Cabbage plants were grown in soil amended with Clandosan (CLA) prepared from crustacean chitin (0.3% w/w). The plants were maintained in constant temperature tanks set to 15° or 30°C, in soils naturally infested with cyst nematodeHeterodera schachtii, or inoculated with the root-knot nematode,Meloidogyne javanica, respectively. At 30°C, after the first month following inoculation, CLA caused an increase in top fresh weight of plants but no reduction in nematode—induced root galling was recorded. However, when fresh plants were planted, CLA induced a large reduction in gall formation and caused an increase in top fresh weight of nematode-inoculated plants. At 15°C, CLA significantly affected the plants only after 60 days: an increase in top fresh weight and a reduction in the number of eggs per cyst were recorded. Ammonium was not detected in soil after 30 days, at 30°C, whereas at 15°C, CLA-treated soil contained twice as much ammonium as non-treated soil. After 60 days, ammonium was not detected at all. After 30 days nitrate concentrations in soil attained higher values at 30°C than at 15°C, whereas after 60 days high levels were detected only at 15°C. At 30°C, CLA induced an increase in the number of fungi, chitinolytic bacteria, and total amount of bacteria; at 15°C, such an increase was detected only with the chitinolytic microorganisms.Contribution from the Agricultural Research Organization (ARO), Bet Dagan, Israel No. 2196-E, 1987 series.  相似文献

The effects of loading cardiopulmonary baroreceptors on basal norepinephrine and renin activity were studied in six normal subjects. Loading of cardiopulmonary baroreceptors was accomplished by a 60-min 30 degrees head-down tilt with small supplemental saline infusions. Central venous pressure was measured continuously by intrathoracic catheter; arterial pressure was measured indirectly by cuff. During the tilt, central venous pressure increased from 5.1 +/- 1.3 to 8.9 +/- 1.7 mmHg (P less than 0.001), whereas arterial pressure was unchanged. Plasma norepinephrine (185 +/- 85 pg/ml) and plasma renin activity (3.9 +/- 5.7 ng . ml-1 . h-1) did not change. Moderate sustained loading of cardiopulmonary baroreceptors is therefore without effect on unstressed plasma norepinephrine and renin activity in normal humans, suggesting that the tonic inhibitory effects of these receptors on these neurohumoral control systems are not readily increased in the basal state.  相似文献

The mouse macrophage Fc gamma 2b/gamma 1R has previously been purified with the aid of the monoclonal antibody 2.4G2. That this Fc gamma R functions as a ligand-dependent ion channel is supported by the following evidence. Employing [3H]tetraphenylphosphonium ([3H]Ph4P+) as a probe for membrane potential changes in intact cells, we found a biphasic change in membrane potential following treatment with immune complexes, monoclonal antibody 2.4G2 IgG and 2.4G2 Fab-Sephadex particles. We observed an immediate depolarization followed by prolonged hyperpolarization. [3H]Ph4P+ uptake experiments with plasma membrane vesicles prepared from J774 macrophages showed that binding of ligands to the FcR led to transmembrane monovalent cation flow. Similar [3H]Ph4+ uptake experiments were done with phospholipid vesicles containing purified and reconstituted Fc gamma 2b/gamma. Following challenge with specific ligands, transmembrane monovalent cation flow was observed. Purified FcR was reconstituted into planar lipid bilayers; exposure to ligands led to transient bilayer conductance increase. THe conductance change was resolved into single channel events. Quin-2 measurements showed an increase of free cytosolic calcium levels in macrophages following exposure of cells to different ligands of the FcR. An optimal range of calcium was found to be required for phagocytosis, below and above which inhibition of ingestion occurred.  相似文献

Atelosteogenesis type II (AO II) is a neonatally lethal chondrodysplasia whose clinical and histological characteristics resemble those of another chondrodysplasia, the much less severe diastrophic dysplasia (DTD). The similarity suggests a shared pathogenesis involving lesions in the same biochemical pathway and perhaps the same gene. DTD is caused by mutations in the recently identified diastrophic dysplasia sulfate-transporter gene (DTDST). Here, we report that AOII patients also have DTDST mutations, which lead to defective uptake of inorganic sulfate and insufficient sulfation of macromolecules by patient mesenchymal cells in vitro. Together with our recent observation that a third even more severe chondrodysplasia, achondrogenesis type IB, is also caused by mutations in DTDST, these results demonstrate a phenotypic series of three chondrodysplasias of increasing severity caused by lesions in a single sulfate-transporter gene. The severity of the phenotype appears to be correlated with the predicted effect of the mutations on the residual activity of the DTDST protein.  相似文献

The metabolism of organic dormancy-breaking chemicals is poorly defined and may provide clues to their mode of action. Therefore, hydrated, dormant seeds of red rice ( Oryza sativa L.) were exposed to dormancy-breaking treatments of propionate-2-13C (22 m W ) or propanol-1-13C (75 m M ) for 24 h at 30°C. Embryo extracts were analyzed by 13C-nuclear magnetic resonance spectroscopy. Metabolism of propionate and propanol to 3-hydroxypropionate, an intermediate of the modified β-oxidation pathway, was detected after 2 and 4 h, respectively. This occurred prior to the onset of dormancy-breaking which required 12 h of chemical exposure. Accumulation of 3-hydroxypropionate was rapid and linear in the embryos of propionate-treated seeds. In the embryos of propanol-treated seeds, the level of 3-hydroxypropionate reached a plateau at 4 h. Following 24 h of contact with propionate, labeled citrate was detected in the embryos. The decrease in tissue pH associated with the dormancy-breaking process was fully accounted for by direct acid uptake and metabolic production of 3-hydroxypropionate.  相似文献

Fructose-2,6-bisphosphate (Fru-2,6-bisP) was evaluated as a potential marker for the dormancy-breaking phase or the germination phase before pericarp splitting in red rice (Oryza sativa). During 4 h of imbibition at 30[deg]C, Fru-2,6-bisP of dehulled dormant and nondormant seeds increased to 0.26 and 0.38 pmol embryo-1, respectively. In nondormant seeds, embryo Fru-2,6-bisP content remained stable until the onset of pericarp splitting (12 h) and increased rapidly thereafter. In dormant seeds, Fru-2,6-bisP declined to 0.09 pmol embryo-1 at 24 h. Embryo Fru-2,6-bisP was correlated with O2 uptake of dormant and nondormant seeds. A 24-h exposure of dehulled, water-imbibed, dormant seeds to treatments yielding >90% germination (sodium nitrite [4 mM], propionic acid [22 mM], methyl propionate [32 mM], propanol [75 mM], and propionaldehyde [40 mM]) led to changes in embryo Fru-2,6-bisP that were unrelated to the final germination percentages. Furthermore, a 2-h pulse of propionaldehyde increased Fru-2,6-bisP 4-fold but did not break dormancy. Whereas nitrite and propionaldehyde increased Fru-2,6-bisP to 0.33 pmol embryo-1 after 2 h of contact, propionic acid and methyl propionate did not increase Fru-2,6-bisP above the untreated control. In all cases, further increases in Fru-2,6-bisP occurred after pericarp splitting. However, the plateau Fru-2,6-bisP attained during chemical contact was inversely correlated with elapsed time to 30% germination (r = -0.978). Therefore, although Fru-2,6-bisP is not a universal marker for dormancy release, its rapid increase during nitrite and propionaldehyde treatments suggests that events associated with dormancy breaking can occur within 2 h of chemical treatment.  相似文献

Physiol. 83(1): 291-296, 1997.Ultrasound has been used to measure diaphragm thickness(Tdi) in thearea where the diaphragm abuts the rib cage (zone of apposition).However, the degree of diaphragm thickening during inspiration reportedas obtained by one-dimensional M-mode ultrasound was greater than thatpredicted by using other radiographic techniques. Becausetwo-dimensional (2-D) ultrasound provides greater anatomic definitionof the diaphragm and neighboring structures, we used this technique toreevaluate the relationship between lung volume andTdi. We firstestablished the accuracy and reproducibility of 2-D ultrasound bymeasuring Tdiwith a 7.5-MHz transducer in 26 cadavers. We found thatTdi measured byultrasound correlated significantly with that measured by ruler (R2 = 0.89), withthe slope of this relationship approximating a line of identity(y = 0.89x + 0.04 mm). The relationship between lung volume andTdi was thenstudied in nine subjects by obtaining diaphragm images at the fivetarget lung volumes [25% increments from residual volume (RV) tototal lung capacity (TLC)]. Plots ofTdi vs. lungvolume demonstrated that the diaphragm thickened as lung volumeincreased, with a more rapid rate of thickening at the higher lungvolumes[Tdi = 1.74 vital capacity (VC)2 + 0.26 VC + 2.7 mm] (R2 = 0.99; P < 0.001) where lung volumeis expressed as a fraction of VC. The mean increase inTdi between RVand TLC for the group was 54% (range 42-78%). We conclude that2-D ultrasound can accurately measureTdi and that theaverage thickening of the diaphragm when a subject is inhaling from RVto TLC using this technique is in the range of what would be predictedfrom a 35% shortening of the diaphragm.

Many chemically dissimilar substances break dormancy of seeds, but the relationship between chemical structure and physiological activity is unknown. In this study, the concentrations of organic acids, esters, aldehydes, alcohols, and inorganic weak acids required to elicit 50% germination of initially dormant, dehulled red rice seeds (Oryza sativa) were determined. The activity of most substances was very highly and inversely correlated to lipophilicity as measured by octanol/water partition coefficients; chemicals with the highest partition coefficients required the lowest concentrations to elicit the germination response. Relative efficacy was also dependent upon the functional group; generally, monocarboxylic acids were more effective than aldehydes, esters, hydroxyacids, and alcohols. Relative hydrophobicity plots supported a modulating role of the functional group. Dormancy-breaking activity of methyl formate, formic acid, nitrite, azide, and cyanide was higher than predicted based on lipophilicity and apparently was related to molecular size; compounds with smaller molecular widths were required at lower concentrations to achieve the 50% germination response.  相似文献

The tryptic hydrolysis of bivine intestinal calcium-binding protein in the presence and absence of excess calcium has been investigated. Calcium-binding activity and immunological reactivity of the protein were not significantly affected in the presence of 1.0 mM CaCl2 following 24 h incubation at 38 degrees C with trypsin at ratios of 1:9 of enzyme to calcium-binding protein. Some modification of the protein did occur under these conditions, however, since analysis by analytical acrylamide gel electrophoresis indicated the formation of a more rapidly-migrating species from the slower-moving original protein band. Omission of added calcium from the incubation medium resulted in rapid and essentially complete destruction of calcium-binding activity and immunological reactivity, and the formation of peptides of low molecular weight. This provides evidence that the conformation of the calcium-binding protein in the presence of calcium differs from that in its absence.  相似文献
