Endometrial carcinoma is a common malignancy in women, being exceeded in incidence only by that of breast, lung, and colorectal cancers. At present, no serum tumor markers are available for the monitoring of endometrial carcinoma patients, and patients with recurrent disease are detected only following the development of symptoms or abnormalities in imaging assessments. Similarly, no screening tools are available for endometrial carcinoma. Protein profiling by matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry (MALDI-TOF MS) has proven to be a sensitive and fast method of analysis for small proteins or peptides to yield specific biomarkers. In this study, a variety of normal and malignant endometrial tissue samples were fractionated and analyzed by SELDI-TOF MS (SELDI is a version of MALDI utilizing protein "chips"). A number of proteins displayed differential expression in malignant endometrial tissues. One of the prominent proteins fractionated by weak cation exchange chromatography and displaying enhanced expression in these malignant tissues was identified as chaperonin 10. The increased expression of chaperonin 10 in malignant endometrial tissues was further confirmed by parallel Western blot and immunohistochemistry analyses. 相似文献

The role of ethylene in regulating sugar, acid, texture and volatile components of fruit quality was investigated in transgenic apple fruit modified in their capacity to synthesize endogenous ethylene. Fruit obtained from plants silenced for either ACS (ACC synthase; ACC-1-aminocyclopropane-1-carboxylic acid) or ACO (ACC oxidase), key enzymes responsible for ethylene biosynthesis, expectedly showed reduced autocatalytic ethylene production. Ethylene suppressed fruits were significantly firmer than controls and displayed an increased shelf-life. No significant difference was observed in sugar or acid accumulation suggesting that sugar and acid composition and accumulation is not directly under ethylene control. Interestingly, a significant and dramatic suppression of the synthesis of volatile esters was observed in fruit silenced for ethylene. However, no significant suppression was observed for the aldehyde and alcohol precursors of these esters. Our results indicate that ethylene differentially regulates fruit quality components and the availability of these transgenic apple trees provides a unique resource to define the role of ethylene and other factors that regulate fruit development. 相似文献

CD1d-restricted T cells are implicated as key players in host defense against various microbial infections. However, the mechanisms involved and the role they play, if any, at the mucosal surfaces where pathogenic infections are initiated is unknown. In a murine pneumonia model established by intranasal application of Pseudomonas aeruginosa, CD1d(-/-) mice showed markedly reduced pulmonary eradication of P. aeruginosa compared with wild-type mice; this was associated with significantly lower amounts of macrophage inflammatory protein-2 and reduced numbers of neutrophils within the bronchoalveolar lavage fluid. Corollarily, treatment of mice with alpha-galactosylceramide--a lipid that activates CD1d-restricted T cells--increased the amount of interferon-gamma; this was associated with rapid pulmonary clearance through enhanced phagocytosis of P. aeruginosa by alveolar macrophages. These results reveal a crucial role played by CD1d-restricted T cells in regulating the antimicrobial immune functions of macrophages at the lung mucosal surface. 相似文献

Predator foraging success and habitat complexity: quantitative test of the threshold hypothesis 总被引:6,自引:2,他引:4
Summary Numerous studies have demonstrated a negative relationship between increasing habitat complexity and predator foraging success. Results from many of these studies suggest a non-linear relationship, and it has been hypothesised that some threshold level of complexity is required before foraging success is reduced significantly. We examined this hypothesis using largemouth bass (Micropterus salmoides) foraging on juvenile bluegill sunfish (Lepomis macrochirus) in various densities of artificial vegetation. Largemouth foraging success differed significantly among the densities of vegetation tested. Regression analysis revealed a non-linear relationship between increasing plant stem density and predator foraging success. Logistic analysis demonstrated a significant fit of our data to a logistic model, from which was calculated the threshold level of plant stem desity necessary to reduce predator foraging success. Studies with various prey species have shown selection by prey for more complex habitats as a refuge from predation. In this stydy, we also examined the effects of increasing habitat complexity (i.e. plant stem density) on choice of habitat by juvenile bluegills while avoiding predation. Plant stem density significantly effected choice of habitat as a refuge. The relationship between increasing habitat complexity and prey choice of habitat was found to be positive and non-linear. As with predator foraging success, logistic analysis demonstrated a significant fit of our data to a logistic model. Using this model we calculated the threshold level of habitat complexity required before prey select a habitat as a refuge. This density of vegetation proved to be considerably higher than that necessary to significantly reduce predator foraging success, indicating that bluegill select habitats safe from predation.Implications of these results and various factors which may affect the relationships described are discussed. 相似文献

We showed previously that p34(cdc2)/cyclin B (MPF) hyperphosphorylates poly(A) polymerase (PAP) during M-phase of the cell cycle, causing repression of its enzymatic activity. Mutation of three cyclin-dependent kinase (cdk) consensus sites in the PAP C-terminal regulatory domain prevented complete phosphorylation and MPF-mediated repression. Here we show that PAP also contains four nearby non-consensus cdk sites that are phosphorylated by MPF. Remarkably, full phosphorylation of all these cdk sites was required for repression of PAP activity, and partial phosphorylation had no detectable effect. The consensus sites were phosphorylated in vitro at a 10-fold lower concentration of MPF than the non-consensus sites. Consistent with this, during meiotic maturation of Xenopus oocytes, consensus sites were phosphorylated prior to the non-consensus sites at metaphase of meiosis I, and remained so throughout maturation, while the non-consensus sites did not become fully phosphorylated until after 12 h of metaphase II arrest. We propose that PAP's multiple cdk sites, and their differential sensitivity to MPF, provide a mechanism to link repression specifically to late M-phase. We discuss the possibility that this reflects a general means to control the timing of cdk-dependent regulatory events during the cell cycle. 相似文献

Summary We examined variation in diet choice by marten (Martes americana) among seasons and between sexes and ages from 1980–1985. During this period prey populations crashed simultaneously, except for ruffed grouse (Bonasa umbellus) which was common at the beginning and end of the study, and masked shrews (Sorex cinereus) which were abundant in 1983. Marten were catholic in selection of prey and made use of most available mammalian prey, ruffed grouse, passerine birds, berries, and insects. Diet niche was widest during the latter three years when prey was scare, particularly in late winter. Diet niche breadth was negatively correlated with abundance of all common prey species. Proportion of small prey species in the diet was correlated with absolute abundance of those species, but proportion of some large prey was related to their relative abundance. Diet choice varied among years and among seasons. Berries and insects were common in summer diets while large prey, particularly varying hare (Lepus americanus), were more frequent in winter diet than in summer diet. We found little evidence that any small mammal species was a preferred prey. Sexual size dimorphism between the sexes did not affect prey choice, nor did age. Reduced foraging effort in winter resulted in a wider diet niche only when prey was scarce. The only prediction of optimal foraging models fully supported by our data was a wider diet niche with reduced prey abundance. However, among the three most profitable prey species choice was dependent on the absolute abundance of the most profitable type (varying hare). We suggest that marten primarily forage for large prey but employ a strategy which results in encounters with small prey as well. Synopsis The field component of this study compared feeding preferences of Umbra limi with available resources in the environment. The mudminnows preferred benthic coleopterans, anisopterans, and amphipods while avoiding planktonic and other benthic food types. The laboratory component tested for possible switching behaviour by offering different ratios of two food types (Enchytraeus worms and Culex larvae). Both components indicated a benthic feeding preference in mudminnows. Although preferences were variable across individuals, switching did not occur, a result consistent with recent predictions on predatory behaviour. Feeding patterns reflected individual differences among fish, runs of feeding on one food type, and a constancy in the total number of the two food types eaten. 相似文献

A unique playback system was used to exaggerate continuously the number of calls heard in royal penguin, Eudyptes schlegeli, colonies to determine whether social facilitation increases a female's exposure to both sexual and agonistic activities, a process that could influence the timing of reproduction in colonies. We also determined whether the source of colony sound or the phase of the breeding cycle influenced the facilitation of sexual or agonistic activities. The system recorded and then played a new 12-min colony sound sample every 24 min throughout the first four phases of the breeding cycle. Because each broadcast was unique, this playback system avoids the possibility of habituation and can modify the acoustic environment in social groups for extended periods of time. The activities of penguins in control colonies (with no playback) were compared with those in colonies that heard samples derived from either their own or a different colony. Colony sound facilitated both sexual and agonistic activities from male and female royal penguins during all four phases of the cycle that we examined. Playbacks from the perceiver's own colony facilitated a greater response than sound originating from a different colony. Studies comparing egg-laying dates in colonies exposed continuously to the sounds of agonistic activities with those in control colonies, or colonies exposed continuously to the sounds of sexual activities are now needed to determine whether the facilitation of agonistic activities opposes the effects of social stimulation on the hormonal changes leading to ovulation in colony-dwelling birds.
