- 1 Although the successful management of the Colorado potato beetle Leptinotarsa decemlineata (Say) depends on the prevention of its dispersal, its walking pattern in the landscape remains poorly understood. In the present study, post‐diapause, early summer, late summer and colony adult beetles, both fed and unfed before release, were tracked with a harmonic radar to establish their walking movement pattern in a bare‐ground field.
- 2 The random walk model successfully described the dispersal of all beetle types, whether fed or unfed.
- 3 The diverse life history of this species was manifested by an increased distance travelled and deviations of individual paths from the random model. Starved post‐diapause beetles travelled furthest and individual paths deviating from random were both local and directed, probably aiming to maximize opportunities for host colonization. Starved early summer beetles also travelled further than fed beetles but relied more on random movement to disperse in the habitat. Starving had little impact on the distance travelled or the path deviations of late summer beetles that are searching for overwintering site rather than hosts.
- 4 The increased displacement of starving beetles over fed beetles corresponded with an increased walking step and index of straightness.
- 5 The impact of starvation on travel distance was greater than expected from laboratory tests.
- 6 In conclusion, the results obtained in the present study suggest a random walking pattern to search arable land until host volatile or visual impulses trigger a more directed walk or flight.

Using two ecotypes of Stellaria longipes with contrasting responses to shade, we found that plants can differ in their responses to similar light cues, reflecting adaptations to their natural habitat. It was also observed that the plants could distinguish between distinct shade signals. Furthermore, the activity of wall modifying proteins, expansins and xyloglucan endotransglucosylase/hydrolase(s) (XTHs) was regulated during these responses. However, only expansin activity and gene expression profiles correlated with observed growth trends. The differential expression of expansins was light signal specific and ecotype specific and could account for both the trends in growth and their magnitude. The mammaglobin gene has been shown to be preferentially expressed in breast tissue. Few genes match its specificity. Mammaglobin has generated much interest, and studies are ongoing to develop diagnostic tests for breast cancer based on the detection of mammaglobin. While searching the Incyte Genomics Lifeseq database for tissue-specific markers, we observed a second secretoglobin, BU101, also known as lipophilin B. We report here that mammaglobin, in breast tissue, is found as a complex with BU101. The complex was isolated from breast cancer tissue and was characterized as the biologically relevant form of mammaglobin. 相似文献

Modulation of voltage-dependent transient K(+) currents (A type K(+) or K(A) current) by Zn(2+) was studied in rat hippocampal neurons by the whole-cell patch-clamp technique. It is found that Zn(2+) selectively binds to the resting (deactivated or closed) K(A) channels with a dissociation constant (K(d)) of approximately 3 &mgr;M, whereas the affinity between Zn(2+) and the inactivated K(A) channels is 1000-fold lower. Zn(2+) therefore produces a concentration-dependent shift of the K(A) channel inactivation curve and enhances the K(A) current elicited from relatively positive holding potentials. It is also found that the kinetics of Zn(2+) action are fast enough to compete with the transition rates between different gating states of the channel. The rapid and selective binding of Zn(2+) to the closed K(A) channels keeps the channel in the closed state and explains the ion's concentration-dependent slowing effect on the activation of K(A) current. This in turn accounts for the inhibitory effect of Zn(2+) on the K(A) current elicited from hyperpolarized holding potentials. Because the molecular mechanisms underlying these gating changes are kinetic interactions between the binding-unbinding of Zn(2+) and the intrinsic gating processes of the channel, the shift of the inactivation curve and slowing of K(A) channel activation are quantitatively correlated with ambient Zn(2+) over a wide concentration range without "saturation"; i.e., The effects are already manifest in micromolar Zn(2+), yet are not saturated even in millimolar Zn(2+). Because the physiological concentration of Zn(2+) could vary over a similarly wide range according to neural activities, Zn(2+) may be a faithful physiological "fine tuner," controlling and controlled by neural activities through its effect on the K(A) current. 相似文献

Dengue virus (DENV) is a mosquito-borne flavivirus that causes serious human disease and mortality worldwide. There is no specific antiviral therapy or vaccine for DENV infection. Alterations in gene expression during DENV infection of the mosquito and the impact of these changes on virus infection are important events to investigate in hopes of creating new treatments and vaccines. We previously identified 203 genes that were ≥5-fold differentially upregulated during flavivirus infection of the mosquito. Here, we examined the impact of silencing 100 of the most highly upregulated gene targets on DENV infection in its mosquito vector. We identified 20 genes that reduced DENV infection by at least 60% when silenced. We focused on one gene, a putative cysteine rich venom protein (SeqID AAEL000379; CRVP379), whose silencing significantly reduced DENV infection in Aedes aegypti cells. Here, we examine the requirement for CRVP379 during DENV infection of the mosquito and investigate the mechanisms surrounding this phenomenon. We also show that blocking CRVP379 protein with either RNAi or specific antisera inhibits DENV infection in Aedes aegypti. This work identifies a novel mosquito gene target for controlling DENV infection in mosquitoes that may also be used to develop broad preventative and therapeutic measures for multiple flaviviruses. 相似文献

The lifespan of herbivorous Rana pipiens larvae is ~3 months, while that of carnivorous Ceratophrys ornata larvae is only about 2 weeks. During metamorphic climax, the larval gut shortens dramatically, especially in R. pipiens, and its luminal epithelium is replaced by adult‐type epithelium. To determine when programmed cell death occurs during the metamorphic restructuring of the gut, we prepared cross‐sections of the stomach, small intestine, and large intestine from representative larval stages and from juvenile frogs of both species. The sections were incubated with monoclonal antibody against active caspase‐3, one of the key enzymes in the apoptotic cascade. We observed apoptosis in some luminal epithelial cells in each of the three regions of the larval gastrointestinal tract of both species. However, apoptotic cells appeared earlier in larval stages of R. pipiens than C. ornata and few were seen in juvenile frogs of either species. The results demonstrate the occurrence of apoptosis in the metamorphic remodeling of the gut of both R. pipiens larvae and C. ornata larvae. J. Morphol., 2011. © 2011 Wiley Periodicals, Inc. 相似文献
