为初步探索叶绿体及其功能与超微弱发光(Ultraweak luminescence,UWL)激发的关系,揭示UWL与植物生长生理的关系及植物中UWL产生的来源,本试验以欧李(Cerasus humilis)为材料,采用室内盆栽试验,设置不同浓度盐胁迫处理,研究盐胁迫下欧李叶片的叶绿体结构和功能(叶绿素代谢、光系统Ⅱ活性、光合性能和能量水平)及UWL的变化规律和相关性。结果表明:(1)盐胁迫降低了欧李叶片的UWL强度,且盐浓度越高,UWL强度下降程度越大;(2)盐胁迫破坏了欧李叶片叶绿体结构,并降低了其功能,具体表现为叶绿素合成主要前体物质(ALA、Mg-ProtoⅨ)含量显著降低,叶绿素降解酶叶绿素酶(Chlase)活性显著升高,导致叶绿素(Chla、Chlb、Car和Chla+b)含量显著降低;同时欧李叶片FV/Fm、FV/FO、PIABS、RC/CSm、φE0和ΨE0快速下降,光系统Ⅱ活性受到严重抑制;进一步Pn、Tr、Gs下降,Ci同时升高,光合性能显著减弱;ATP含量和EC的显著降低,导致能量水平整体下降;(3)欧李叶片UWL强度与其叶绿素代谢物质及叶绿素含量(ALA、Mg-ProtoⅨ、Chla、Chlb、Car和Chla+b)、光系统Ⅱ活性(FV/Fm、FV/FO、PIABS、RC/CSm、φE0、ΨE0)、光合性能(Pn、Tr、Gs)及能量水平(ATP、EC)等参数均呈显著或极显著性正相关关系。(4)盐浓度越高,胁迫时间越长,欧李叶片UWL强度与叶绿体功能各指标变化程度越大,且高浓度处理下的相关性整体高于低浓度处理。可见,在盐胁迫条件下,欧李叶片叶绿体结构被破坏,同时其功能受到损伤活力下降,从而导致UWL强度降低;UWL强度与叶绿体及其功能关系密切,叶绿体可能是UWL的细胞器之一;UWL强度可以用来反映欧李叶片受盐胁迫伤害的程度。  相似文献

河蚬是一类在我国广泛分布的底栖贝类,具有重要的经济价值及生态价值。近年来,河蚬野生资源量锐减,了解河蚬在国内的潜在分布能为河蚬的保护和合理利用提供重要参考。基于河蚬在中国的136个分布点和8个环境因子,采用ENMeval包和biasfile优化后的最大熵模型(MaxEnt)预测分别河蚬现代和未来(2041—2060年和2081—2100年) 6个气候情景下的潜在分布。综合Jackknife检验、置换重要值和环境因子贡献率评估影响现代河蚬潜在分布的主要因子,比较未来气候情景下潜在适生区差异从而分析预测河蚬适宜分布的变化。结果表明:(1)优化后的MaxEnt模型预测准确度极高,平均AUC值为0.900±0.037,平均AUCDIFF值为0.019,现代河蚬潜在分布区域总面积为188.33×104 km2,主要集中在长江流域、海河流域、淮河流域、珠江流域、东南沿海区域以及黄河流域下游和渤海湾沿岸区域。(2)影响河蚬潜在分布的主要环境因子为海拔、温度(年均温和温度年较差)和降水(年降水量)。(3)在未来6种气候情景下,河蚬主要潜...  相似文献

通过构建表达人膜辅因子蛋白MCP编码区全长cDNA的重级载体pcDNA3MCP,以磷酸钙沉淀法转染NIH3T3细胞,用G418筛选阳性克隆,并鉴定MCP cDNA在细胞中的表达。将血清梯度稀释并与细胞孵育,然后检测细胞活力:灭活的人血清不能引起对照细胞与MCP转染细胞的胞毒作用,而新鲜人血清可使对照细胞发生补体依赖的细胞毒反应,MCP转染细胞由于MCP蛋白的合成,细胞受到保护不发生该反应。另外,M  相似文献

Since April 2010, Tembusu virus (TMUV) which is a contagious pathogen of waterfowls, causing symptoms of high fever, loss of appetite and fall in egg production, has been reported in east of China. A double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) which detects for TMUV was developed, using two monoclonal antibodies (mAbs) against the TMUV envelope (E) protein. BALB/c mice were immunized with purified recombinant E protein expressed in E. coli. Three hybridoma cell lines designated as 12B1, 10C6 and 2D2, were screened by cell fusion and indirect ELISA for their ability to recognize different linear epitopes on the E protein, and were characterized subsequently. High-affinity mAbs 12B1 and 2D2 were used as capture and detection antibodies, respectively. The reaction conditions for the DAS-ELISA were optimized for TMUV detection. The cross-reactivity of the DAS-ELISA was determined using TMUV, duck plague virus, avian influenza virus subtype H9, Newcastle disease virus, duck hepatitis A virus type 1 and duck reovirus samples. A total of 191 homogenized tissues of field samples were simultaneously detected by DAS-ELISA and by RT-PCR. The former was found to have a high specificity of 99.1% and a sensitivity of 93.1%. These results reveal a positive coincidence between DAS-ELISA and RT-PCR at a coincidence rate of 95.8%. The method developed in this study can be used for the diagnosis of TMUV infection of duck origin.  相似文献

A process for human influenza H1N1 virus vaccine production from Madin–Darby canine kidney (MDCK) cells using a novel packed-bed bioreactor is described in this report. The mini-bioreactor was used to study the relationship between cell density and glucose consumption rate and to optimize the infection parameters of the influenza H1N1 virus (A/New Caledonia/20/99). The MDCK cell culture and virus infection were then monitored in a disposable perfusion bioreactor (AmProtein Current Perfusion Bioreactor) with proportional–integral–derivative control of pH, dissolved O2 (DO), agitation, and temperature. During 6 days of culture, the total cell number increased from 2.0?×?109 to 3.2?×?1010 cells. The maximum virus titers of 768 hemagglutinin units/100 μL and 7.8?×?107 50 % tissue culture infectious doses/mL were obtained 3 days after infection. These results demonstrate that using a disposable perfusion bioreactor for large-scale cultivation of MDCK cells, which allows for the control of DO, pH, and other conditions, is a convenient and stable platform for industrial-scale production of influenza vaccines.  相似文献

In rabbit corneal epithelial cells (RCEC), we determined whether capacitative calcium entry (CCE) mediates the mitogenic response to epidermal growth factor, EGF. [Ca2+]i was measured with single-cell fluorescence imaging of fura2-loaded RCEC. EGF (5 ng/ml) maximally increased [Ca2+]i 4.4-fold. Following intracellular store (ICS) calcium depletion in calcium-free medium with 10 µM cyclopiazonic acid (CPA) (endoplasmic reticulum calcium ATPase inhibitor), calcium addback elicited plasma membrane Ca2+ influx as a result of activation of plasma membrane store operated channel (SOC) activity. Based on Mn2+ quench measurements of fura2 fluorescence, 5 ng/ml EGF enhanced such influx 2.3-fold, whereas with Rp-cAMPS (protein kinase A inhibitor) plus EGF it increased by 5.3-fold. In contrast, SOC activation was blocked with 100 µM 2-aminoethyldiphenylborate (2-APB, store-operated channel inhibitor). During exposure to either 50 µM UO126 (MEK-1/2 inhibitor) or 10 µM forskolin (adenylate cyclase activator), 5 ng/ml EGF failed to affect [Ca2+]i. RT-PCR detected gene expression of: 1) transient receptor potential (TRP) protein isoforms 1, 3, 4, 6 and 7; 2) IP3R isoforms 1–3. Immunocytochemistry, in conjunction with confocal and immunogold electron microscopy, detected plasma membrane localization of TRP4 expression. Inhibition of CCE with 2-APB and/or CPA, eliminated the 2.5-fold increase in intracellular [3H]-thymidine incorporation induced by EGF. Taken together, CCE in RCEC mediates the mitogenic response to EGF. EGF induces CCE through its stimulation of Erk1/2 activity, whereas PKA stimulation suppresses these effects of EGF. TRP4 may be a component of plasma membrane SOC activity, which is stimulated by ICS calcium depletion.  相似文献

Introduction

DNA methyltransferase-1(DNMT1) is an important enzyme in determining genomic methylation patterns in mammalian cells. We investigated the associations between SNPs in the DNMT1 gene and risks of developing H. pylori seropositivity, gastric atrophy and gastric cancer in the Chinese population.

Methods

The study consisted of 447 patients with gastric cancer; 111 patients with gastric atrophy; and 961 healthy controls. Five SNPs, rs10420321, rs16999593, rs8101866, rs8111085 and rs2288349 of the DNMT1 gene were genotyped. Anti-H.pylori IgG was detected by ELISA. Gastric atrophy was screened by the level of serum pepsinogen Ιand II and then confirmed by endoscopy and histopatholgical examinations.

Results

The age- and sex-adjusted OR of H. pylori seropositivity was 0.67 (95%CI: 0.51–0.87) for rs8111085 TC/CC genotypes, significantly lower than the TT genotype in healthy controls. The adjusted OR of H.pylori seropositivity was 0.68 (95%CI: 0.52–0.89) for rs10420321 AG/GG genotypes. In addition, patients carrying rs2228349 AA genotype have a significantly increased risk for H.pylori seropositivity (OR = 1.67; 95%CI: 1.02–2.75). Further haplotype analyses also showed that the ATTTG and ATCTA are significantly associated with increased risks in H.pylori infection compared to the GTCCG haplotype (OR = 1.38, 95%CI: 1.08–1.77; OR = 1.40, 95% CI: 1.09–1.80). The adjusted ORs of gastric atrophy were 1.66 (95%CI: 1.06–2.61) for rs10420321 GG genotype, and 1.67 (95%CI 1.06–2.63, P = 0.03) for rs8111085 CC genotype, but no association was found between SNPs in the DNMT1 gene and risk of developing gastric cancer.

Conclusions

Individuals with rs10420321 GG and rs8111085 CC genotype of the DNMT1 gene were associated with reduced risks for H.pylori infection. On the other hand, higher risks of gastric atrophy were found in the carriers with these two genotypes compared to other genotypes. Our results suggested that SNPs of DNMT1 could be used as genotypic markers for predicting genetic susceptibilities to H.pylori infection and risks in gastric atrophy.  相似文献

砍伐是人类对植物种群最常见的干扰形式之一,种群结构及其动态在砍伐后会经历快速变化和调整时期。以长汀侵蚀退化地为研究区,基于砍伐迹地上马尾松种群4年生活史调查,利用积分投影模型(IPM,Integral Projection Model)结合传统种群统计分析方法,揭示砍伐迹地上马尾松种群增长率变化过程及个体生命率对种群的影响。结果表明:砍伐迹地上马尾松种群处于衰退状态(λ1),且衰退程度不断加深。4年内幼苗数量大幅度减少,幼年树和成树数量增加。死亡在生活史各个阶段均有出现,幼苗是死亡的主要来源;植株生长量随着个体的增大和时间的增加而增加,生长增量由大到小依次为:成年树,幼年树和幼苗;植株繁殖概率逐年提升,但新生幼苗数量逐年下降。弹性分析显示,个体尤其是幼苗的存活对种群至关重要,小径级植株的正生长有利于种群恢复,大径级植株的正生长不利于种群发展,而负增长和繁殖贡献均十分有限。  相似文献
