Microglia are immunocompetent cells in the brain that have manysimilarities with macrophages of peripheral tissues. In normal adultbrain, microglial cells are in a resting state, but they becomeactivated during inflammation of the central nervous system, afterneuronal injury, and in several neurological diseases. Patch-clamp studies of microglial cells in cell culture and in tissue slices demonstrate that microglia express a wide variety of ion channels. Sixdifferent types of K+ channelshave been identified in microglia, namely, inward rectifier, delayedrectifier, HERG-like, G protein-activated, as well as voltage-dependentand voltage-independentCa2+-activatedK+ channels. Moreover, microgliaexpress H+ channels,Na+ channels, voltage-gatedCa2+ channels,Ca2+-release activatedCa2+ channels, andvoltage-dependent and voltage-independentCl channels. With respectto their kinetic and pharmacological properties, most microglial ionchannels closely resemble ion channels characterized in othermacrophage preparations. Expression patterns of ion channels inmicroglia depend on the functional state of the cells. Microglial ionchannels can be modulated by exposure to lipopolysaccharide or variouscytokines, by activation of protein kinase C or G proteins, by factorsreleased from astrocytes, by changes in the concentration of internalfree Ca2+, and by variations ofthe internal or external pH. There is evidence suggesting that ionchannels in microglia are involved in maintaining the membranepotential and are also involved in proliferation, ramification, and therespiratory burst. Understanding the evolutionary potential of morphological evolution is still a major problem in evolutionary biology. In this study, we tried to quantify the amount of variation of different traits among species of a Drosophila clade reared under standard conditions. Nineteen different traits have been measured on nine species of the same clade, the Neotropical saltans group of Drosophila. Measured traits can be distributed into five categories: size‐traits (wing and thorax), shape indices (ratios among the size traits), number of sternopleural bristles on the thorax, number of abdominal bristles on successive sternites, and dorsal pigmentation of abdomen. All species are of medium size with a generally dark pigmentation. A remarkable feature is the presence of numerous bristles on the 6th sternite of the males, while this segment is bare in other Drosophila species. A multivariate analysis revealed that it was possible to discriminate all the investigated species by using the complete set of measured traits. For each trait, phenotypic variability was investigated at the within‐ and between‐species levels. As a rule, the interspecific coefficient of variation (CV) was much greater than the within species CV, and it is proposed to call it realized evolvability. All possible correlations were calculated between traits across species, providing many unexpected results. Size traits were highly correlated among them, but not correlated with shape indices. Abdominal traits (bristles and pigmentation) were correlated, but often in opposite directions, with thoracic shape indices. Tergite pigmentation was negatively correlated with bristle number on sternite. For the moment, most of the correlations cannot be explained by developmental processes or parallel selective pressures. Nonetheless, mapping the evolution of the two characters on a molecular phylogeny of the studied species revealed two opposite phylogenetic trends for abdominal pigmentation and setation, respectively. Microtubules (MTs) are polymers of alpha and beta tubulin dimers that mediate many cellular functions, including the establishment and maintenance of cell shape. The dynamic properties of MTs may be influenced by tubulin isotype, posttranslational modifications of tubulin, and interaction with microtubule-associated proteins (MAPs). End-binding (EB) family proteins affect MT dynamics by stabilizing MTs, and are the only MAPs reported that bind MTs via a calponin-homology (CH) domain (J Biol Chem 278 (2003) 49721-49731; J Cell Biol 149 (2000) 761-766). Here, we describe a novel 27 kDa protein identified from an inner ear organ of Corti library. Structural homology modeling demonstrates a CH domain in this protein similar to EB proteins. Northern and Western blottings confirmed expression of this gene in other tissues, including brain, lung, and testis. In the organ of Corti, this protein localized throughout distinctively large and well-ordered MT bundles that support the elongated body of mechanically stiff pillar cells of the auditory sensory epithelium. When ectopically expressed in Cos-7 cells, this protein localized along cytoplasmic MTs, promoted MT bundling, and efficiently stabilized MTs against depolymerization in response to high concentration of nocodazole and cold temperature. We propose that this protein, designated CLAMP, is a novel MAP and represents a new member of the CH domain protein family. 相似文献

The Cyanidiales is a group of asexual, unicellular red algae, which thrive in acidic and high temperature conditions around hot springs. These unicellular taxa have a relatively simple morphology and are currently classified into three genera, Cyanidium, Cyanidioschyzon and Galdieria. Little is known, however, about the biodiversity of Cyanidiales, their population structure and their phylogenetic relationships. Here we used a taxonomically broadly sampled three-gene data set of plastid sequences to infer a robust phylogenetic framework for the Cyanidiales. The phylogenetic analyses support the existence of at least four distinct Cyanidiales lineages: the Galdieria spp. lineage (excluding Galdieria maxima), the Cyanidium caldarium lineage, a novel monophyletic lineage of mesophilic Cyanidium spp. and the Cyanidioschyzon merolae plus Galdieria maxima lineage. Our analyses do not support the notion of a mesophilic ancestry of the Cyanidiales and suggest that these algae were ancestrally thermo-acidotolerant. We also used environmental polymerase chain reaction (PCR) for the rbcL gene to sample Cyanidiales biodiversity at five ecologically distinct sites at Pisciarelli in the Phlegrean Fields in Italy. This analysis showed a high level of sequence divergence among Cyanidiales species and the partitioning of taxa based on environmental conditions. Our research revealed an unexpected level of genetic diversity among Cyanidiales that revises current thinking about the phylogeny and biodiversity of this group. We predict that future environmental PCR studies will significantly augment known biodiversity that we have discovered and demonstrate the Cyanidiales to be a species-rich branch of red algal evolution. 相似文献

Acid mine drainage (AMD) barrens result from destruction of vegetation within AMD flow paths. When exposed to air, soluble iron in AMD undergoes oxidation and hydrolysis to form ferric iron (oxyhydr)oxides which accumulate on soil surfaces. A restoration experiment was conducted at a 50‐year‐old AMD barrens created by discharge from an abandoned underground coal mine. The objective was to determine whether vegetation could be established by altering rather than removing surface layers of acidic precipitates at a site representative of other mining‐degraded areas. Three zones in the barrens were identified based on moisture content, pH (2.7–3.3), and thickness of precipitates (0–35 cm). Our hypothesis was that application of the same reclamation method to all zones would fail to sustain >70% vegetative cover in each zone after four growing seasons. The method consisted of applying 11 t/ha lime and 27 or 54 t/ha compost before rototilling (top 15 cm) and mulching with oat straw containing viable seeds for a nurse crop. Lime‐only plots were included for comparison, and all amended plots were sown with a mine reclamation seed mix. Oats, sown species, and indigenous species dominated cover in the first, second, and fourth growing seasons, respectively. In the fourth year following reclamation, compost‐amended plots had >70% cover and improved soil properties in all three zones, providing evidence to reject our hypothesis. Vegetative restoration of AMD barrens did not require removal of highly acidic precipitates, since they could be transformed at low‐cost into a medium that supports indigenous plants. 相似文献

Mezcal from Tamaulipas (México) is produced by spontaneous alcoholic fermentation using Agave spp. musts, which are rich in fructose. In this study eight Saccharomyces cerevisiae isolates obtained at the final stage of fermentation from a traditional mezcal winery were analysed in three semi-synthetic media. Medium M1 had a sugar content of 100 g l?1 and a glucose/fructose (G/F) of 9:1. Medium M2 had a sugar content of 100 g l?1 and a G/F of 1:9. Medium M3 had a sugar content of 200 g l?1 and a G/F of 1:1. In the three types of media tested, the highest ethanol yield was obtained from the glucophilic strain LCBG-3Y5, while strain LCBG-3Y8 was highly resistant to ethanol and the most fructophilic of the mezcal strains. Strain LCBG-3Y5 produced more glycerol (4.4 g l?1) and acetic acid (1 g l?1) in M2 than in M1 (1.7 and 0.5 g l?1, respectively), and the ethanol yields were higher for all strains in M1 except for LCBG-3Y5, -3Y8 and the Fermichamp strain. In medium M3, only the Fermichamp strain was able to fully consume the 100 g of fructose l?1 but left a residual 32 g of glucose l?1. Regarding the hexose transporters, a high number of amino acid polymorphisms were found in the Hxt1p sequences. Strain LCBG-3Y8 exhibited eight unique amino acid changes, followed by the Fermichamp strain with three changes. In Hxt3p, we observed nine amino acid polymorphisms unique for the Fermichamp strain and five unique changes for the mezcal strains. 相似文献

BackgroundPythium insidiosum is an oomycete classified in the kingdom Stramenopila. P. insidiosum hyphae are not able to initiate infection without the secretion of hydrolytic enzymes, which are considered an important factor in microbial virulence.AimsTo evaluate the extracellular enzymatic activity of 14 Brazilian P. insidiosum isolates and a standard strain (ATCC 58637) by the API-ZYM System screening method.MethodsZoospores were grown in RPMI 1640 broth, and 65 μL of the liquid phase were inoculated in each cupule of the API-ZYM strips.ResultsDifferences in the enzymatic activities were observed among the isolates, although phosphohydrolases and ester hydrolases were conspicuous among all isolates. β-glucosidase was also present in most of the isolates. Enzymatic activities of α-glucosidase and chymotrypsin were not observed, differing from a previous study involving Australian isolates and intracellular enzymes.ConclusionsThe discrepancy in the enzymatic profile observed among Brazilian P. insidiosum isolates reflects the phenotypic variations found in susceptibility tests. 相似文献
