High-molecular-mass polysaccharides were released by mild acid degradation of the lipopolysaccharides of two wild-type Vibrio vulnificus strain, a flagellated motile strain CECT 5198 and a non-flagellated non-motile strain S3-I2-36. Studies by sugar analysis and partial acid hydrolysis along with 1H and 13C NMR spectroscopies showed that the polysaccharides from both strains have the same trisaccharide repeating unit of the following structure:→4)-β-d-GlcpNAc3NAcylAN-(1→4)-α-l-GalpNAmA-(1→3)-α-d-QuipNAc-(1→where QuiNAc stands for 2-acetamido-2,6-dideoxyglucose, GalNAmA for 2-acetimidoylamino-2-deoxygalacturonic acid, GlcNAc3NAcylAN for 2-acetamido-3-acylamino-2,3-dideoxyglucuronamide and acyl for 4-d-malyl (∼30%) or 2-O-acetyl-4-d-malyl (∼70%). The structure of the polysaccharide studied resembles much that of a marine bacterium Pseudoalteromonas rubra ATCC 29570 reinvestigated in this work. The latter differs in (i) the absolute configuration of malic acid (l vs d), (ii) 3-O-acetylation of GalNAmA and (iii) replacement of QuiNAc with its 4-keto biosynthetic precursor.  相似文献

A video camera mounted in an underwater housing and remotely operated was used to monitor the behaviour of five different Sakhalin taimen (Parahucho perryi), females and attendant males spawning in three coastal tributary streams in Northern Hokkaido, Japan. Based on three complete and two incomplete spawnings, we describe in detail for the first time the complete spawning behavioural repertoire of this species. The Sakhalin taimen was originally placed within Hucho, then removed from that genus based on morphological, life history and molecular data. Our study supports that removal—none of the behavioural traits we recorded clustered Parahucho with Hucho uniquely. Similarities between the two genera were all plesiomorphic traits that are widespread throughout the salmonines. The immediate behaviour right after spawning was found to be a major difference between Hucho and Parahucho. Like female Oncorhynchus and Salmo, Sakhalin taimen females cover their eggs by beats of their tails immediately after spawning. This is different from the “rest, then cover” behaviour shown by Siberian taimen (Hucho taimen) as well as lenok (Brachymystax lenok), supporting again that the Sakhalin taimen be removed from Hucho and placed in its own genus.  相似文献

Contradictory findings regarding the gene expression of the main lipogenic enzymes in human adipose tissue depots have been reported. In this cross‐sectional study, we aimed to evaluate the mRNA expression of fatty acid synthase (FAS) and acetyl‐CoA carboxilase (ACC) in omental and subcutaneous (SC) fat depots from subjects who varied widely in terms of body fat mass. FAS and ACC gene expression were evaluated by real time‐PCR in 188 samples of visceral adipose tissue which were obtained during elective surgical procedures in 119 women and 69 men. Decreased sex‐adjusted FAS (?59%) and ACC (?49%) mRNA were found in visceral adipose tissue from obese subjects, with and without diabetes mellitus type 2 (DM‐2), compared with lean subjects (both P < 0.0001). FAS mRNA was also decreased (?40%) in fat depots from overweight subjects (P < 0.05). Indeed, FAS mRNA was significantly and positively associated with ACC gene expression (r = 0.316, P < 0.0001) and negatively with BMI (r = ?0.274), waist circumference (r = ?0.437), systolic blood pressure (r = ?0.310), serum glucose (r = ?0.277), and fasting triglycerides (r = ?0.226), among others (all P < 0.0001). Similar associations were observed for ACC gene expression levels. In a representative subgroup of nonobese (n = 4) and obese women (n = 6), relative FAS gene expression levels significantly correlated (r = 0.657, P = 0.034; n = 10) with FAS protein values. FAS protein levels were also inversely correlated with blood glucose (r = ?0.640, P = 0.046) and fasting triglycerides (r = ?0.832, P = 0.010). In conclusion, the gene expression of the main lipogenic enzymes is downregulated in visceral adipose tissue from obese subjects.  相似文献

Abstract Several purple and green sulfur bacteria (genera Chromatium, Thiocapsa and Chlorobium ) were tested for their sensitivity to different antimicrobial agents by a disc diffusion assay, using thioacetamide as a source of hydrogen sulfide for plate growth. Chlorobium limicola strains were more sensitive to amoxicillin, erythromycin and nalidixic acid, whereas gentamicin and netilmicin were more active against the purple bacteria tested. None of the organisms were sensitive to oxacillin and trimethoprim + sulfamethoxazole. The critical concentrations at the edge of the inhibition zone were also calculated for three organisms and the antimicrobials colistin, mitomycin C, penicillin G, rifampicin, and streptomycin. The results obtained suggest that colistin, mitomycin C, penicillin G would provide selective conditions against the growth of Chlorobium limicola strains, while streptomycin and other aminoglycoside antibiotics would select against purple bacteria.  相似文献

The content and accretion of fatty acids in 30, 45 and 60-day old Wistar rats fed either reference chow or a cafeteria diet has been studied, together with their actual fatty acid intake during that period. Diet had a small overall effect on the pattern of deposition of fatty acids, but the deposition of fat was much higher in cafeteria rats. The fat-rich cafeteria diet allowed the direct incorporation of most fatty acids into lipid storage, whilst chow-feeding activated lipogenesis and the deposition of a shorter chain and more saturated type of fatty acids. During the second month of the rat's life, the elongation pathway as well as 9-desaturase became functional, thus helping to shape the pattern of fatty acids actually accrued. The 60-day rats showed a relative impairment in the operation of 5-desaturase, since their lipids had a higher C20:4/C20:3 ratio than those of the diet ingested. Cafeteria-diet feeding minimized this effect since the large supply of dietary polyunsaturated fatty acids made the operation of the elongation-desaturase pathways practically unnecessary.  相似文献

During the last decades, agricultural intensification has modified the hydrology of Mediterranean wetlands, as has occurred in the Mar Menor coastal lagoon (SE Spain). Salt-steppe dominated wetlands, characteristic of transitional areas surrounding this lagoon and rich in biodiversity values, are threatened by changes in their water regime originated by land-use changes in the watershed. Traditional dryland cultures have also been replaced by irrigated ones. We assess the direct and indirect changes induced by agriculture on a terrestrial vertebrate community (steppe birds) especially sensitive to these ecosystem changes. This is made on the basis of several surveys of terrestrial birds (excluding aerial feeders and raptors) carried out between 1984 and 2008 in a representative wetland of the lagoon's continental margin (Marina del Carmolí). The changes in this bird assemblage reflect the hydrological modifications induced by agriculture at the watershed scale, which have significant effects on the relative representation of wetland habitats. Bird metrics and indices (species abundance, taxonomic composition, conservation value) describe these community changes as the combination of early declines in some species and families, and transient or late increases in other. In the long term, the family Alaudidae (and particularly species like Melanocorypha calandra) have lost importance to the benefit of Turdidae and Fringillidae. The area of salt-steppe explains a large part of the variation in the abundance of Alaudidae, while most variation of Turdidae and Fringillidae respond to the area of saltmarsh. Some Alaudidae seem to take advantage of the intermediate stages of saltmarsh expansion (Calandrella rufescens), or from the marginal irrigated crops fringing the wetland (Calandrella brachydactyla) that could compensate the loss of original agricultural habitats. Habitat changes in the wetland have occurred in three differentiated stages, and modify the steppe bird community towards a more heterogeneous assemblage including scrubland and palustrine species. Among three indices of ornithological value, only that based on the EU Bird's Directive Annex I species was negatively affected, but since the wetland has been designated a Specially Protected Area under this regulation, this represents a management failure. There exists some chance, however, to manage peripheral cropland in favour of biodiversity. The importance of monitoring in conservation evaluation and management is also stressed, since the terrestrial bird community of this wetland has not been regularly surveyed. In fact, its evaluation against the Bird's Directive criteria was made in a period of quick departure from the original, good ecological state of the wetland.  相似文献

Objective: To establish whether single daily oral doses of oleoyl‐estrone result in dose‐dependent slimming effects on normal weight rats, and to determine the changes in energy parameters induced by this treatment. Research Methods and Procedures: The effects of a daily oral gavage of oleoyl‐estrone (0, 0.2, 0.5, 1, 2, 5, 10, and 20 μmol/kg per day) in 0.2 ml of sunflower oil given over a 10‐day period were studied in groups, each of which contained six adult female Wistar rats initially weighing 190 to 230 g. A group of intact control rats receiving no gavage was included for comparison. Body weight and food intake were measured daily. Rats were killed on day 10 of treatment, and body composition (protein nitrogen, lipids, and water), liver lipids, and plasma parameters (glucose, triacylglycerols, total cholesterol, free fatty acids, 3‐hydroxybutyrate, urea, aspartate, alanine transaminases, insulin, leptin, and free and acyl‐estrone) were measured. Results: The administration of oleoyl‐estrone resulted in a dose‐dependent loss of body fat, because of a partly maintained energy expenditure combined with decreased food intake. The differences in the energy budget were met by internal fat pools. The changes recorded did not affect the levels of the main plasma energy homeostasis indicators: unaltered glucose, triacylglycerols, free fatty acids, 3hydroxybutyrate, and urea. Protein was accrued even under conditions of severe lipid store drainage. There were no changes in transaminases. No lipid accumulation was recorded in the liver. Plasma insulin and leptin levels decreased with increased oleoyl‐estrone doses, whereas the levels of free and esterified estrone increased with treatment, although not in proportion to the dose received. Discussion: Oral treatment with oleoyl‐estrone resulted in the specific dose‐related loss of fat reserves with little change to other metabolic parameters. These results agree with the postulated role of oleoyl‐estrone as a ponderostat signal.  相似文献

Human GTPBP3 is an evolutionarily conserved, multidomain protein involved in mitochondrial tRNA modification. Characterization of its biochemical properties and the phenotype conferred by GTPBP3 inactivation is crucial to understanding the role of this protein in tRNA maturation and its effects on mitochondrial respiration. We show that the two most abundant GTPBP3 isoforms exhibit moderate affinity for guanine nucleotides like their bacterial homologue, MnmE, although they hydrolyze GTP at a 100-fold lower rate. This suggests that regulation of the GTPase activity, essential for the tRNA modification function of MnmE, is different in GTPBP3. In fact, potassium-induced dimerization of the G domain leads to stimulation of the GTPase activity in MnmE but not in GTPBP3. The GTPBP3 N-terminal domain mediates a potassium-independent dimerization, which appears as an evolutionarily conserved property of the protein family, probably related to the construction of the binding site for the one-carbon-unit donor in the modification reaction. Partial inactivation of GTPBP3 by small interfering RNA reduces oxygen consumption, ATP production, and mitochondrial protein synthesis, while the degradation of these proteins slightly increases. It also results in mitochondria with defective membrane potential and increased superoxide levels. These phenotypic traits suggest that GTPBP3 defects contribute to the pathogenesis of some oxidative phosphorylation diseases.  相似文献

In order to detect xenoestrogens which induce perturbations of mammalian cells, design of biosensor using a mammalian cell line enable to detect these compounds is necessary. MELN cell line is suitable to detect estrogen activity, since they are stably transfect with an estrogen regulated luciferase gene. To realize this biosensor, it appeared necessary to add a protection to the mamalian cell, which is devoided, of the wall protecting yeasts or plant cells. With this aim in view, MELN cells have been isolated with a polyelectrolyte shell using the layer-by-layer technique. Among several polyelectrolyte-couples, the best cell survival (>80%) was obtained by alternating the polycation poly-diallyldimethyl ammonium chloride layer and the negatively charged poly-styrene sulfonate. We observed that the composition of the buffer used for layer-deposition was crucial to preserving cell viability, e.g. potassium ions were preferred to sodium ions during the coating. Furthermore, viability was increased when cells were allowed to recover for 2 h between each bilayer deposition. The use of engineered mammalian cells that synthesize luciferase as a response to exposure to estradiol, demonstrated that coating not only permits cell survival, but also allows essential metabolic functions, such as RNA and protein synthesis to take place. Capsule formation allows free diffusion of small molecules, while it prevents internalization in the cells of proteins larger than 60 kDa.  相似文献
