The aims of this study were to evaluate the time-course of the familiarization process associated with a test of multiple sprint running performance and to determine the reliability of various performance indices once familiarization had been established. Eleven physically active men (mean age: 21 +/- 2 years) completed 4 multiple sprint running trials (12 x 30 m; repeated at 35-s intervals) with 7 days between trials. All testing was conducted indoors, and times were recorded by twin-beam photocells. Results revealed no apparent learning effects as evidenced by no significant (p > 0.05) between-trial differences in measures of fastest or mean 30-m sprint time. Within-subject test-retest reliability determined over 4 trials by coefficient of variation (CV) and intraclass correlation coefficient (ICC) showed excellent reliability for measures of fastest and mean sprint times (CV range: 1.34-2.24%; ICC range: 0.79-0.94). Pre- and posttrial blood lactate concentrations showed good reliability when judged in context with typical values (CV range: 12.08-18.21%; ICC range: 0.72-0.78). In contrast, and in line with previous research, fatigue data showed much greater variability (CV: 26.43%; ICC: 0.66). The results of this study suggest that high degrees of test-retest reliability can be obtained in many multiple sprint running indices without the need for prior familiarization. 相似文献

Toll-like receptors (TLRs) serve to initiate inflammatory signalling in response to the detection of conserved microbial molecules or products of host tissue damage. Recent evidence suggests that TLR-signalling plays a considerable role in a number of inflammatory diseases, including atherosclerosis and arthritis. Agents which modulate TLR-signalling are, therefore, receiving interest in terms of their potential to modify inflammatory disease processes. One such family of molecules, the oxidised phospholipids (OxPLs), which are formed as a result of inflammatory events and accumulate at sites of chronic inflammation, have been shown to modulate TLR-signalling in both in vitro and in vivo systems. As the interaction between OxPLs and TLRs may play a significant role in chronic inflammatory disease processes, consideration is given in this review to the potential role of OxPLs in the regulation of TLR-signalling. 相似文献

The first step in the olfactory perception is the activation by odorants of sensory neurones in the olfactory epithelium. In humans, this sensory epithelium is located at 2 narrow passages, the olfactory clefts, at the upper part of the nasal cavities. Little is known about the physiology of these clefts. We examined, in 34 patients, the impact of obstructed clefts upon detection and postlearning identification of 5 odorants. The location and extension of the obstructions were assessed using endoscopy, CT scans, and MRI. The inflammatory obstruction was usually bilateral, extending anteroposteriorly, and confined to the clefts, with no sign of obstruction or any inflammatory disease in the rest of the nasal cavities and sinuses. When tested with 5 odorants, these patients showed greatly impaired olfaction compared with a group of 73 normosmic subjects. The majority of these 34 patients had sensory deficits equivalent to that found in another group of 41 congenital anosmic patients, where inspection with MRI indicated the lack of olfactory bulbs. This study demonstrates that the olfactory clefts, in human, function as an entity that is different from other regions of the nasal cavity and is the target for local inflammatory events that are apparently not responding to corticoid and antibiotic treatments. 相似文献

The probiotic Escherichia coli strain Nissle 1917 (EcN) has been used for decades in human medicine in Central Europe for the treatment and prevention of intestinal disorders and diseases. However, the molecular mechanisms underlying its beneficial effects are only partially understood. To identify molecular responses induced by EcN that might contribute to its probiotic properties polarized T84 cells were investigated employing DNA microarrays, quantitative RT-PCR, Western blotting, immunofluorescence and specific protein kinase C (PKC) inhibitors. Polarized T84 epithelial cell monolayers were used as a model to monitor barrier disruption by infection with the enteropathogenic E. coli (EPEC) strain E2348/69. Co-incubation of EPEC with EcN or addition of EcN following EPEC infection abolished barrier disruption and, moreover, restored barrier integrity as monitored by transepithelial resistance. DNA-microarray analysis of T84 cells incubated with EcN identified 300+ genes exhibiting altered expression. EcN altered the expression, distribution of zonula occludens-2 (ZO-2) protein and of distinct PKC isotypes. ZO-2 expression was enhanced in parallel to its redistribution towards the cell boundaries. This study provides evidence that EcN induces an overriding signalling effect leading to restoration of a disrupted epithelial barrier. This is transmitted via silencing of PKCzeta and the redistribution of ZO-2. We suggest that these properties contribute to the reported efficacy in the treatment of inflammatory bowel diseases and in part rationalize the probiotic nature of EcN. 相似文献

The phenomenon of aposematism, or the pairing of antipredator defence with conspicuous or distinctive signals, serves as an excellent example of how traits act in concert to shape fitness. Not only does this complex phenotype require the integration of multiple traits, it alters the fitness pay‐offs of yet others. The protection offered by aposematism may, for example, reduce the costs associated with foraging or sexual display. Thus, well‐protected aposematic lineages should be bolder, more active and less likely to respond to perceived threats of predation than more cryptic lineages. Comparisons of differently coloured morphs of the polytypic strawberry poison frog (Oophaga pumilio) have supported the predicted behavioural correlates of aposematism, with the exception of those regarding responsiveness to simulated predators. We tested the key prediction that aposematic coloration will be associated with reduced sensitivity to predators in two polymorphic O. pumilio populations. The novel approach of studying polymorphic populations allowed us to assess the effect of colour in the absence of potentially confounding habitat differences. We found that colour was associated with the probability that a frog would attempt escape and the distance at which it fled, but only in one population, and not in the predicted direction. An overall comparison of the two populations superficially supported our predictions, but this pattern actually arose because frogs occupying higher perches were less likely to respond, a pattern that may reflect the value of high perches and the costs associated with returning to them after attempted escape. These results highlight the complexity of the relationship between predators and prey, the challenges associated with understanding how and why traits are correlated, and the intimate ties between behaviour and morphological evolution. 相似文献

Pseudomonas fluorescens is commonly considered a saprophytic rhizobacterium devoid of pathogenic potential. Nevertheless, the recurrent isolation of strains from clinical human cases could indicate the emergence of novel strains originating from the rhizosphere reservoir, which could be particularly resistant to the immune system and clinical treatment. The importance of type three secretion systems (T3SSs) in the related Pseudomonas aeruginosa nosocomial species and the occurrence of this secretion system in plant-associated P. fluorescens raise the question of whether clinical isolates may also harbor T3SSs. In this study, isolates associated with clinical infections and identified in hospitals as belonging to P. fluorescens were compared with fluorescent pseudomonads harboring T3SSs isolated from plants. Bacterial isolates were tested for (i) their genetic relationships based on their 16S rRNA phylogeny, (ii) the presence of T3SS genes by PCR, and (iii) their infectious potential on animals and plants under environmental or physiological temperature conditions. Two groups of bacteria were delineated among the clinical isolates. The first group encompassed thermotolerant (41°C) isolates from patients suffering from blood infections; these isolates were finally found to not belong to P. fluorescens but were closely related and harbored highly conserved T3SS genes belonging to the Ysc-T3SS family, like the T3SSs from P. aeruginosa. The second group encompassed isolates from patients suffering from cystic fibrosis; these isolates belonged to P. fluorescens and harbored T3SS genes belonging to the Hrp1-T3SS family found commonly in plant-associated P. fluorescens. 相似文献

The Extracellular A-loop of Dual Oxidases Affects the Specificity of Reactive Oxygen Species Release

NADPH oxidase (Nox) family proteins produce superoxide (O2⨪) directly by transferring an electron to molecular oxygen. Dual oxidases (Duoxes) also produce an O2⨪ intermediate, although the final species secreted by mature Duoxes is H2O2, suggesting that intramolecular O2⨪ dismutation or other mechanisms contribute to H2O2 release. We explored the structural determinants affecting reactive oxygen species formation by Duox enzymes. Duox2 showed O2⨪ leakage when mismatched with Duox activator 1 (DuoxA1). Duox2 released O2⨪ even in correctly matched combinations, including Duox2 + DuoxA2 and Duox2 + N-terminally tagged DuoxA2 regardless of the type or number of tags. Conversely, Duox1 did not release O2⨪ in any combination. Chimeric Duox2 possessing the A-loop of Duox1 showed no O2⨪ leakage; chimeric Duox1 possessing the A-loop of Duox2 released O2⨪. Moreover, Duox2 proteins possessing the A-loops of Nox1 or Nox5 co-expressed with DuoxA2 showed enhanced O2⨪ release, and Duox1 proteins possessing the A-loops of Nox1 or Nox5 co-expressed with DuoxA1 acquired O2⨪ leakage. Although we identified Duox1 A-loop residues (His1071, His1072, and Gly1074) important for reducing O2⨪ release, mutations of these residues to those of Duox2 failed to convert Duox1 to an O2⨪-releasing enzyme. Using immunoprecipitation and endoglycosidase H sensitivity assays, we found that the A-loop of Duoxes binds to DuoxA N termini, creating more stable, mature Duox-DuoxA complexes. In conclusion, the A-loops of both Duoxes support H2O2 production through interaction with corresponding activators, but complex formation between the Duox1 A-loop and DuoxA1 results in tighter control of H2O2 release by the enzyme complex. 相似文献
