Rabbit polymorphonuclear leucocytes (PMN, neutrophils) obtained from peritoneal exudate were exposed in vitro for one-half or one hour to continuous wave or amplitude-modulated (20-Hz) 100-MHz RF radiation in a temperature-controlled coaxial exposure chamber at field strengths from 2.5 to 4.1 V/cm (SARs of 120 to 341 W/kg). RF exposure at 37 +/- 0.2 degrees C had no detectable effect on PMN viability or phagocytosis compared to sham-exposed cells simultaneously subjected to the same time-temperature regime. Temperature control studies indicated that at 37 degrees C no effect on PMN viability would be expected but phagocytosis would be reduced by approximately 6%/degrees C temperature increase. The absence of an effect of RF exposure suggests that there was minimal undetected intrasample heating and that phagocytosis was not affected by 100-MHz RF radiation under the conditions of this study. 相似文献

Turtles are key components of modern vertebrate faunas and their diversity and distributions are likely to be affected by anthropogenic climate change. However, there is limited baseline data on turtle taxonomic richness through time or assessment of their past responses to global environmental change. We used the extensive Triassic–Palaeogene (252–223 Ma) fossil record of terrestrial and freshwater turtles to investigate diversity patterns, finding substantial variation in richness through time and between continents. Globally, turtle richness was low from their Triassic origin until the Late Jurassic. There is strong evidence for high richness in the earliest Cretaceous of Europe, becoming especially high following the Cretaceous Thermal Maximum and declining in all continents by the end-Cretaceous. At the K–Pg boundary, South American richness levels changed little while North American richness increased, becoming very high during the earliest Palaeogene (Danian). Informative data are lacking elsewhere for this time period. However, the Selandian–Thanetian interval, approximately 5 myr after the K–Pg mass extinction, shows low turtle richness in Asia, Europe and South America, suggesting that the occurrence of exceptional turtle richness in the post-extinction Paleocene fauna of North America is not globally representative. Richness decreased over the Eocene–Oligocene boundary in North America but increased to its greatest known level for Europe, implying very different responses to dramatic climatic shifts. Time series regressions suggest number of formations sampled and palaeotemperature are the primary influencers of face-value richness counts, but additional factors not tested here may also be involved. 相似文献

Chronic food restriction in Sprague-Dawley rats has been shown to alter adipose glucose metabolism. In the present study, lean and obese male Zucker rats were food restricted from 5 weeks until either 10 or 26 weeks of age and adipocyte glucose metabolism was measured. Adipocytes from restricted-fed lean and obese Zucker rats converted more glucose to CO2 and fatty acids than those from their ad libitum-fed counterparts in both the absence and the presence of increasing doses of insulin at 10 weeks of age. At the highest insulin dose, adipocytes from restricted-fed obese rats converted significantly more glucose to CO2 and fatty acids than did those from restricted-fed lean rats. Basal glyceride-glycerol values were similar in all groups at this age. At the 0.4 and 2.0 ng/ml insulin levels, glyceride-glycerol production was highest in restricted-fed lean rats; restricted-fed obese and ad libitum-fed lean rats had similar values; and ad libitum-fed obese rats had the lowest. At the 20 ng/ml dose, glyceride-glycerol values of restricted-fed rats were higher than those of ad libitum-fed rats. Basal and insulin-stimulated values were compared within each group. Most basal versus insulin-stimulated values were significantly different for the two lean groups. For ad libitum-fed obese rats, only 0 versus 20 ng/ml insulin values were significant. Restricted-fed obese rats had significant increases in 0 versus both 2 and 20 ng/ml insulin values. Restricted-fed obese rats had significantly lower serum insulin levels relative to ad libitum-fed obese rats at 10 weeks of age. Adipocytes from all rats at 26 weeks of age had similar basal rates of conversion of glucose metabolism to all three metabolites. In the presence of insulin, adipocytes from restricted-fed lean rats metabolized significantly more glucose to CO2 and glyceride-glycerol than adipocytes prepared from the three other groups. Fatty acid production was similar in all groups at each insulin level. Only restricted-fed lean rats showed consistent significant responses to insulin stimulation for the three metabolites. ABSTRACTMarine lakes are landlocked bodies of seawater, isolated to varying degrees from the surrounding marine habitat. Isolated lakes generally have lower pH values, salinities and higher temperatures than more open lakes. We used a 16S rRNA gene barcoded pyrosequencing approach to study the bacterial communities of two sponge species, sediment and seawater in one enclosed and two open marine lakes. Bacterial communities of the sponge Spheciospongia solida mainly consisted of Proteobacteria, Cyanobacteria and Bacteroidetes. In contrast, Proteobacteria, Chloroflexi and Acidobacteria dominated the bacterial communities of the sponge Spongia ceylonensis. Although only a limited amount of samples were collected, both water and S. ceylonensis sponge had higher relative abundances of Cyanobacteria in the enclosed lake, which mainly consisted of OTUs assigned to the genus Synechococcus. This is in line with a number of previous studies, which have shown that environmental conditions found within low pH environments such as marine lakes benefit the growth of Synechococcus spp. Future studies should address the mechanism by which Synechococcus spp. may help host sponges and their bacterial communities adapt to low pH conditions in isolated marine lakes and other low-pH environments. 相似文献

Dutch rabbits were acutely exposed to electromagnetic pulsed (EMP) fields (pulse duration 0.4μs, field strengths of 1–2 kV/cm and pulse repetition rates in the range of 10 to 38 Hz) for periods of up to two hours. The dependent variables investigated were pentobarbital-induced sleeping time and serum chemistry (including serum triglycerides, creatine phosphokinase (CPK) isoenzymes, and sodium and potassium). Core temperature measured immediately pre-exposure and postexposure revealed no exposure-related alterations. Genomic imprinting is an epigenetic process that results in the preferential silencing of one of the two parental copies of a gene. Although the precise mechanisms by which genomic imprinting occurs are unknown, the tendency of imprinted genes to exist in chromosomal clusters suggests long-range regulation through shared regulatory elements. We characterize a 800-kb region on the distal end of mouse chromosome 7 that contains a cluster of four maternally expressed genes, H19, Mash2, Kvlqt1, and p57Kip2, as well as two paternally expressed genes, Igf2 and Ins2, and assess the expression and imprinting of Mash2, Kvlqt1, and p57Kip2 during development in embryonic and extraembryonic tissues. Unlike Igf2 and Ins2, which depend on H19 for their imprinting, Mash2, p57Kip2, and Kvlqt1 are unaffected by a deletion of the H19 gene region, suggesting that these more telomeric genes are not regulated by the mechanism that controls H19, Igf2, and Ins2. Mutations in human p57Kip2 have been implicated in Beckwith-Wiedemann syndrome, a disease that has also been associated with loss of imprinting of IGF2. We find, however, that a deletion of the gene has no effect on imprinting within the cluster. Surprisingly, the three maternally expressed genes are regulated very differently by DNA methylation; p57Kip2 is activated, Kvlqt1 is silenced, and Mash2 is unaffected in mice lacking DNA methyltransferase. We conclude that H19 is not a global regulator of imprinting on distal chromosome 7 and that the telomeric genes are imprinted by a separate mechanism(s). 相似文献
