In 2009, the Fundy Ocean Research Centre for Energy (FORCE) developed a 1.6 x 1.0 km Crown Lease Area (CLA) to test Marine Hydrokinetic devices (MHKs) in Minas Passage (MP), a strait that connects the Bay of Fundy to Minas Basin (MB), Nova Scotia. Minas Basin is an important summer feeding aggregation site for numerous fishes including Atlantic sturgeon (Acipenser oxyrinchus Mitchill, 1815) stocks from Canada and the US. In this study, acoustic tagging technology was used to describe sturgeon presence in the CLA, as well as an adjacent site proposed for MHKs deployment. During 2018 and 2019, 33.9% (n = 19/56 Atlantic sturgeon tagged) and 29.9% (n = 23/77 Atlantic sturgeon tagged) respectively, of sturgeon acoustically tagged in MB during 2014–2019 were detected in these two FORCE sites. Binomial General Linear Models (GLMs) were fit to determine if environmental parameters influenced the presence, or depth of Atlantic sturgeon in the FORCE sites. The only significant predictor of Atlantic sturgeon depth and presence at the FORCE sites was tide type. Atlantic sturgeon was found to have a higher likelihood of being detected during ebb tide (12%) as opposed to flood tide (3%). Apparent decline in the presence of sturgeon during flood tide was probably due to poor signal reception caused by turbulence from higher current speeds. Tagged sturgeon travelled through the FORCE sites pelagically at similar depths (27.5 ± 14.57 m) to those proposed for seabed mounted MHKs (~30 m), travelling deeper during flood tide than ebb, suggesting they may be at risk of spatial overlap with MHKs. While this study indicated that the duration Atlantic sturgeon utilized the FORCE sites throughout the summer appeared to be low, sturgeon from both endangered and threatened stocks migrated through this region during their yearly entry and exit to MB and were present for approximately 12% of the days assessed in this study. 相似文献

Early studies with Geobacter sulfurreducens suggested that outer-surface c-type cytochromes might play a role in U(VI) reduction, but it has recently been suggested that there is substantial U(VI) reduction at the surface of the electrically conductive pili known as microbial nanowires. This phenomenon was further investigated. A strain of G. sulfurreducens, known as Aro-5, which produces pili with substantially reduced conductivity reduced U(VI) nearly as well as the wild type, as did a strain in which the gene for PilA, the structural pilin protein, was deleted. In order to reduce rates of U(VI) reduction to levels less than 20% of the wild-type rates, it was necessary to delete the genes for the five most abundant outer surface c-type cytochromes of G. sulfurreducens. X-ray absorption near-edge structure spectroscopy demonstrated that whereas 83% ± 10% of the uranium associated with wild-type cells correspond to U(IV) after 4 h of incubation, with the quintuple mutant, 89% ± 10% of uranium was U(VI). Transmission electron microscopy and X-ray energy dispersion spectroscopy revealed that wild-type cells did not precipitate uranium along pili as previously reported, but U(IV) was precipitated at the outer cell surface. These findings are consistent with those of previous studies, which have suggested that G. sulfurreducens requires outer-surface c-type cytochromes but not pili for the reduction of soluble extracellular electron acceptors. 相似文献

Iron-reducing bacteria (FeRB) play key roles in anaerobic metal and carbon cycling and carry out biogeochemical transformations that can be harnessed for environmental bioremediation. A subset of FeRB require direct contact with Fe(III)-bearing minerals for dissimilatory growth, yet these bacteria must move between mineral particles. Furthermore, they proliferate in planktonic consortia during biostimulation experiments. Thus, a key question is how such organisms can sustain growth under these conditions. Here we characterized planktonic microbial communities sampled from an aquifer in Rifle, Colorado, USA, close to the peak of iron reduction following in situ acetate amendment. Samples were cryo-plunged on site and subsequently examined using correlated two- and three-dimensional cryogenic transmission electron microscopy (cryo-TEM) and scanning transmission X-ray microscopy (STXM). The outer membranes of most cells were decorated with aggregates up to 150 nm in diameter composed of ∼3 nm wide amorphous, Fe-rich nanoparticles. Fluorescent in situ hybridization of lineage-specific probes applied to rRNA of cells subsequently imaged via cryo-TEM identified Geobacter spp., a well-studied group of FeRB. STXM results at the Fe L2,3 absorption edges indicate that nanoparticle aggregates contain a variable mixture of Fe(II)–Fe(III), and are generally enriched in Fe(III). Geobacter bemidjiensis cultivated anaerobically in the laboratory on acetate and hydrous ferric oxyhydroxides also accumulated mixed-valence nanoparticle aggregates. In field-collected samples, FeRB with a wide variety of morphologies were associated with nano-aggregates, indicating that cell surface Fe(III) accumulation may be a general mechanism by which FeRB can grow while in planktonic suspension. 相似文献

Mexico is the main producer, consumer and exporter
of avocado in the world, being Michoacan the main producer state
contributing more than 80% of the national production. There
are phytopathogens that decimate the production causing the
death of the tree. Root samples were collected in avocado trees
that showed the characteristic symptomatology of the disease
known as avocado sadness, the sampling was carried out in four
of the main avocado producing towns, in the state of Michoacan,
Mexico. The isolation consisted in sowing root tissue in Petri
dishes with V8®-PARPH culture medium, subsequently they were
identified morphologically and for species level it was determined
by molecular biology, with the PCR-ITS technique. Pathogenicity
tests were performed in triplicate with avocado seedlings with more
than six leaves. After 24 hours, the inoculated plants expressed
decay in the apical part, after 120 hours the leaves showed yellowing
and after 15 days there was a generalized wilt on the stem and
leaves, re-isolating the phytopathogen Phytopythium vexans.
This study confirms the first report of the oomycete P. vexans
affecting avocado trees in the most important producing region of

Expression of the adhesion molecules, ICAM-1, VCAM-1, NCAM, CD44, CD49d (VLA-4, a chain), and CDlla (LFA-1, a chain) on mouse oocytes, and pre- and peri-implantation stage embryos was examined by quantitative indirect immunofluorescence microscopy. ICAM-1 was most strongly expressed at the oocyte stage, gradually declining almost to undetectable levels by the expanded blastocyst stage. NCAM, also expressed maximally on the oocyte, declined to undetectable levels beyond the morula stage. On the other hand, CD44 declined from highest expression at the oocyte stage to show a second maximum at the compacted 8-cell/morula. This molecule exhibited high expression around contact areas between trophecto-derm and zona pellucida during blastocyst hatching. CD49d was highly expressed in the oocyte, remained significantly expressed throughout and after blastocyst hatching was expressed on the polar trophecto-derm. Cell-free cytosolic extracts from the Yoshida (AH 130) rat ascites hepatoma cell line, grown in vivo, showed high ribosomal protein S6 kinase activity in vitro, as measured by transfer of 32P to exogenous 40S rat liver ribosomal subunits, in both exponential growing and stationary phase cells. A significant decrease of protein synthesis (3H-leucine incorporation into total cell protein) was found to occur in cells reaching the stationary phase of growth, suggesting that S6 phosphorylation was not tightly coupled to the rate of the intraperitoneal cell growth and of protein synthesis in these tumor cells. When the cell-free cytosolic extracts were prepared from cells exposed to amiloride, at concentrations that inhibit the Na+/H+ exchange, a decrease of S6 kinase activity was observed only in exponential growing cells, suggesting the possibility of coupling of the Na+/H+ exchange with phosphorylation of intracellular proteins in these tumor cells. Actually, stationary phase cells showed unchanged S6 kinase activity under the same conditions, possibly due to the extremely low Na+/H+ exchange activity, previously demonstrated (Cell Biol. Int. Rep., 1985, 9, 1017-1025). The present experiments support the hypothesis that the regulation of protein synthesis is not tightly coupled to phosphorylation-dephosphorylation cycles, at least of ribosomal protein S6, in cells characterized by a rather uncontrolled growth such as the Yoshida (AH 130) rat ascites hepatoma. Microsatellite allele frequencies in humans and chimpanzees, with implications for constraints on allele size 总被引:24,自引:6,他引:18
The distributions of allele sizes at eight simple-sequence repeat (SSR) or
microsatellite loci in chimpanzees are found and compared with the
distributions previously obtained from several human populations. At
several loci, the differences in average allele size between chimpanzees
and humans are sufficiently small that there might be a constraint on the
evolution of average allele size. Furthermore, a model that allows for a
bias in the mutation process shows that for some loci a weak bias can
account for the observations. Several alleles at one of the loci (Mfd 59)
were sequenced. Differences between alleles of different lengths were found
to be more complex than previously assumed. An 8-base-pair deletion was
present in the nonvariable region of the chimpanzee locus. This locus
contains a previously unrecognized repeated region, which is imperfect in
humans and perfect in chimpanzees. The apparently greater opportunity for
mutation conferred by the two perfect repeat regions in chimpanzees is
reflected in the higher variance in repeat number at Mfd 59 in chimpanzees
than in humans. These data indicate that interspecific differences in
allele length are not always attributable to simple changes in the number
of repeats.
