Activation energy and phospholipid requirements of membrane-bound adenosine triphosphatases 总被引:1,自引:0,他引:1
In order to evaluate the role of lipids in the function of membrane ATPase reactions, the apparent activation energies of membrane-bound (Na+ + K+)-ATPase and membrane-bound Mg2+-ATPase have been measured under conditions frequently supposed to alter the membrane lipids in vitro.In the case of (Na+ + K+)-ATPase, the untreated enzyme was shown to have two different activation energies as shown by an Arrhenius plot comprising two straight lines which intersect at the “critical temperature.” Treatment of the preparation with detergents or with phospholipase C causes some alteration in the spécifie activity of the enzyme but did not significantly alter the activation energies or the critical temperature. After treatment with phospholipase A, however, the Arrhenius plot appeared linear over the entire temperature range studied. Subsequent treatment of phospholipase A-treated preparations with phosphatidylserine restored the control response.Conversely, untreated preparations of Mg2+-ATPase give an Arrhenius plot which is neither linear nor composed of two intersecting straight lines. This plot, which we regard as curvilinear, does not permit a unique value of the activation energy to be determined. The shape of this plot is unaltered by detergent or by treatment with phospholipase C. In contrast to (Na+ + K+)-ATPase, it is also unaffected by treat-with phospholipase A or phospholipase A followed by phosphatidylserine.We conclude that although (Na+ + K+)-ATPase and Mg2+-ATPase are frequently closely associated in many membranes, their functions involve the presence of different membrane lipids. 相似文献

The first experiment was a selection procedure designed to increase the receptivity of Drosophila melanogaster females to males whose courtship had become less effective through attenuation of wing vibration. The clearest response was in the percentage of females receptive per generation, but changes were also detected in courtship duration and latency to courtship. The two selected and two control lines were hybridized in experiment 2, and the hybrid of the selected lines (Selx) was found to maintain the adaptation to selection of its parents. The relative receptivity of all female types was compared in experiment 3. The results demonstrated that adaptations had occurred in the females' courtship processing, rather than merely in the males' ability to court. 相似文献

Summary The fine structure of the various hormone-producing cell types (with the exclusion of the prolactin cells) in the pituitary gland (pars distalis) of migratory sockeye salmon is described. All fish were in an advanced stage of sexual maturation. In the proximal pars distalis five cell types were distinguished: growth hormone cells, ACTH cells, gonadotrops, vesicular cells, and chromophobe cells. Gonadotrops were also found throughout the rostral pars distalis. A conspicuous feature of the gonadotrops was the presence of two kinds of secretory inclusions: small electron-dense granules (200–375 m) and large, relatively electron-translucent globules (400–2 000 m). The large vesicular cells, so called because of their conspicuous vesicular endoplasmic reticulum, were numerous and often appeared to contain some small granules. It is argued that they may represent a second type of gonadotropic cell, which, in earlier stages of gonad development, contains many granules but becomes largely degranulated near the time of reproduction when the other gonadotrops (globular gonadotrops) abound. The chromophobes, which were smaller and far less abundant than the vesicular cells, also appeared to contain small granules (120–280 m). They are probably thyrotrops.The assistance of Mr. S. Four microbiological assay procedures (Escherichia coli, Lactobacillus leichmanni, Euglena gracilis, and Ochromonas malhamensis) for the vitamin B(12) content of rat liver and kidney tissue were compared. Significant differences in the growth response for portions of the same sample based on the same standard preparations were noted. The liver values with E. coli were strikingly higher than those obtained with the other microorganisms, about which there was rather general agreement. With kidney tissue, E. coli again gave higher values, but not as significantly high as those obtained with L. leichmannii and E. gracilis. O. malhamensis values were significantly lower. These differences were greater than were expected and could not be explained by the differences of specificity and sensitivity of the assay organisms. Several methods of sample preparation, the possible presence of an alkali-heat-stable factor, different methods of sterilization of the sample plus medium, and different chromatographic procedures were explored in an attempt to find explanations for the variations in growth response, with the thought that these various methods would clarify in what form or forms the free and bound vitamin B(12) might exist in the samples.
