High hydrostatic pressure has been shown to produce neurological changes in humans which manifest, in part, as tremor, myoclonic jerks, electroencephalographic changes, and convulsions. This clinical pattern has been termed high-pressure nervous syndrome (HPNS). These symptoms may represent an alteration in synaptic transmission in the central nervous system with the inhibitory neural pathways being affected in particular. Since gamma-aminobutyric acid (GABA) transmission has been implicated in other seizure disorders, it was of interest to study GABAergic function at high pressure. Isolated synaptosomes were used to follow GABA release at 67.7 ATA of pressure. The major observation was a 33% depression in total [3H]GABA efflux from depolarized cerebrocortical synaptosomes at 67.7 ATA. The Ca2+-dependent component of release was found to be completely blocked during the 1st min of [3H]GABA efflux with a slow rise over the subsequent 3 min. These findings lead us to conclude that high pressure interferes with the intraterminal cascade for Ca2+-dependent release of GABA. 相似文献

《Inorganica chimica acta》1994,220(1-2):99-105Electrospray mass spectra have been observed for a number of alkene and arene complexes of Ag(I) formed by the interaction of AgNO3 and the organometallic ligand in water/methanol solution. The ES mass spectra show that almost all the alkene and arene ligands in stoichiometric excess form labile 1:2 cationic complexes with Ag(I) which are easily decomposed by collisional activation to the 1:1 species. However, with a deficiency of organic ligand polymeric species are observed. The cation [Ag(cod)2]+ (cod=1,5-cyclooctadiene) was reacted with a variety of other potential ligands, such as PPh3, AsPh3, PhSCH2SPh etc. In most cases, mixed complexes [Ag(cod)(ligand)]+ were observed, and excess ligand usually produced [Ag(ligand)2]+. 相似文献

The relative efficiency of acetate and glucose as substrates for the biosynthesis of lipids in the skin of the rat and horse was examined using in vivo pulse labelling of skin with [1-14C]acetate and [U-14C]glucose by intradermal injections. The resulting radiolabelled lipids were recovered in the rat by punch biopsy as well as by daily, long-term skin surface lipid collections and in the horse by punch biopsy of the injection sites. The lipids were examined by liquid scintillation and by a combination of thin-layer chromatography and autoradiography. In both species the recovery of radiolabel in the non-polar lipids was much higher after a pulse of [1-14C]acetate than after a pulse of [U-14C]glucose. In the rat, the skin surface lipids labelled through acetate contained sufficient radiolabel to allow observation of the time course of excretion of 14C in the major non-polar lipid classes. The results suggest that the biosynthesis of these lipid classes in the sebaceous glands of the rat are not entirely synchronous. In the skin lipid extracts of the horse, all of the major lipid classes, including phospholipids and glycolipids, were labelled through acetate. Sampling in a soybean field established that presence of velvetleaf (A. theophrasti) weeds interfered with soybean production. Number of soybean pods and number of pods/stem were significantly lower in transect segments adjacent to velvetleaf plants. In bioassays for phytotoxicity of velvetleaf, several dilutions of aqueous extracts from fresh field-collected leaves depressed germination of radish seeds and inhibited growth of soybean seedlings. Seed germination bioassays from eluates of chromatograms developed in one dimension showed that two of three bands containing phenolic compounds were inhibitory to radish seed germination. Soybeans inhibited by aqueous velvetleaf extracts had increased diffusive resistance, suggesting partial stomatal closure. Inhibited plants also gave evidence of water stress, with leaf water potentials often as low as –20 bars and reduced water content, when compared with controls. Quantification of chlorophyll on a leaf area basis showed that chlorophyll of inhibited plants was below controls. These data demonstrate the allelopathic potential of velvetleaf and suggest that interference with water balance and chlorophyll content may be two mechanisms of inhibitory action of toxins present in the leaves of velvetleaf. 相似文献

A continuous flow reactor (perifusion system) was fabricated and tested for measuring the kinetics of insulin secretion from isolated pancreatic islets of Langerhans in response to step changes in the glucose concentration and oxygen partial pressure in the perfusate flowing around the islets. The system was capable of making rapid changes in perfusate glucose concentration and pO2, had rapid dynamic response for measuring the change in insulin secretion rate as a result of these changes in perfusate, and was suitable for studying very small volumes of tissue. Initial experiments with this system demonstrated that (1) the response of isolated rat islets to glucose stimulation was very fast, with the first phase peak occurring in as little as about 10 s, (2) bulk perfusate oxygen partial pressure levels of 30 mmHg or less reduced the second-phase insulin secretion rate in graded fashion, (3) the reduction in secretion rate began within 1 min following an oxygen partial pressure decrease, and (4) the reduction in secretion rate was reversible, with a burst of insulin secretion occurring during the first minute after partial pressure restoration. 相似文献
