Summary Comparison of transintegumental membrane permeability and partition coefficients of selected nonelectrolytes was attempted to correlate the parameters of lipid solubility, hydrophilicity, and membrane permeation in male and female schistosomes (parasites of the portal venous tributaries of man). Surface permation (measured by the triple isotope technique) and octanol/water partition coefficients were determined for 17 compounds (acetamide, aminopyrine, antipyrine, benzyl alcohol, butanol, caffeine, ethanol, ethylene glycol, glycerol, inosine, mannitol, methanol, polyethylene glycol, propylene glycol, sucrose, thiourea, and urea).Linear regression analyses comparing the logarithm of the partition coefficient to transintegumental uptakes indicate a positive correlation in both sexes:R=0.76 (P<0.001) for males, andR=0.77 (P<0.001) for females. Similarly, linear regression analyses comparing hydrogen bond number with the logarithm of tissue uptake index demonstrate a high (negative) correlation in both males (R=–0.85,P<0.001) and females (R=–0.90,P<0.001). The male and female schistosomes showed no statistically significant differences in correlation of these parameters. Surface permeation was the same in male and female schistosomes, suggesting that male-female variations in integumental uptake rates previously observed may be restricted to metabolites which enter by way of a selective carrier system.  相似文献

The rate of phosphorylation of 2-deoxy-D-glucose (2dGlc) was determined by incubating Schistosoma mansoni in vitro in [3H]2-deoxy-D-glucose; 60 sec after exposure to the [3H]dGlc, [14C]dGlc was added to the medium, and metabolic activity was arrested at 2 min by immersion of the tissue in ice-cold silicone oil. Column chromatographic separation of the neutral [3H]- and [14C]dGlc from the [3H]- and [14C]2-deoxy-D-glucose-6-phosphate permitted estimation of the quantity of [3H]dGlc phosphorylated in 2 min, and the proportion of [14C]dGlc phosphorylated in 1 min; thus a phosphorylation rate was determined from a single tissue sample. In male schistosomes derived from mouse infections 4.4 +/- 0.8% of the dGlc was phosphorylated each minute, and 4.2 +/- 0.9% in the females. Lower rates of phosphorylation were measured in schistosomes taken from hamsters where males phosphorylated 2.4 +/- 1.1% of the dGlc each minute, and in females 2.7 +/- 1.0%. These studies suggest the high rate of hexose utilization by schistosomes compares to the conscious rat brain, where 11% of the dGlc is phosphorylated each minute.  相似文献

Hymenolepis diminuta is able to acquire plasma-borne glucose 3-O-[14C]methylglucose in vivo. Free glucose concentrations estimated for this helminth in vivo are comparable to that of the host intestine. Both in vivo and in vitro examinations indicate that the scolex-neck regions (first quartile) of this tapeworm have the highest glucose content, and an anterior-posterior gradient along the second, third, and fourth quartiles was observed. Substrate concentration was rate affecting for glucose utilization rates (measured as substrate depletion from the medium in vitro). Glucose utilization per minute exceeds glucose content by a factor of more than 5. The half-life of glucose was about 10 sec, emphasizing that sugar metabolism is a very rapid process. In addition, utilization was highest in the first quartile and decreased in succession in the second, third, and fourth quartiles. It is concluded that while the exogenous glucose concentration remains stable, regional differences in glucose utilization rates are linked (R = 0.98; P less than 0.01) to free glucose content in H. diminuta.  相似文献
