To investigate the activity of the regulatory region of the maize (Zea mays L.) proteinase inhibitor (mpi) gene, we transferred into rice (Oryza sativa L.) plants the –689/+197 (C1) fragment of the mpi genomic clone fused to either theuidA gene or a synthetic Bacillus thuringiensis cry1B gene. Although uidA and cry1B encode very different proteins consistent results were obtained from their respective histochemical and fluorometric and immunoblot detections in T3 transgenic rice lines. In response to mechanical wounding, a 4–5 fold increase in GUS activity and a Cry1B accumulation reaching 0.1–0.2% of total soluble proteins were observed from basal and undetectable levels respectively in leaf tissue. The establishment of the time-course of wound response in both systems revealed a maximum induction level 12–16 h after treatment. From both systems we also deduced that the C1 region is not active in pollen and seed endosperm. Three independent transformation events expressing cry1B under the control of the C1 region exhibited protection against striped stem borer damage and showed 100% mortality of second instar larvae 8 days after release. These results illustrate the first evidence that wound-inducible expression of a Bacillus thuringiensis endotoxin gene affords full protection to transgenic rice plants.  相似文献

We reconsider the deterministic haploid mutation–selection equation with two types. This is an ordinary differential equation that describes the type distribution (forward in time) in a population of infinite size. This paper establishes ancestral (random) structures inherent in this deterministic model. In a first step, we obtain a representation of the deterministic equation’s solution (and, in particular, of its equilibria) in terms of an ancestral process called the killed ancestral selection graph. This representation allows one to understand the bifurcations related to the error threshold phenomenon from a genealogical point of view. Next, we characterise the ancestral type distribution by means of the pruned lookdown ancestral selection graph and study its properties at equilibrium. We also provide an alternative characterisation in terms of a piecewise-deterministic Markov process. Throughout, emphasis is on the underlying dualities as well as on explicit results.

Females of socially monogamous bird species frequently accept or solicit copulations from males other than their social mates. At the interspecific level, it has been proposed that species with few or no sexual plumage differences have lower levels of extra-pair fertilizations (EPFs) than more dichromatic species. This is because it is easier for females to assess the quality of extra-pair mates in dichromatic than monochromatic species. To test this, by using genetic profiling, we compared the occurrence of extra-pair young in nests of Tree Sparrows Passer montanus , a monochromatic species, in Switzerland and Spain, with published estimates for the House Sparrow Passer domesticus , a closely related dichromatic species. We found that 25% (10/40) of Tree Sparrow broods in Spain and 23% (8/35) in Switzerland had at least one extra-pair offspring, and that 8% (12/151) of nestlings in Spain and 10% (12/114) in Switzerland were sired by extra-pair mates. These proportions did not differ significantly between the two populations. Tree Sparrow EPF rates did not differ significantly from available data for the House Sparrow, either in the proportion of broods with EPF young (24%, 18/75 in Tree vs. 26%, 92/359 in House Sparrows) or the overall proportion of extra-pair young (9%, 24/265 in Tree vs. 11%, 142/1110 in House Sparrows). Our results suggest that a close taxonomic relationship and similarities in behaviour and ecology may be more important in determining levels of EPF in these species than sexual phenotypic differences.  相似文献

Four decades ago, it was proposed that environmental sex determination (ESD) evolves when individual fitness depends on the environment in a sex‐specific fashion—a form of condition‐dependent sex allocation. Many biological processes have been hypothesized to drive this sex asymmetry, yet a general explanation for the evolution of sex‐determining mechanisms remains elusive. Here, we develop a mathematical model for a novel hypothesis of the evolution of ESD, and provide a first empirical test using data across turtles. ESD is favored when the sex‐determining environment affects annual survival rates equivalently in males and females, and males and females mature at different ages. We compare this hypothesis to alternative hypotheses, and demonstrate how it captures a crucially different process. This maturation process arises naturally from common life histories and applies more broadly to condition‐dependent sex allocation. Therefore, it has widespread implications for animal taxa. Across turtle species, ESD is associated with greater sex differences in the age at maturity compared to species without ESD, as predicted by our hypothesis. However, the effect is not statistically significant and will require expanded empirical investigation. Given variation among taxa in sex‐specific age at maturity, our survival‐to‐maturity hypothesis may capture common selective forces on sex‐determining mechanisms.  相似文献

Bordetella bronchiseptica produces respiratory disease primarily in mammals including humans. Although a considerably amount of research has been generated regarding lipopolysaccharide (LPS) role during infection and stimulating innate and adaptive immune response, mechanisms involved in LPS synthesis are still unknown. In this context we searched in B. bronchiseptica genome for putative glycosyltransferases. We found possible genes codifying for enzymes involved in sugar substitution of the LPS structure. We decided to analyse BB3394 to BB3400 genes, closed to a previously described LPS biosynthetic locus in B. pertussis. Particularly, conservation of BB3394 in sequenced B. bronchiseptica genomes suggests the importance of this gene for bacteria normal physiology. Deletion of BB3394 abolished resistance to naive serum as described for other LPS mutants. When purified LPS was analyzed, differences in the LPS core structure were found. Particularly, a GalNA branched sugar substitution in the core was absent in the LPS obtained from BB3394 deletion mutant. Absence of GalNA in core LPS alters immune response in vivo but is able to induce protective response against B. bronchiseptica infection.  相似文献

The study of the neutral and/or selective processes driving genetic variation in natural populations is central to determine the evolutionary history of species and lineages and understand how they interact with different historical and contemporary components of landscape heterogeneity. Here, we combine nuclear and mitochondrial data to study the processes shaping genetic divergence in the Mediterranean esparto grasshopper (Ramburiella hispanica). Our analyses revealed the presence of three main lineages, two in Europe that split in the Early-Middle Pleistocene and one in North Africa that diverged from the two European ones after the Messinian. Lineage-specific potential distribution models and tests of environmental niche differentiation suggest that the phylogeographic structure of the species was driven by allopatric divergence due to the re-opening of the Gibraltar strait at the end of the Messinian (Europe–Africa split) and population fragmentation in geographically isolated Pleistocene climatic refugia (European split). Although we found no evidence for environment as an important driver of genetic divergence at the onset of lineage formation, our analyses considering the spatial distribution of populations and different aspects of landscape composition suggest that genetic differentiation at mitochondrial loci was largely explained by environmental dissimilarity, whereas resistance-based estimates of geographical distance were the only predictors of genetic differentiation at nuclear markers. Overall, our study shows that although historical factors have largely shaped concordant range-wide patterns of mitonuclear genetic structure in the esparto grasshopper, different contemporary processes (neutral gene flow vs. environmental-based selection) seem to be governing the spatial distribution of genetic variation in the two genomes.  相似文献

The narrow genetic base of commercial arabica resulting from intensive selection for quality during domestication and self-pollination has been well documented, raising the need for new diverse germplasm sources. Beans of 232 diverse arabica coffee accessions originating from 27 countries were harvested from the germplasm collection at CATIE, Costa Rica. Substantial variation was observed for bean morphology including 100 bean weight, bean length, width, thickness and bulk density. Non-volatiles including caffeine and trigonelline were analysed and showed larger variation in range than has previously been reported. Results of targeted analysis of 18 volatiles from 35 accessions also showed significant variation, with coefficients of variation from 140% for 4-vinylguaiacol to 62% for geraniol. There were strong correlations between some volatile compounds, suggesting that representative volatiles used in selection would save analytical costs. However, no strong correlation was found between bean morphology and the levels of non-volatile or volatile compounds, implying that it is difficult to select for low or high composition of these compounds based on bean physical characteristics. Utilizing the large variation observed for bean morphology and biochemical traits, it should be possible to select for desirable combinations of traits in arabica coffee breeding.  相似文献

During Trypanosoma cruzi cell invasion, signal transduction pathways are triggered in parasite and host cells, leading to a rise in intracellular Ca(2+) concentration. We posed the question whether calcineurin (CaN), in particular the functional regulatory subunit CaNB, a Ca(2+)-binding EF-hand protein, was expressed in T. cruzi and whether it played a role in cell invasion. Here we report the cloning and characterization of CL strain CaNB gene, as well as the participation of CaNB in cell invasion. Treatment of metacyclic trypomastigotes (MT) or tissue-culture trypomastigotes (TCT) with the CaN inhibitors cyclosporin or cypermethrin strongly inhibited (62-64%) their entry into HeLa cells. In assays using anti-phospho-serine/threonine antibodies, a few proteins of MT were found to be dephosphorylated in a manner inhibitable by cyclosporin upon exposure to HeLa cell extract. The phosphatase activity of CaN was detected by a biochemical approach in both MT and TCT. Treatment of parasites with antisense phosphorothioate oligonucleotides directed to TcCaNB-CL, which reduced the expression of TcCaNB and affected TcCaN activity, resulted in approximately 50% inhibition of HeLa cell entry by MT or TCT. Given that TcCaNB-CL may play a key role in cell invasion and differs considerably in its primary structure from the human CaNB, it might be considered as a potential chemotherapeutic target.  相似文献
