Irradiation in the ultraviolet wavelength range is found to be up to 50% lower in the European summer compared to sites with comparable latitudes in New Zealand. We have developed a method to quantitatively attribute the causes for such differences between sites by analysis of spectra. We conclude that these large differences are caused mainly by differences in total ozone, cloudiness, aerosol loading and Sun-Earth separation. The relative contribution of clouds varies from year to year and it is site dependent. Averaged over several years we find a strong latitudinal gradient of the cloud impact within Europe, with much less cloud attenuation in southern Europe. Due to the differences in total ozone and aerosol loading, the UV-B levels are generally lower in Europe compared to New Zealand. It is likely that inter-hemispheric differences will change in coming decades due to a combination of changes in ozone concentrations, air pollution and cloudiness as a result of climate change. However, since the future evolution of these major parameters is highly uncertain, the magnitude and even the sign of such changes are not known yet. 相似文献

Abstract: Gerbils ( Meriones unguiculatus ) are known for their seizure sensitivity, which is dependent on an intact perforant path from the entorhinal cortex to the hippocampus. In contrast with other species, the perforant path in gerbils contains parvalbumin, a cytosolic high-affinity calcium-binding protein. Parvalbumin is known to be present in a subpopulation of GABA-containing neurons and is thought to be responsible for their physiological characteristics of fast spiking activity and lack of spike adaptation. Therefore, the question arose of whether this projection in gerbils is GABAergic or glutamatergic as in other species. In a first approach to this question, the effect of lesioning the origin of the perforant path, the entorhinal cortex, on levels of GABA and glutamate was determined by enzymatic-luminometric assay in single layers of the dentate gyrus of lyophilized brain sections. Parallel sections were cryofixed using an acidified acetone-formaldehyde mixture at -20°C for 48 h, and subsequently stained for parvalbumin immunocytochemistry. Seven days after ablation of the entorhinal cortex, parvalbumin staining was undetectable in the termination zone of the perforant path, the outer two-thirds of the stratum moleculare. In parallel, glutamate content was reduced to 80% of controls (and of the unoperated contralateral side) but unchanged in the inner third of the stratum moleculare and in stratum granulare. GABA content was not significantly altered by the lesion. From these results, we conclude that in the gerbil as in other species, the perforant path contains glutamate. The association of glutamate with parvalbumin suggests, however, that in the gerbil the calcium-dependent release of the transmitter is carefully controlled in the entorhinal perforant path. 相似文献

Phalaenopsis hybrids contain two 1,2-saturated pyrrolizidine monoesters, T-phalaenopsine (necine base trachelanthamidine) and its stereoisomer Is-phalaenopsine (necine base isoretronecanol). T-Phalaenopsine is the major alkaloid accounting for more than 90% of total alkaloid. About equal amounts of alkaloid were genuinely present as free base and its N-oxide. The structures were confirmed by GC-MS. The quantitative distribution of phalaenopsine in various organs and tissues of vegetative rosette plants and flowering plants revealed alkaloid in all tissues. The highest concentrations were found in young and developing tissues (e.g., root tips and young leaves), peripheral tissues (e.g., of flower stalks) and reproductive organs (flower buds and flowers). Within flowers, parts that usually attract insect visitors (e.g., labellum with colorful crests as well as column and pollinia) show the highest alkaloid levels. Tracer feeding experiments with (14)C-labeled putrecine revealed that in rosette plants the aerial roots were the sites of phalaenopsine biosynthesis. However active biosynthesis was only observed in roots still attached to the plant but not in excised roots. There is a slow but substantial translocation of newly synthesized alkaloid from the roots to other plant organs. A long-term tracer experiment revealed that phalaenopsine shows neither turnover nor degradation. The results are discussed in the context of a polyphyletic molecular origin of the biosynthetic pathways of pyrrolizidine alkaloids in various scattered angiosperm taxa. The ecological role of the so called non-toxic 1,2-saturated pyrrolizidine alkaloids is discussed in comparison to the pro-toxic 1,2-unsaturated pyrrolizidine alkaloids. Evidence from the plant-insect interphase is presented indicating a substantial role of the 1,2-saturated alkaloids in plant and insect defense. 相似文献

26S proteasomes are multisubunit protease complexes that play the central role in the ubiquitin-dependent protein degradation pathway. The proteolytically active core is formed by the 20S proteasome. Regulatory subunits, principally the 19S cap complex, confer the specificity towards ubiquitinated substrates and an ATP-dependence on proteolysis. Green fluorescence protein (GFP)-tagged versions of either an -subunit of the 20S core or an ATPase subunit of the 19S cap complex were functionally incorporated into the protease complex, thus allowing to monitor the subcellular distribution of 26S proteasomes in living yeast. Our localization studies suggest that proteasomal proteolysis mainly occurs at the nuclear envelope (NE)/rough ER. Implications of proteasomal functions at the NE/rough ER are discussed in the context of published work on ER degradation and with regard to possible targeting mechanisms. 相似文献

Properdin (FP) is an essential positive regulator of the complement alternative pathway (AP) providing stabilization of the C3 and C5 convertases, but its oligomeric nature challenges structural analysis. We describe here a novel FP deficiency (E244K) caused by a single point mutation which results in a very low level of AP activity. Recombinant FP E244K is monomeric, fails to support bacteriolysis, and binds weakly to C3 products. We compare this to a monomeric unit excised from oligomeric FP, which is also dysfunctional in bacteriolysis but binds the AP proconvertase, C3 convertase, C3 products and partially stabilizes the convertase. The crystal structure of such a FP-convertase complex suggests that the major contact between FP and the AP convertase is mediated by a single FP thrombospondin repeat and a small region in C3b. Small angle X-ray scattering indicates that FP E244K is trapped in a compact conformation preventing its oligomerization. Our studies demonstrate an essential role of FP oligomerization in vivo while our monomers enable detailed structural insight paving the way for novel modulators of complement. 相似文献

The formation of gametes is a key step in the life cycle of any sexually reproducing organism. In flowering plants, gametes develop in haploid structures termed gametophytes that comprise a few cells. The female gametophyte forms gametic cells and flanking accessory cells. During a screen for regulators of egg-cell fate, we isolated three mutants, lachesis (lis), clotho (clo) and atropos (ato), that show deregulated expression of an egg-cell marker. We have previously shown that, in lis mutants, which are defective for the splicing factor PRP4, accessory cells can differentiate gametic cell fate. Here, we show that CLOTHO/GAMETOPHYTIC FACTOR 1 (CLO/GFA1) is necessary for the restricted expression of egg- and central-cell fate and hence reproductive success. Surprisingly, infertile gametophytes can be expelled from the maternal ovule tissue, thereby preventing the needless allocation of maternal resources to sterile tissue. CLO/GFA1 encodes the Arabidopsis homologue of Snu114, a protein that is considered to be an essential component of the spliceosome. In agreement with their proposed role in pre-mRNA splicing, CLO/GFA1 and LIS co-localize to nuclear speckles. Our data also suggest that CLO/GFA1 is necessary for the tissue-specific expression of LIS. Furthermore, we demonstrate that ATO encodes the Arabidopsis homologue of SF3a60, a protein that has been implicated in pre-spliceosome formation. Our results thus establish that the restriction of gametic cell fate is specifically coupled to the function of various core spliceosomal components. 相似文献

The regional assessment of global change effects on plant phenology usually relies on local observations that need to be up-scaled. Therefore, methodological difficulties mostly related to data spatial resolution and congruency arise while performing broader-scale evaluations. Geostatiscs could be a useful tool to solve this type of problem, provided that a database with adequate spatial and temporal resolution is available. An assessment of variations in air temperature and plant phenology was carried out at the country level by using two German datasets regarding spring phenological phases of 15 plant species and air temperature. The data were collected from 1961-2002 at 1,279 and 675 sites, respectively. The annual mean air temperature in Germany was found to rise from 8.3 degrees C in the 1961-1990 period to 9.1 degrees C in the 1991-2002 term. The overall 15-species mean for the start of spring was found to be 6 days earlier in the latter period. The geostatistical analysis of the data revealed the suitability of Syringa vulgaris to be used as an indicator species to detect phenological changes in German forests. Moreover, their spatial patterns were found to be related to altitude and latitude. Therefore, geostatistics proved to be a useful tool to overcome some of the methodological problems related to the regional assessments of global change impacts on terrestrial ecosystems. 相似文献
