A rat spleen cDNA library was screened for clones carrying the cDNAs for prothymosin alpha and parathymosin. Sequence analysis of a clone carrying the entire coding region for prothymosin alpha confirmed and completed the amino acid sequence for this polypeptide and established the number of amino acid residues as 111. Rat prothymosin alpha differs from human prothymosin alpha at six positions, including four substitutions and two insertions. The nucleotide sequences of the cDNAs for the rat and human polypeptides are more than 90% identical in the open reading frames, with significant homology extending into the 5' and 3' flanking regions. From the same library, we also isolated a clone carrying 80% of the coding region for rat parathymosin. The number of amino acid residues in rat parathymosin is 101, based on the sequence deduced from the cDNA insert and earlier information on the sequence in the amino-terminal portion of this polypeptide. Despite their similarity in size and amino acid composition, rat prothymosin alpha and rat parathymosin show only limited sequence homology, primarily in the segment including residues 14 through 25, where 10 of 12 positions are identical in the two polypeptides. this is also the region of significant sequence similarity to a 12-amino-acid segment in the p17 protein of the human immunodeficiency disease associated virus (HTLV-IIIB).  相似文献

Phorbol myristate acetate (PMA)-stimulated human monocyte-like cells (U-937) were found to synthesize the third component of complement (C3), as shown by enzyme-linked immunosorbent assay and immunoprecipitation from [35S]methionine-labelled culture supernatants. C3 synthesis occurred at a rate of about 160 ng of C3/24 h per 10(6) cells on day 7 after addition of PMA; it was blocked by cycloheximide treatment and was restored after removal of the inhibitor. SDS/polyacrylamide-gel-electrophoretic analysis of the immunoprecipitated protein showed that the size and subunit structure of the newly synthesized C3 were identical with those of plasma C3, and that a single-chain intracellular precursor was present in the cell lysates. Haemolytic assays showed that the synthesized C3 fully expressed functional activity in early culture within 4 h. After longer culture, a loss of haemolytic activity was observed. The possibility that newly secreted C3 is cleaved by U-937 cells themselves was suggested.  相似文献

Alterations in prostatic acid phosphatase (PAcP), a phosphotyrosyl phosphatase, corresponded to changes in overall tyrosyl kinase activity. PAcP added to extracts of prostate carcinoma cells with a low endogenous level of PAcP activity and elevated tyrosyl kinase activity decreased the tyrosyl kinase activity. On the other hand, when PAcP activity was decreased by the addition of androgens to cells, there was a corresponding increase in tyrosyl kinase activity.  相似文献

Uptake of labeled amino acids occurred at –4 C to 50 C accompanied by amino acid pool formation and protein synthesis. Maximum assimilation rates of both amino acids occurred at a temperature at which growth of this yeast was inhibited. Over a wide range of temperature the organism took up more exogenous lysine than glutamic acid, even though glutamic acid was present in the cellular protein in greater quantities. At 25 C the uptake and incorporation rates of glutamic acid was significantly higher than at 3 C; however, the size of the glutamic acid pools, at these two temperatures, appeared to be equal and independent of temperature.  相似文献

The general fine structure of the giant coenocyte Caulerpa prolifera (Forsskål) Lamouroux is presented. The cytoplasm forms a parietal layer throughout the plant without any regular membranous separations within organs or between organs. The vacuome is similar in structure. There are distinct patterns of organelle distribution in the highly polar cytoplasm of rhizoids and blades. The organelles are compared to those of other members of the division Chlorophyta and are typical. Amyloplast structure is compared to that of chloroplasts. A possible developmental sequence from chloroplast buds through an unusual circular body is suggested.  相似文献
