The growth characteristics of threeRumex species were determined under different solution oxygen concentrations in hydroculture. These species all occur in a river foreland ecosystem and they were found to differ in their flood tolerance. The flood-tolerantR. maritimus undR. crispus developed a large number of new, aerenchymatous roots within a short period under low solution oxygen concentrations. Biomass production was not affected. In the flood-intolerantR. thyrsiflorus, however, only few slow-growing new roots were developed and biomass production was significantly reduced at solution oxygen concentrations below 2% (v:v). These different responses could be partly explained by a differential aerenchyma formation in new roots of the flood-tolerant species. Aerenchyma can relieve the oxygen stress of the root systems via internal aeration.The fast development of new roots of the flood-tolerantR. maritimus andR. crispus after the onset of anaerobiosis coincided with the reduction or cessation of growth of the primary roots. Notwithstanding the cessation of growth, however, primary roots of both species were able to recover following restoration of aerobic conditions after a 13-day anaerobic period. However, the roots ofR. thyrsiflorus ceased growing very soon after the onset of anaerobiosis. All had died within 10 days.The balance between the growth rates of the primary and the newly formed root system are discussed and related to the differential tolerance of theRumex species to transient flooding.  相似文献

Ice algae are a case study in photo-autotrophic growth and metabolism under chronically low temperature and irradiance. We measured the allocation of 14C-labelled photosynthate among major classes of intracellular carbon (low molecular weight compounds, or LMW; lipid; protein; and polysaccharide) and found light-dependent changes in allocation broadly similar to photo-adaptive responses known in phytoplankton at higher temperatures; average relative allocation to protein varied inversely (10–37%) and allocation to lipids and polysaccharides directly (10–23%, and 16–21%, respectively) with the sub-inhibiting irradiance levels we employed (3.5–33.0 μE.M−2. s−1). Unlike many observations at higher temperatures, ours indicated (on average) a large and light-insensitive allocation to LMW (ca. 40%) and a greater light-sensitivity in lipid than in polysaccharide allocation. At the higher incubation irradiances, resembling in situ levels typical of areas with little (0–5 cm) snow cover, allocation to protein was often low (10–13%) compared to many observations of nutrient-sufficient or light-limited phytoplankton. Allocation to protein increased substantially (to ca. 40%) during a period of intensified tidal mixing, and assimilation numbers also attained a maximum at about the same time. Such dynamics show that the ice algae are not constrained to their often protein-poor allocation by the constantly low ambient temperature. Rather, they display marked shifts in metabolism consistent with major changes in light and inorganic nutrient supply, driven in part by the physical process of tidal mixing.  相似文献

Senolytic drugs are designed to selectively clear senescent cells (SnCs) that accumulate with injury or aging. In a mouse model of osteoarthritis (OA), senolysis yields a pro-regenerative response, but the therapeutic benefit is reduced in aged mice. Increased oxidative stress is a hallmark of advanced age. Therefore, here we investigate whether senolytic treatment differentially affects joint oxidative load in young and aged animals. We find that senolysis by a p53/MDM2 interaction inhibitor, UBX0101, reduces protein oxidative modification in the aged arthritic knee joint. Mass spectrometry coupled with protein interaction network analysis and biophysical stability prediction of extracted joint proteins revealed divergent responses to senolysis between young and aged animals, broadly suggesting that knee regeneration and cellular stress programs are contrarily poised to respond as a function of age. These opposing responses include differing signatures of protein-by-protein oxidative modification and abundance change, disparate quantitative trends in modified protein network centrality, and contrasting patterns of oxidation-induced folding free energy perturbation between young and old. We develop a composite sensitivity score to identify specific key proteins in the proteomes of aged osteoarthritic joints, thereby nominating prospective therapeutic targets to complement senolytics.  相似文献

Background

There is renewed vigour in efforts to eliminate neglected tropical diseases including sleeping sickness (human African trypanosomiasis or HAT), including attempts to develop more cost-effective methods of tsetse control. In the West Nile region of Uganda, newly designed insecticide-treated targets are being deployed over an area of ∼500 km2. The operational area covers villages where tsetse control has not been conducted previously. The effectiveness of the targets will depend, in part, on their acceptance by the local community.

Methodology/Principal Findings

We assessed knowledge, perceptions and acceptance of tsetse baits (traps, targets) in villages where they had or had not been used previously. We conducted sixteen focus group discussions with male and female participants in eight villages across Arua District. Discussions were audio recorded, translated and transcribed. We used thematic analysis to compare the views of both groups and identify salient themes.

Conclusions/Significance

Despite the villages being less than 10 km apart, community members perceived deployed baits very differently. Villagers who had never seen traps before expressed fear, anxiety and panic when they first encountered them. This was related to associations with witchcraft and “ghosts from the river” which are traditionally linked with physical or mental illness, death and misfortune. By contrast, villagers living in areas where traps had been used previously had positive attitudes towards them and were fully aware of their purpose and benefits. The latter group reported that they had similar negative perceptions when tsetse control interventions first started a decade ago. Our results suggest that despite their proximity, acceptance of traps varies markedly between villages and this is related to the duration of experience with tsetse control programs. The success of community-based interventions against tsetse will therefore depend on early engagements with communities and carefully designed sensitization campaigns that reach all communities, especially those living in areas new to such interventions.  相似文献

Homegardens on Amazonian Dark Earths, Non-anthropogenic Upland, and Floodplain Soils along the Brazilian Middle Madeira River Exhibit Diverging Agrobiodiversity. We test the hypothesis that the agrobiodiversity associated with homegardens on three different soils—upland Amazonian Dark Earths (ADE) and Oxisols (OX), and Fluvent Entisols (FL)—commonly found along the middle Madeira River in the municipality of Manicoré, Amazonas State, Brazil, is different due to the contrasting biotic, abiotic, and cultural settings specific to each of these soils. Using data from interviews with 63 farmers about food and utility species, we compare structural and floristic characteristics of homegarden agrobiodiversity. The density of individuals is higher on ADE than on the other soils (mean ± standard deviation: 715 ± 363 on ADE, 474 ± 283 on OX, 642 ± 399 on FL). ADE and OX have higher species richness (28.2 ± 5.6 on ADE, 25 ± 3.7on OX, 23.6 ± 5 on FL), while ADE and FL have a greater degree of domestication (2 ± 0.6 on ADE, 1.3 ± 0.5 on OX, 2.3 ± 0.6 on FL). ADE and OX have greater proportions of richness, density, and coverage composed of South American species, while FL has greater proportions of richness and density composed of Old World species. ADE has higher proportions of density and coverage of Mesoamerican species. Floristic composition is also different between soils: ADE occupies an intermediate position, composed of species associated with each of the other soil types and species that are most common on ADE. These differences in agrobiodiversity emerge through the interaction of human agency, plant responses, and the unique properties of soils in relation to socioeconomic and historical trajectories over time.  相似文献

The study of eye movements and oculomotor disorders has, for four decades, greatly benefitted from the application of control theoretic concepts. This paper is an example of a complementary approach based on the theory of nonlinear dynamical systems. Recently, a nonlinear dynamics model of the saccadic system was developed, comprising a symmetric piecewise-smooth system of six first-order autonomous ordinary differential equations. A preliminary numerical investigation of the model revealed that in addition to generating normal saccades, it could also simulate inaccurate saccades, and the oscillatory instability known as congenital nystagmus (CN). By varying the parameters of the model, several types of CN oscillations were produced, including jerk, bidirectional jerk and pendular nystagmus. The aim of this study was to investigate the bifurcations and attractors of the model, in order to obtain a classification of the simulated oculomotor behaviours. The application of standard stability analysis techniques, together with numerical work, revealed that the equations have a rich bifurcation structure. In addition to Hopf, homoclinic and saddlenode bifurcations organised by a Takens-Bogdanov point, the equations can undergo nonsmooth pitchfork bifurcations and nonsmooth gluing bifurcations. Evidence was also found for the existence of Hopf-initiated canards. The simulated jerk CN waveforms were found to correspond to a pair of post-canard symmetry-related limit cycles, which exist in regions of parameter space where the equations are a slow-fast system. The slow and fast phases of the simulated oscillations were attributed to the geometry of the corresponding slow manifold. The simulated bidirectional jerk and pendular waveforms were attributed to a symmetry invariant limit cycle produced by the gluing of the asymmetric cycles. In contrast to control models of the oculomotor system, the bifurcation analysis places clear restrictions on which kinds of behaviour are likely to be associated with each other in parameter space, enabling predictions to be made regarding the possible changes in the oscillation type that may be observed upon changing the model parameters. The analysis suggests that CN is one of a range of oculomotor disorders associated with a pathological saccadic braking signal, and that jerk and pendular nystagmus are the most probable oscillatory instabilities. Additionally, the transition from jerk CN to bidirectional jerk and pendular nystagmus observed experimentally when the gaze angle or attention level is changed is attributed to a gluing bifurcation. This suggests the possibility of manipulating the waveforms of subjects with jerk CN experimentally to produce waveforms with an extended foveation period, thereby improving visual resolution.  相似文献
