As a part of the study of the bacteriophage T4-induced deoxyribonucleotide synthetase complex, an investigation has been made of the T4 ribonucleoside diphosphate reductases formed by a series of mutants of nrdA and B, the genes coding, respectively, for the alpha 2 and beta 2 subunits of the enzyme. dATP affinity columns were used to isolate the enzyme by a single-step procedure. The molecular weights of the alpha and beta chains have been found to be 84,000 and 43,500, respectively, by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Since alpha 2 beta 2 is bound to dATP affinity columns through allosteric effector sites on alpha 2, it is possible to monitor the binding of beta 2 to alpha 2. dTTP- and ATP-Sepharose columns did not bind T4 alpha 2 beta 2, although the corresponding nucleoside triphosphates are effectors of the enzyme and although the alpha 2 subunit of the host enzyme binds to these columns. Missense mutants of nrdA and B forming alpha 2 and beta 2 subunits that lacked catalytic activity but retained the ability to form the alpha 2 beta 2 complex have been described. The 50,000-dalton fragment formed by an amber mutant of nrdA did not bind to the dATP affinity column, providing evidence that a region of the carboxyl-terminal segment of the alpha chain is required for retention. The beta 2 subunit appears to protect the alpha 2 protein. On infection by nrdB mutants not forming beta 2, the alpha protein chain was cleaved specifically to form 3 protein chains of 61,000, 57,000, and 24,500 daltons, which retain the ability to bind to dATP-Sepharose. Some effects of mutation on the interaction of the alpha and beta chains of the enzyme with the deoxyribonucleotide synthetase complex have been examined.  相似文献

The aim was to control specifically the development of the mycotic and bacterial components of the microflora. Effective control of the microflora and temperature was achieved. A small circular extrachromosomal DNA of the flagellate protozoan Euglena gracilis has been characterized as having a contour length of 11.3 kb, with a consistent restriction map. The buoyant density (rho = 1.717) and melting temperature (tm = 89 degrees C) both indicate a base content of 59% G + C. The DNA is found in both wild-type cells and those lacking plastids. The copy number is estimated to be about 1000.  相似文献

Summary The fine structure of erythrocytic stages of Plasmodium knowlesi was compared with that of the same parasite isolated from its host cell by a saponin technique. Rhesus monkeys experimentally infected with Plasmodium knowlesi were the source of parasitized red cells. The erythrocytic stages of this Plasmodium showed all the organelles described in other mammalian forms; the nucleus lacked a typical nucleolus but contained a cluster of granules. P. knowlesi did not have protozoan-type mitochondria as do the avian and reptilian forms, but had double-membrane-bounded bodies as observed in other mammalian malarial parasites.The isolation procedure caused a slight swelling of the parasite, but in general, the structure and structural relationships of the parasite were preserved. However, the isolation technique gave a new insight into the connection of the host cell cytoplasm with the large, so-called food vacuoles of the parasite. The parasite freed from its host cell showed clear spaces where the large vacuoles had been. The content of these vacuoles had been removed together with the red cell cytoplasm. As the nature of the isolation procedure precluded any disruption of the parasite itself, these findings support our view that the vacuoles are not true food vacuoles. If these were true food vacuoles, they would be completely enclosed by a parasite membrane within the parasite cytoplasm. However, we have demonstrated that they represent extensions of host cell cytoplasm in direct communication with the rest of the red cell. The outer membrane surrounding the intra-erythrocytic parasites disappeared after isolation of the parasite from the host cell. This strongly suggested that the outer membrane is of host cell origin. Partially purified nerve varicosities (PV) prepared from guinea pig ileal myenteric plexus were found to contain, by radioimmunoassay, gastrin-releasing polypeptide (GRP), substance P (SP), galanin, Leu-enkephalin (LE), Met-enkephalin (ME), and vasoactive intestinal polypeptide (VIP). SP was present in the highest concentration followed by, in descending order, ME, LE, VIP, GRP and galanin. On reverse-phase HPLC, SP-, LE- and ME-like immunoreactivity in the PV preparation eluted at retention times similar to their synthetic analogues, galanin-like immunoreactivity eluted at a retention time different from that of synthetic porcine galanin and VIP-like immunoreactivity eluted at the retention time of synthetic guinea pig VIP. GRP-like immunoreactivity, on reverse-phase HPLC, eluted at retention times close to that of synthetic porcine GRP-(1-27) and its major oxidized form. Evidence was obtained for the presence of an alpha-neurokinin-like immunoreactive entity and an unidentified SP-like immunoreactive entity in guinea pig myenteric plexus.  相似文献

Soils that had a high binding capacity for inorganic orthophosphate (Pi) had reduced capacities to bind ionic alkyl phosphorus compounds. Only ionic methylphosphonate (MPn) and ionic phenylphosphonate exhibited moderate binding. Pseudomonas testosteroni used either MPn or Pi as a sole phosphorus source and exhibited diauxic utilization of MPn and Pi. The utilization of MPn was suppressed in the presence of Pi. This suppression was abolished by a Pi-binding soil. The soil did not have a significant effect on the maximum rate of degradation of either MPn or the poorly bound ionic O-isopropyl methylphosphonate, whereas the amount of MPn (but not the amount of O-isopropyl methylphosphonate) metabolized was reduced in the presence of soil  相似文献
