AIMS: The goal of this study was to determine whether nisin and lactoferrin would act synergistically to inhibit the growth of Listeria monocytogenes and Escherichia coli O157:H7. METHODS AND RESULTS: Lactoferrin and nisin separately or in combination were suspended in peptone yeast glucose broth and following inoculation with L. monocytogenes or E. coli O157:H7 growth inhibition of each pathogen was determined. At 1000 microg ml(-1) lactoferrin L. monocytogenes was effectively inhibited. However, E. coli O157:H7 initially was inhibited and then grew to cell density similar to the control. A combination of 500 microg ml(-1) of lactoferrin and 250 IU ml(-1) of nisin effectively inhibited the growth of E. coli O157:H7, whereas, 250 microg ml(-1) of lactoferrin and 10 IU ml(-1) of nisin were inhibitory to L. monocytogenes. CONCLUSIONS: The results suggest that lactoferrin and nisin act synergistically to inhibit the growth of L. monocytogenes and E. coli O157:H7. SIGNIFICANCE AND IMPACT OF THE STUDY: Natural preservatives that are active against gram-positive and gram-negative pathogens are desirable to the food industry and consumers. This study demonstrates that lactoferrin and nisin work synergistically reducing the levels required independently inhibiting growth of two major foodborne pathogens. Previous reported results indicated a low level of antimicrobial activity; however, this work was not performed in low divalent cation concentration media. It has been suggested that nondivalent cation-limiting medium such as trypticase soy broth (TSB), can reduce or completely eliminate the inhibitory activity. Further knowledge of these interactions can increase the understanding of the antimicrobial activity of lactoferrin. This should make the use of these compounds by industry more attractive.  相似文献

Aflatoxins are toxic and carcinogenic secondary metabolites produced by the fungi Aspergillus flavus and Aspergillus parasiticus. To better understand the molecular mechanisms that regulate aflatoxin production, the biosynthesis of the toxin in A. flavus and A. parasticus grown in yeast extract sucrose media supplemented with 50 mM tryptophan (Trp) were examined. Aspergillus flavus grown in the presence of 50 mM tryptophan was found to have significantly reduced aflatoxin B1 and B2 biosynthesis, while A. parasiticus cultures had significantly increased B1 and G1 biosynthesis. Microarray analysis of RNA extracted from fungi grown under these conditions revealed 77 genes that are expressed significantly different between A. flavus and A. parasiticus, including the aflatoxin biosynthetic genes aflD (nor-1), aflE (norA), and aflO (omtB). It is clear that the regulatory mechanisms of aflatoxin biosynthesis in response to Trp in A. flavus and A. parasiticus are different. These candidate genes may serve as regulatory factors of aflatoxin biosynthesis.  相似文献

Hirano bodies are paracrystalline F-actin-rich structures associated with diverse conditions, including neurodegeneration and aging. Generation of model Hirano bodies using altered forms of Dictyostelium 34-kDa actin-bundling protein allows studies of their physiological function and mechanism of formation. We describe a novel 34-kDa protein mutant, E60K, with a point mutation within the inhibitory domain of the 34-kDa protein. Expression of E60K in Dictyostelium induces the formation of model Hirano bodies. The E60K protein has activated actin binding and is calcium regulated, unlike other forms of the 34-kDa protein that induce Hirano bodies and that have activated actin binding but lack calcium regulation. Actin filaments in the presence of E60K in vitro show enhanced resistance to disassembly induced by latrunculin B. Actin filaments in model Hirano bodies are also protected from latrunculin-induced depolymerization. We used nocodazole and blebbistatin to probe the role of the microtubules and myosin II, respectively, in the formation of model Hirano bodies. In the presence of these inhibitors, model Hirano bodies can form but are smaller than controls at early times of formation. The ultrastructure of model Hirano bodies did not reveal any major difference in structure and organization in the presence of inhibitors. In summary, these results support the conclusion that formation of model Hirano bodies is promoted by gain-of-function actin filament bundling, which enhances actin filament stabilization. Microtubules and myosin II contribute to but are not required for formation of model Hirano bodies.  相似文献

Centromere protein E (CENP-E) is a highly elongated kinesin that transports pole-proximal chromosomes during congression in prometaphase. During metaphase, it facilitates kinetochore–microtubule end-on attachment required to achieve and maintain chromosome alignment. In vitro CENP-E can walk processively along microtubule tracks and follow both growing and shrinking microtubule plus ends. Neither the CENP-E–dependent transport along microtubules nor its tip-tracking activity requires the unusually long coiled-coil stalk of CENP-E. The biological role for the CENP-E stalk has now been identified through creation of “Bonsai” CENP-E with significantly shortened stalk but wild-type motor and tail domains. We demonstrate that Bonsai CENP-E fails to bind microtubules in vitro unless a cargo is contemporaneously bound via its C-terminal tail. In contrast, both full-length and truncated CENP-E that has no stalk and tail exhibit robust motility with and without cargo binding, highlighting the importance of CENP-E stalk for its activity. Correspondingly, kinetochore attachment to microtubule ends is shown to be disrupted in cells whose CENP-E has a shortened stalk, thereby producing chromosome misalignment in metaphase and lagging chromosomes during anaphase. Together these findings establish an unexpected role of CENP-E elongated stalk in ensuring stability of kinetochore–microtubule attachments during chromosome congression and segregation.  相似文献

Two steroidal saponins have been purified from cayenne pepper (Capsicum frutescens). Both have the same steroidal moiety but differ in the number of glucose moieties: the first saponin has four glucose moieties (molecular mass 1081 Da) and the second contains three glucose moieties (molecular mass 919 Da). Solubility in aqueous solution is less for the saponin containing three glucose moieties than for the one containing four glucose moieties. The larger saponin was slightly fungicidal against the nongerminated and germinating conidia of Aspergillus flavus, A. niger, A. parasiticus, A. fumigatus, Fusarium oxysporum, F. moniliforme, and F. graminearum, whereas, the second saponin (molecular mass 919 Da) was inactive against these fungi. Results indicate that the absence of one glucose molecule affects the fungicidal and aqueous solubility properties of these similar molecules.  相似文献

为阐明青海省森林生态系统乔木层植被碳储量现状及其分布特征, 该研究利用240个标准样地实测的乔木数据, 估算出青海省森林生态系统不同林型处于不同龄级阶段的平均碳密度, 并结合青海省森林资源清查资料所提供的不同龄级的各林型面积, 估算了青海省森林生态系统乔木层的固碳现状、速率和潜力。结果表明: 1) 2011年青海省森林乔木层平均碳密度为76.54 Mg·hm -2, 总碳储量为27.38 Tg。云杉(Picea spp.)林、柏木(Cupressus funebris)林、桦木(Betula spp.)林、杨树(Populus spp.)林是青海地区的主要林型, 占青海省森林面积的96.23%, 占青海省乔木层碳储量的86.67%, 其中云杉林的碳储量(14.78 Tg)和碳密度(106.93 Mg·hm -2)最高。按龄级划分, 乔木层碳储量表现为过熟林>中龄林>成熟林>近熟林>幼龄林。2)青海省乔木层总碳储量从2003年的23.30 Tg增加到2011年的27.38 Tg, 年平均碳增量为0.51 Tg·a -1。乔木层固碳速率为1.06 Mg·hm -2·a -1, 其中柏木林的固碳速率最大(0.44 Mg·hm -2·a -1); 桦木林的固碳速率为负值(-1.06 Mg·hm -2·a -1)。3)青海省乔木层植被固碳潜力为8.50 Tg, 其中云杉林固碳潜力最高(3.40 Tg)。该研究结果表明青海省乔木层具有较大的固碳潜力, 若对现有森林资源进行合理管理和利用, 将会增加青海省森林的碳固存能力。  相似文献

Tropical rainforests are reservoirs of terrestrial carbon and biodiversity. Large and often emergent trees store disproportionately large amounts of aboveground carbon and greatly influence the structure and functioning of tropical rainforests. Despite their importance, controls on the abundance and distribution of emergent trees are largely unknown across tropical landscapes. Conventional field approaches are limited in their ability to characterize patterns in emergent trees across vast landscapes with varying environmental conditions and floristic composition. Here, we used a high‐resolution light detection and ranging (LiDAR) sensor aboard the Carnegie Airborne Observatory Airborne Taxonomic Mapping System (CAO‐AToMS) to examine the abundance and distribution of tall emergent tree crowns (ETC) relative to surrounding tree crowns (STC) across the Osa Peninsula, a geologically and topographically diverse region of Costa Rica. The abundance of ETC was clearly influenced by fine‐scale topographic variation, with distribution patterns that held across a variety of geologic substrates. Specifically, the density of ETC was much greater on lower slopes and in valleys, compared to upper slopes and ridges. Furthermore, using the CAO high‐fidelity imaging spectrometer, ETC had a different spectral signature than that of STC. Most notably, ETC had lower remotely sensed foliar nitrogen than STC, which was verified with an independent field survey of canopy leaf chemistry. The underlying mechanisms to explain the topographic‐dependence of ETCs and linkages to canopy N are unknown, and remain an important area of research.  相似文献

为了了解滨海湿地土壤中溶解性有机质的结构特征及来源,2014年1月在胶州湾采集光滩、碱蓬、芦苇和大米草湿地的土壤样品,测定土壤溶解性有机质(DOM)含量,利用三维荧光技术进行光谱分析.结果表明: 胶州湾4种湿地类型土壤溶解性有机碳(DOC)含量表现为大米草湿地>光滩>碱蓬湿地>芦苇湿地,垂直剖面上随土层深度的增加DOC含量均呈减少的趋势.经光谱分析,胶州湾湿地土壤DOM的三维荧光光谱(3DEEMs)中出现了B、T、A、D和C等5种荧光峰,分别代表类酪氨酸、类色氨酸、类富里酸、类可溶性微生物副产物和类腐殖酸5种组分.利用荧光区域积分(FRI)法对5种组分进行定量分析,类色氨酸、类富里酸和类酪氨酸在DOM各组分含量中居前3位,类可溶性微生物副产物和类腐殖酸的含量次之,二者含量差异不显著.DOM的5种组分相互之间均呈显著正相关,与DOC含量呈显著正相关,与总磷、有效磷、总氮有不同程度的相关性.胶州湾4种湿地类型土壤DOM主要由生物相互作用内源产生,且腐殖化程度较低.  相似文献
