The management of subsurface soil acidity remains a challenge. We tested the ability of calcium nitrate fertilization to force net anion uptake by wheat within acidic subsurface layers and hence its ability to increase, or at least maintain, soil pH within the acidic layer. The trials were conducted at two field sites: the moderately acidic site ran for 3 years (2006?C08) while the trial at the most acidic site was conducted for 2 years (2006?C07). Five treatments (nitrogen form and application method) were compared. Uptake efficiency of nitrogen (N) as measured by 15N was similar (at ~40 to 70%) for both urea and nitrate sources at both sites. The urea source acidified the soils (up to 0.2 pH in 3 years) whilst the nitrate form resulted in increases in soil pH (up to 0.3 pH). The increases in pH were of a similar magnitude to the acidification rate measured in a nearby long-term trial. The dry matter production and grain yield in our trials were compromised by the decade long drought in our region, so the work should be repeated during wetter seasons. Calcium nitrate fertilization is a useful tool for at least maintaining soil pH, and even reversing soil acidification, in acidic subsurface soil layers.  相似文献

Drought significantly constrains higher yield of alfalfa (Medicago sativa L.) in arid and semiarid areas all over the world. This study evaluated the responses of leaf cuticular wax constituents to drought treatment and their relations to gas-exchange indexes across six alfalfa cultivars widely grown in China. Water deficit was imposed by withholding water for 12 d during branching stage. Cuticular waxes on alfalfa leaves were dominated by primary alcohols (41.7?C54.2%), alkanes (13.2?C26.9%) and terpenes (17.5?C28.9%), with small amount of aldehydes (1.4?C3.4%) and unknown constituents (4.5?C18.4%). Compared to total wax contents, the wax constituents were more sensitive to drought treatment. Drought decreased the contents of primary alcohol and increased alkanes in all cultivars. Alkane homologs, C25, C27, and C29, were all negatively correlated with photosynthetic rate, transpiration rate, stomatal conductance, and leaf water potential. Under drought conditions, both stomatal and nonstomatal factors were involved in controlling water loss from alfalfa leaves. No direct relationship was observed between wax contents and drought resistance among alfalfa cultivars. An increase in alkane content might be more important in improving drought tolerance of alfalfa under water deficit, which might be used as an index for selecting and breeding drought resistant cultivars of alfalfa.  相似文献

Children with neurofibromatosis type 1 (NF1) frequently have cognitive and behavioral deficits. Some of these deficits have been successfully modeled in Nf1 genetically-engineered mice that develop optic gliomas (Nf1 OPG mice). In the current study, we show that abnormal motivational influences affect the behavior of Nf1 OPG mice, particularly with regard to their response to novel environmental stimuli. For example, Nf1 OPG mice made fewer spontaneous alternations in a Y-maze and fewer arm entries relative to WT controls. However, analysis of normalized alternation data demonstrated that these differences were not due to a spatial working memory deficit. Other reported behavioral results (e.g., open-field test, below) suggest that differential responses to novelty and/or other motivational influences may be more important determinants of these kinds of behavior than simple differences in locomotor activity/spontaneous movements. Importantly, normal long-term depression was observed in hippocampal slices from Nf1 OPG mice. Results from elevated plus maze testing showed that differences in exploratory activity between Nf1 OPG and WT control mice may be dependent on the environmental context (e.g., threatening or non-threatening) under which exploration is being measured. Nf1 OPG mice also exhibited decreased exploratory hole poking in a novel holeboard and showed abnormal olfactory preferences, although L-dopa (50 mg/kg) administration resolved the abnormal olfactory preference behaviors. Nf1 OPG mice displayed an attenuated response to a novel open field in terms of decreased ambulatory activity and rearing but only during the first 10 min of the session. Importantly, Nf1 OPG mice demonstrated investigative rearing deficits with regard to a novel hanging object suspended on one side of the field which were not rescued by L-dopa administration. Collectively, our results provide new data important for evaluating therapeutic treatments aimed at ameliorating NF1-associated cognitive/behavioral deficits.  相似文献

Tumour necrosis factor (TNF) did not stimulate lipolysis in isolated rat adipocytes, though preincubation with TNF increased adrenaline-stimulated fatty acid release. Glycogenolysis, gluconeogenesis and ketogenesis in isolated rat hepatocytes were not influenced by TNF in short-term (30-60 min) incubations. TNF stimulated 14CO2 production from [U-14C]glucose in rat hemidiaphragm preparations, but lactate production and alanine release were not significantly altered. It is concluded that TNF does not regulate short-term metabolism in adipocytes, hepatocytes and muscle preparations in the manner of a catabolic hormone.  相似文献

The landscapes colonized by invasive earthworms in the eastern U.S. are often patchworks of forest stands in various stages of successional development. We established six field sites in tulip poplar dominated forests in the Smithsonian Environmental Research Center in Edgewater, MD, that span mid (50–70 years-three plots) and late (120–150 years-three plots) successional stages where younger sites had greater earthworm density and biomass than older sites and were dominated by non-native lumbricid species. In particular Lumbricus rubellus, a litter-feeding species, was abundant in mid successional forests. Here, we separated particulate organic matter (POM) from the bulk soil by a combination of size and density fractionation and found that patterns in soil POM chemistry were similar to those found previously during litter decay: in younger forests with high abundance of earthworms, organic carbon normalized cutin- and suberin-derived substituted fatty acid (SFA) concentration was lower and lignin-derived phenols greater than in older forests where earthworms were less abundant. The chemistry of the dominant litter from mid versus late successional tree species did not fully explain the differences in POM chemistry between age classes. Instead, the differences in leaf body versus petiole and leaf versus root chemistry were the dominant drivers of POM chemistry in mid versus late successional stands, although aspects of stand age and tree species also impacted POM chemistry. Our results indicate that preferential ingestion of leaf body tissue by earthworms and the subsequent shifts in sources of plant biopolymers in soil influenced POM chemistry in mid successional forests. These results indicate that invasive earthworm activity in North American forests contributes to a shift in the aromatic and aliphatic composition of POM and thus potentially influences carbon stabilization in soil.  相似文献

Venom from the parasitoid wasp Pimpla hypochondriaca contains numerous proteins, has potent in vitro anti-haemocytic properties, and disrupts host encapsulation responses. By sequencing 500 cDNAs randomly isolated from a venom gland library, we have identified 60 clones that encode proteins containing potential secretory signal sequences. To identify cDNAs encoding particular venom proteins, N-terminal amino acid sequences were determined for large (>30 kDa) venom proteins that had been separated using a combination of gel filtration and SDS-PAGE. We describe five of these cDNAs, which encoded residues that matched with the N-terminal sequences of previously undescribed venom proteins. cDNAs vpr1 and vpr3 encoded related proteins of approximately 32 kDa that were found in widely different fractions of gel filtration-separated venom. Neither vpr1 nor vpr3 were closely related to any other protein in the GenBank database, suggesting that they are highly specialised venom components. vpr2 encoded a 57-kDa polypeptide that was similar to a Drosophila protein, of unknown function, which lacks a signal sequence. A fourth clone, tre1, encoded a 61-kDa protein with extensive sequence similarity to trehalases. The 76-kDa sequence encoded by lac1 contained three regions which were very similar to histidine-rich copper-binding motifs, and could be aligned with the laccase from the fungus Coprinus cinereus. This study represents a significant step towards a holistic view of the molecular composition of a parasitoid wasp venom.  相似文献
