A study of blood pressure control in elderly outpatients was carried out with the participation of 444 Italian general practitioners. Of 4096 patients aged 65 years or over who were considered for recruitment, 3959 (96.7%) fulfilled all the criteria of admission and were followed up for 12 months. The findings regarding one of the aims of the study--that is, to assess the feasibility of a large scale trial in general practice--are reported. Most (87%) of the doctors completed the study. Their adherence to the protocol was highly satisfactory, leading to an acceptable quality of work. Patients'' compliance was also good; 98.6% (3898) of the patients who had fulfilled the admission criteria agreed to participate in the study, and only 4% (158) dropped out. Both of these observations support the feasibility of carrying out prospective studies in general practice. The creation of networks of general practitioners who are prepared to carry out research in their practices would allow treatment and preventive measures to be studied simply and at low cost in the appropriate setting.  相似文献

Co-cultures of rat ventricular myocytes and sympathetic neurons were established. Superior cervical ganglia and ventricles from newborn rats were enzymatically dissociated and plated in a culture dish. Experiments were done between the 3rd (when evidence of neuron-myocyte proximity arises) and the 5th day in culture (before the myocytes become confluent). Simultaneous intracellular recording from a cardiomyocyte and an attached neuron was done using conventional microelectrode techniques (resistance of 60-100 Mohm). The myocytes in co-culture were either quiescent or spontaneously contracting. The contracting cells were either latent pacemaker or ventricular-like myocytes. The action potential (AP) characteristics of cardiomyocytes in co-cultures were comparable to those recorded in cardiomyocytes in pure cultures. Sympathetic innervation of the cardiomyocytes in co-cultures was evidenced by stimulating the neuron and observing an increase in rate of beating in latent pacemaker myocytes (average increase of 19.4 +/- 4.6%). In quiescent cardiomyocytes, neural stimulation evoked a slow depolarization that can reach threshold and initiate APs in the cell. This response is similar to slow excitatory postsynaptic potentials (EPSPs) observed in other synapses. Slow ESPSs could also be recorded in spontaneous beating cells, made quiescent by nifedipine (1x10(-6)-1x10(-7) M). These results indicate that functional synaptic contacts are developed in co-culture of sympathetic neurons and cardiac myocytes, and slow EPSPs can be evoked in cardiomyocytes as well as in other excitable cells. The sympathetic innervation occurring in culture did not significantly modify the spontaneous AP characteristics of the cardiomyocytes.  相似文献

Human lymphocytes were separated by centrifugation and grown for 48--72 h in TC 199 medium. Synchronous cells, obtained by the addition of colchicine, were treated with arabinosylcytosine (Ara-C), an antimitotic substance which is known to produce nuclear changes in vivo and in vitro. In this case, alterations in the nuclear envelope closely resembling the malignancy associated changes described in human leukaemic cells were observed. The results allow us to suggest that such alterations may be due to a cellular derangement involving the renewal of the nuclear envelope in th G1 phase of the mitotic cycle.  相似文献

The ultrastructural changes of human leukemic cell nuclei have been investigated. Particular attention is paid to the alteration of the nuclear envelope and its constituents, i.e., the pores and the Zonula Nucleum Limitans which appear constantly involved in these pathologic processes. An alteration of the relationships between the components of the nuclear envelope and the chromatin itself may be responsible for the appearance of the most nuclear changes.  相似文献

Summary The pigmentary system of the planaria, Dugesia gonocephala s.l. (Platyhelminthes, Turbellaria, Tricladida), consists of granules contained in chromatophore cells distributed in the parenchyma tissue. The administration of MSH release-Inhibiting Factor (M.I.F.) leads to an easily observable general decolouration of the animal due to the migration of the pigment granules towards the deeper-lying cell nucleus.In planarians bisected transversely through the pharyngeal region, the decolouration occurs only in the cephalic segment, and the caudal segment remains dark. When, however, the decapitated caudal segment regenerates a head region, a decolouration response occurs when exposed to M.I.F.The significance of these results is discussed, and an hypothesis on the hormonal regulation of the pigmentary system is proposed.  相似文献
