increased with increasing chain length. Concentrations required for the effective inhibition of proliferation in SM cultures by 14:0, 16:0 and 18:0 were 220 μm, 95μm and 18μm, respectively. The fatty acids used in these studies are all endogenous components of sera used as growth supplements in in vitro systems. Their roles as prostaglandin and hydroperoxy fatty-acid precursors (20:3 and 20:4), inhibitors of prostaglandin biosynthesis (18:1), or as calcium ionophores (14:0, 16:0, and 18:0) may allow them to function as endogenous controls of cell proliferation.
