Seasonal patterns of allozyme variation are examined for 12 polymorphic enzyme loci in Drosophila melanogaster. The data derive from a total of 56 samples taken from a natural population in the Summer and Fall of 1978 and 1979. Samples were obtained at approximately five-day intervals and assayed for 6-phosphogluconate dehydrogenase (6Pgd), phosphoglucomutase (Pgm) and glucose-6-phosphate dehydrogenase (G6pd). The remaining nine enzymes were assayed in an average of eight samples per season. None of the loci exhibit regular seasonal cycles of gene-frequency change, although 6Pgd does show significant, but irregular, frequency oscillations. There is also little evidence for gene-frequency differences between years, although 6Pgd is again exceptional in showing significant frequency changes between years. In addition, genotypic frequency distributions are usually consistent with random mating expectations. With the notable exception of 6Pgd, the data give a strong impression of gene-frequency homogeneity within and among years, despite obvious seasonal changes in climate and in the distribution of breeding sites.  相似文献

1. Through the use of isoelectric focusing and peptide analysis, the hemoglobins of Przewalski's horse. Equus przewalskii and the domestic horse, E. caballus have been compared. 2. Przewalski's horses have two separate alpha-globin chain polymorphisms similar to domestic horses. Each hemoglobin phenotype could be accurately determined by isoelectric focusing. 3. Confirmation of the electrofocusing hemoglobin determinations was made by comparison to amino acid composition analyses of purified tryptic peptides and by analysis of the rare hemoglobins phenotypes observed in a family of Norwegian trotting horses. 4. Hemoglobin genotypes of fifteen Przewalski's horses were determined and inheritance of hemoglobin haplotypes has been observed.  相似文献

1. Nitrate reductase was purified 134-fold from Escherichia coli K12. The purification procedure involves the release by Triton X-100 of the enzyme from the cell envelope. i. The purified enzyme exists in aqueous solution either as a monomer (mol. wt. about 220 000) or as an associated form (probably a tetramer; mol.wt. about 880 000). 3. The purified enzyme has three subunits with apparent mol.wts. of 150 000, 67000 and 65000. An additional subunit of apparent mol.wt. 20000 is present in a haem-containing fraction that is also produced by the preparative procedure described. 4. None of the enzyme subunits is present in the cell envelope of cells grown in the absence of nitrate. 5. Reversible changes in the activity of nitrate reductase in vitro with FMNH2 as reductant can be induced under circumstances which are without effect on the reduced Benzyl Viologen-NO3-activity.  相似文献

The ability of cysts of the brine shrimp, Artemia salina, to incorporate 14CO2 into organic compounds soluble in cold-trichloroacetic acid was examined over a broad range of cellular water concentrations. Carbon dioxide was not incorporated by cysts containing less than about 0.3 g H2O/g dried cysts, the “critical hydration” for CO2-fixation. This relationship held whether the cysts were hydrated from the liquid or the vapor phase. The incorporation of radioactivity was shown to be due exclusively to metabolic activity in the cellular component of the cyst. Above the critical hydration, the amount of 14CO2 incorporated was a function of cyst water content, but the kinds of metabolites labelled with this precursor, and their relative proportions, were found to be similar in cysts of greatly different hydration. Almost all of the radioactivity was associated with amino acids, Krebs cycle intermediates and related acids, and pyrimidine nucleotides. The fact that the pathway involved with CO2-fixation, and subsequent metabolism of the fixation products are all initiated in cysts containing as little as 0.3 g H2O/g is particularly noteworthy since this hydration level is well within the range of the amounts of “bound water” described in the literature for a wide array of cells and tissues.  相似文献

The dynamical behavior of chromosomal segments undergoing strong selection was investigated in four replicate populations of Drosophila melanogaster. This was accomplished by following the joint behavior of allozyme markers at the loci phosphoglucomutase and esterase C, adjacent to the recessive lethal locus Glued, during and following the course of selection against Glued. The results show strong selection at other loci in the region of the marked segment. Examination of the joint dynamic of the two markers indicates that there must be more than one, and probably several, selected loci in the region under observation, with large epistatic effects. The mode of selection on the segment often results in excess heterozygosity at the markers, but does not appear to be constant in time. It is concluded that the density of selective effects in the region under study is substantial.  相似文献

Studies on the respiratory nitrate reductase (EC 1.7.99.4) from Escherichia coli K12 by electron-paramagnetic-resonance spectroscopy indicate that its molybdenum centre is comparable with that in other molybdenum-containing enzymes. Two Mo(V) signals may be observed; one shows interaction of Mo(V) with a proton exchangeable with the solvent and has: A (1H) 0.9-1.2mT; g1 = 1.999; g2=1.985; g3 = 1.964; gav. = 1.983. Molybdenum of both signal-giving species may be reduced with dithionite and reoxidized with nitrate.  相似文献

Growth-promoting action of simazine and other s-triazine herbicides was detected by the use of sorghum (Sorghum bicolor [L]. Moench) callus tissue and the chlorophyll retention test. Soil application of simazine [2-chloro-4, 6-bis(ethylamino)-s-triazine] at sublethal levels nearly doubled the growth-promoting action of sorghum root exudates. Treated plants yielded up to 26% more total protein than untreated plants. This indicated that the level of callus growth-promoting action in the root exudate of the plant has a positive effect on its final total protein yield and confirms a positive effect of simazine on total protein content in certain instances. The results may provide a new understanding of the mode of action of s-triazines applied at sublethal levels in increasing protein content and certain enzymic activities of treated plants. It is speculated that the growth-promoting action of these herbicides is hormonal in nature and most likely kinetin-like.  相似文献

To investigate the use of whole blood manganese (Mn) as an indicator of total body Mn, we measured Mn in whole blood and liver of rats fed purified diets containing adequate (45 micrograms Mn/g diet) or deficient (1 microgram Mn/g diet) Mn. The mean hepatic Mn concentration was significantly lower (P less than 0.001) in the Mn-deficient group compared to the control group, 0.36, microgram Mn/g and 1.73 micrograms Mn/g, respectively. Furthermore, whole blood Mn was significantly reduced (P less than 0.001) in the deficient group when compared to the control group, 4.0 ng Mn/ml and 8.6 ng Mn/ml, respectively. Hepatic Mn linearly regressed against whole blood Mn yielded a statistically significant (P less than 0.001) correlation coefficient of 0.775. These data suggest that whole blood Mn is a valid indicator of body Mn status and thus may be useful, in addition to the measurements of serum copper and zinc, for the diagnosis and prognosis of diseases in which the metabolism of trace elements is affected. In addition, this paper describes and delineates operational parameters for the measurement of whole blood Mn using the IL 551 atomic absorption spectrophotometer and the IL 555 B flameless atomizer.  相似文献

Tissue-binding specificity of the type-3 fimbriae of pathogenic enteric bacteria was determined using frozen sections of human kidney. A wild-type Klebsiella sp. strain and the recombinant strain Escherichia coli HB101(pFK12), both expressing type-3 fimbriae, as well as the purified type-3 fimbriae effectively bound to sites at or adjacent to tubular basement membranes, Bowman's capsule, arterial walls, and the interstitial connective tissue. Bacterial adherence to kidney was decreased after collagenase treatment of the tissue sections. Recombinant strains expressing type-3 fimbriae specifically adhered to type V collagen immobilized on glass slides, whereas other collagens, fibronectin or laminin did not support bacterial adherence. In accordance with these findings, specific binding of purified type-3 fimbriae to immobilized type V collagen was demonstrated. Specific adhesion to type V collagen was also seen with the recombinant strain HB101(pFK52/pDC17), which expresses the mrkD gene of the type-3 fimbrial gene cluster in association with the pap-encoded fimbrial filament of E. coli, showing that the observed binding was mediated by the minor lectin (MrkD) protein of the type-3 fimbrial filament. The interaction is highly dependent on the conformation of type V collagen molecules since type V collagen in solution did not react with the fimbriae. Specific binding to type V collagen was also exhibited by type-3 fimbriate strains of Yersinia and Salmonella, showing that the ability to use type V collagen as tissue target is widespread among enteric bacteria.  相似文献
