The aim of the present study was to evaluate the response of adrenocorticotropin ([ACTH]) and growth hormone ([GH]) concentrations
to a typical aerobic swimming set during a training season. Nine top-level male endurance swimmers (age range 17–23 years)
were tested during three training sessions occurring 6, 12 and 18 weeks after the beginning of the season. During each session,
after a standard warm-up, the swimmers performed a training set of 15 × 200-m freestyle, with 20 s of rest between repetitions,
at a predetermined individual speed. Blood samples were collected before warm-up and at the end of the training set. A few
days before each session, the individual swimming velocity corresponding to the 4 mmol · l−1 blood lactate concentration (v
4) was assessed as a standard of aerobic performance. Aerobic training affected v
4 levels, which were highest 18 weeks after the beginning of the season; at the same time, while [ACTH] response was attenuated,
[GH] response was enhanced. These results could be considered as adaptations to the exercise intensity. In our training programme,
these adaptations seemed to have occurred between the 12th and 18th weeks of the training season.

In cell cultures of human lung fibroblasts, we found that oxidized LDL (oxLDL), after 24-h treatment, stimulated arachidonic acid release. A putative role for phospholipases A(2) and MAPK activities in this process was postulated. Consequently, we studied the contribution of either Ca(2+)-dependent, cytosolic phospholipase A(2) (cPLA(2)) or Ca(2+)-independent phospholipase A(2) (iPLA(2)), and the role of the MAP kinase family in oxLDL toxicity to fibroblastic cells in vitro. Activation of extracellular signal-regulated kinases ERK1/2, p38 and c-Jun NH(2)-terminal kinase (JNK) was also assessed with Western blotting. Compared with cellular samples untreated or treated with native LDL, treatment with oxLDL (50-100 microM hydroperoxides) for 24 h significantly increased the levels of either cPLA(2) protein expression or constitutively phosphorylated cPLA(2) protein; in addition we observed enzyme translocation to membranes. iPLA(2) activity was not stimulated by oxLDL. Arachidonic acid release appeared to be associated with phosphorylation of ERK1/2 which was significantly enhanced in a dose-dependent manner whereas no activation of p38 and JNKs was found, indicating that these MAPKs are not involved in mediating the maximal oxLDL response. Western blotting on subcellular fractions and confocal microscopy analyses confirmed an increase in 15-lipoxygenase (15-LO) protein expression and translocation upon activation. A significant increase of cyclooxygenase-2 expression into membrane fraction was also found. Collectively, the data presented link the stimulation of ERK-cPLA(2)-15-LO pathway by oxLDL to the prooxidant mechanism of the lipoprotein complex. We report the application of an integrated computational approach for biomolecular structure determination at a low resolution. In particular, a neural network is trained to predict the spatial proximity of C-alpha atoms that are less than a given threshold apart, whereas a Kalman filter algorithm is employed to outline the biomolecular fold, with a constraints set that includes these pairwise atomic distances, and the distances and angles that define the structure as it is known from the protein's sequence. The results for Crambin demonstrate that this integrated approach is useful for molecular structure prediction at a low resolution and may also complement existing experimental distance data for a protein structure determination. © 1996 John Wiley & Sons, Inc. 相似文献

The palynological characterisation of 157 honey samples from three northwest regions of Argentina (Prepuna, Yungas and Chaco) are presented to determine their botanical origin and species associations to be able to define their geographic origin. Samples were harvested during 2003–2011 and processed by means of melissopalynological conventional techniques. One-hundred and nine pollen types were identified. Representative pollen types with a frequency of occurrence greater than 50% in descending order of importance are: Salix humboldtiana, Allophylus edulis, Baccharis, Solanaceae, Eucalyptus, Schinus, Brassicaceae, Papilionoideae, Celtis, Scutia/Condalia-type and Parapiptadenia excelsa. The most important monofloral honeys are from the following: Salix humboldtiana, Scutia/Condalia-type, Allophylus edulis, Baccharis, Blepharocalyx salicifolius, Gleditsia amorphoides, Myrtaceae, Sicyos, Ziziphus mistol, Schinopsis-type, Agonandra excelsa, Anadenathera colubrina, Mimosa, all of them native species, and among introduced species are Eucalyptus, Citrus and Tithonia. Three apicultural zones and their corresponding pollen association indicators were determined: Zone I, Prepuna: Arquita trichocarpa, Prosopis ferox, Schinus areira, Baccharis, Buddleja and Mutisieae; Zone II, Yungas: Myrtaceae, Parapiptadenia excelsa, Baccharis, Salix humboldtiana, Allophylus edulis, Scutia/Condalia-type and Zanthoxylum coco; Zone III, transitional area Yungas-Chaco: Prosopis, Salix humboldtiana, Schinus, Anadenanthera colubrina and Allophylus edulis. 相似文献

Leishmaniasis and Chagas disease afflict the poorest countries in the world. The Brazilian flora represents a rich source for the screening of potential antiparasitic compounds. In this work, we tested the total alkaloid and ethanol extracts of nine different plants from Brazilian families which produce isoquinoline alkaloids, to determine their in vitro antiparasitic effect against L. chagasi and T. cruzi parasites. Promastigotes of L. chagasi were shown to be susceptible only to the total alkaloid extracts of A. crassiflora (EC50 value = 24.89 microg/ml), A. coriacea (EC50 value = 41.60 microg/ml), C. ovalifolia (EC50 value = 63.88 microg/ml) and G. australis (EC50 value = 37.88 microg/ml). Except for the G. australis total alkaloids, all the three extracts presented a considerable activity when tested against intracellular amastigotes. The most effective alkaloid extracts were those from A. crassiflora and C. ovalifolia, which reduced the number of infected macrophages at 25 microg/ml by 86.1% and 89.8%, respectively. Among the 18 tested extracts, 16 showed anti-Trypanosoma activity. Eight extracts (A. crassiflora, A. coriacea, C. ovalifolia, D. furfuracea, D. lanceolata, S. guianensis, X. emarginata and G. australis) were the most effective against the trypomastigotes, killing approximately 100% of the parasites at the maximal concentration of 100 microg/ml. Cytotoxicity against mammalian cells was evaluated for all extracts, but potential ones showed little or no cytotoxicity and a considerable antiparasitic effect, including D. furfuracea, D. lanceolata, G. australis, S. guianensis and X. emarginata. The dystrophin-associated protein complex (DAPC) is essential for skeletal muscle, and the lack of dystrophin in Duchenne muscular dystrophy results in a reduction of DAPC components such as syntrophins and in fiber necrosis. By anchoring various molecules, the syntrophins may confer a role in cell signaling to the DAPC. Calcium disorders and abnormally elevated cation influx in dystrophic muscle cells have suggested that the DAPC regulates some sarcolemmal cationic channels. We demonstrated previously that mini-dystrophin and α1-syntrophin restore normal cation entry in dystrophin-deficient myotubes and that sarcolemmal TRPC1 channels associate with dystrophin and the bound PDZ domain of α1-syntrophin. This study shows that small interfering RNA (siRNA) silencing of α1-syntrophin dysregulated cation influx in myotubes. Moreover, deletion of the PDZ-containing domain prevented restoration of normal cation entry by α1-syntrophin transfection in dystrophin-deficient myotubes. TRPC1 and TRPC4 channels are expressed at the sarcolemma of muscle cells; forced expression or siRNA silencing showed that cation influx regulated by α1-syntrophin is supported by TRPC1 and TRPC4. A molecular association was found between TRPC1 and TRPC4 channels and the α1-syntrophin-dystrophin complex. TRPC1 and TRPC4 channels may form sarcolemmal channels anchored to the DAPC, and α1-syntrophin is necessary to maintain the normal regulation of TRPC-supported cation entry in skeletal muscle. Cation channels with DAPC form a signaling complex that modulates cation entry and may be crucial for normal calcium homeostasis in skeletal muscles. 相似文献
