割手密作为现代甘蔗遗传杂交育种史上最为成功的野生亲本,对多种不良环境都具有很强的抗逆性,被公认为是抗逆基因的主要来源。但目前真正被有效利用的割手密抗逆亲本和抗逆基因非常有限,我国自育和引进甘蔗主栽品种的抗逆性仍然比较单一且普遍偏弱,因此加强割手密优良抗逆亲本筛选和抗逆基因挖掘利用研究意义重大。本文综述了不同基因型割手密在非生物逆境(干旱、低温等理化因素)和生物逆境(病虫害侵染)下的抗逆性鉴定及其抗逆基因克隆和功能验证等国内外相关研究进展;并探讨了当前割手密资源抗逆材料筛选和抗逆基因挖掘利用中存在的问题和今后的研究方向,希望为高效利用割手密优异抗逆基因资源开展甘蔗多抗逆性聚合育种提供参考。  相似文献

One robust result from many small-scale experiments has been that plant community productivity often increases with increasing plant diversity. Most frequently, resource-based or competitive interactions are thought to drive this positive diversity-productivity relationship. Here, we ask whether suppression of plant productivity by soil fungal pathogens might also drive a positive diversity-productivity relationship. We created plant assemblages that varied in diversity and crossed this with a ± soil fungicide treatment. In control (non-fungicide treated) assemblages there was a strong positive relationship between plant diversity and above-ground plant biomass. However, in fungicide-treated assemblages this relationship disappeared. This occurred because fungicide increased plant production by an average of 141% at the lower ends of diversity but boosted production by an average of only 33% at the higher ends of diversity, essentially flattening the diversity-productivity curve. These results suggest that soil pathogens might be a heretofore unappreciated driver of diversity-productivity relationships.  相似文献

Primary tropical rainforests are generally considered to be relatively nitrogen (N) rich, with characteristically large hydrologic and gaseous losses of inorganic N. However, emerging evidence suggests that some tropical ecosystems can exhibit tight N cycling, with low biologically available losses. In this study, we combined isotopic data with a well-characterized watershed N mass balance to close the N budget and characterize gaseous N losses at the ecosystem scale in a lowland tropical rainforest on the Osa Peninsula in southwestern Costa Rica. We measured δ15N and δ18O of nitrate (NO3 ?) in precipitation, surface, shallow and deep soil lysimeters and stream water biweekly for 1 year. Enrichment of both isotopes indicates that denitrification occurs predominantly as NO3 ? moves from surface soil down to 15 cm depth or laterally to stream water, with little further processing in deeper soil. Two different isotopic modeling approaches suggested that the gaseous fraction comprises 14 or 32% of total N loss (2.7 or 7.5 kg N ha?1 y?1), though estimates are sensitive to selection of isotopic fractionation values. Gas loss estimates using the mass balance approach (3.2 kg N ha?1 y?1) fall within this range and include N2O losses of 0.9 kg N ha?1 y?1. Overall, gaseous and soluble hydrologic N losses comprise a modest proportion (~ 25%) of the total N inputs to this ecosystem. By contrast, relatively large, episodic hydrologic losses of non-biologically available particulate N balance the majority of N inputs and may contribute to maintaining conservative N cycling in this lowland tropical forest. Similar patterns of N cycling may occur in other tropical forests with similar state factor combinations—high rainfall, steep topography, relatively fertile soils—such as the western arc of the Amazon Basin and much of IndoMalaysia, but this hypothesis remains untested.  相似文献

Independent of the approach used, the ability to correctly interpret tandem MS data depends on the quality of the original spectra. Even in the case of the highest quality spectra, the majority of spectral peaks can not be reliably interpreted. The accuracy of sequencing algorithms can be improved by filtering out such 'noise' peaks. Preprocessing MS/MS spectra to select informative ion peaks increases accuracy and reduces the processing time. Intuitively, the mix of informative versus non-informative peaks has a direct effect on the quality and size of the resulting candidate peptide search space. As the number of selected peaks increases, the corresponding search space increases exponentially. If we select too few peaks then the ion-ladder interpretation of the spectrum will contain gaps that can only be explained by permutations of combinations of amino acids. This will result in a larger candidate peptide search space and poorer quality candidates. The dependency that peptide sequencing accuracy has on an initial peak selection regime makes this preprocessing step a crucial facet of any approach, whether de novo or not, to MS/MS spectra interpretation.We have developed a novel approach to address this problem. Our approach uses a staged neural network to model ion fragmentation patterns and estimate the posterior probability of each ion type. Our method improves upon other preprocessing techniques and shows a significant reduction in the search space for candidate peptides without sacrificing candidate peptide quality.  相似文献

An organism, identified as Mycobacterium phlei GTIS10, was isolated based on its ability to use dibenzothiophene (DBT) as a sole source of sulfur for growth at 30-52 degrees C. Similar to other biodesulfurization-competent organisms, M. phlei GTIS10 converts DBT to 2-hydroxybiphenyl (2-HBP), as detected by HPLC. The specific desulfurization activity of the 50 degrees C M. phlei GTIS10 culture was determined to be 1.1+/-0.07 micromol 2-HBP min(-1) (g dry cell)(-1). M. phlei GTIS10 can also utilize benzothiophene and thiophene as sulfur sources for growth. The dszABC operon of M. phlei GTIS10 was cloned and sequenced and was found to be identical to that of Rhodococcus erythropolis IGTS8. The presence of the R. erythropolis IGTS8 120-kb plasmid pSOX, which encodes the dszABC operon, has been demonstrated in M. phlei GTIS10. Even though identical dsz genes are contained in both cultures, the temperature at which resting cells of R. erythropolisIGTS8 reach the highest rate of DBT metabolism is near 30 degrees C whereas the temperature that shows the highest activity in resting cell cultures of M. phlei GTIS10 is near 50 degrees C, and activity is detectable at temperatures as high as 57 degrees C. In M. phlei GTIS10, the rate-limiting step in vivo appears to be the conversion of DBT to dibenzothiophene sulfone catalyzed by the product of the dszC gene, DBT monooxygenase. The thermostability of individual desulfurization enzymes was determined and 2-hydroxybiphenyl-2-sulfinate sulfinolyase, encoded by dszB, was found to be the most thermolabile. These results demonstrate that the thermostability of individual enzymes determined in vitro is not necessarily a good predictor of the functional temperature range of enzymes in vivo.  相似文献

为了分析LITAF、RAB7、LMNA和MTMR2基因在中国人腓骨肌萎缩症(Charcot-Marie-Tooth disease, CMT)的突变特点, 文章分别应用PCR结合DNA序列分析方法和PCR-单链构象多态性(PCR-SSCP)结合DNA序列分析方法对6个常染色体显性遗传家系先证者和27个散发病例进行LITAF和RAB7基因突变分析; 应用PCR-SSCP结合DNA序列分析方法对14个常染色体遗传的CMT家系先证者和27个散发患者进行LMNA和MTMR2基因突变分析。结果发现: LITAF基因c.269G→A、c.274A→G序列变异和LMNA基因c.1243G→A、c.1910C→T序列变异, 未发现RAB7和MTMR2基因的序列变异。其中LITAF基因c.269G→A、LMNA基因c.1243G→A和c.1910C→T为新发现的单核苷酸多态; LITAF基因c.274A→G为已知多态。说明LITAF、RAB7、LMNA和MTMR2基因突变在中国人CMT患者中罕见。  相似文献

为了了解滨海湿地土壤中溶解性有机质的结构特征及来源,2014年1月在胶州湾采集光滩、碱蓬、芦苇和大米草湿地的土壤样品,测定土壤溶解性有机质(DOM)含量,利用三维荧光技术进行光谱分析.结果表明: 胶州湾4种湿地类型土壤溶解性有机碳(DOC)含量表现为大米草湿地>光滩>碱蓬湿地>芦苇湿地,垂直剖面上随土层深度的增加DOC含量均呈减少的趋势.经光谱分析,胶州湾湿地土壤DOM的三维荧光光谱(3DEEMs)中出现了B、T、A、D和C等5种荧光峰,分别代表类酪氨酸、类色氨酸、类富里酸、类可溶性微生物副产物和类腐殖酸5种组分.利用荧光区域积分(FRI)法对5种组分进行定量分析,类色氨酸、类富里酸和类酪氨酸在DOM各组分含量中居前3位,类可溶性微生物副产物和类腐殖酸的含量次之,二者含量差异不显著.DOM的5种组分相互之间均呈显著正相关,与DOC含量呈显著正相关,与总磷、有效磷、总氮有不同程度的相关性.胶州湾4种湿地类型土壤DOM主要由生物相互作用内源产生,且腐殖化程度较低.  相似文献

Radio frequency (RF) dosimetry is the quantification of the magnitude and distribution of absorbed electromagnetic energy within biological objects that are exposed to RF fields. At RF, the dosimetric quantity, which is called the specific absorption rate (SAR), is defined as the rate at which energy is absorbed per unit mass. The SAR is determined not only by the incident electromagnetic waves but also by the electrical and geometric characteristics of the irradiated subject and nearby objects. It is related to the internal electric field strength (E) as well as to the electric conductivity and the density of tissues; therefore, it is a suitable dosimetric parameter, even when a mechanism is determined to be “athermal.” SAR distributions are usually determined from measurements in human models, in animal tissues, or from calculations. This tutorial describes experimental techniques that are used commonly to determine SAR distributions along with the SAR limitations and unresolved problems. The methods discussed to obtain point, planar, or whole-body averaged SARs include the use of small E-field probes or measurement of initial rate of temperature rise in an irradiated object. © 1996 Wiley-Liss, Inc.  相似文献
