DNA minor groove-binding compounds (polyamides) that target insect and vertebrate telomeric repeats with high specificity were synthesized. Base pair recognition of these polyamides is based on the presence of the heterocyclic amino acids pyrrole and imidazole. One compound (TH52B) interacts uniquely and with excellent specificity (K(d) = 0.12 nM) with two consecutive insect-type telomeric repeats (TTAGG). A related compound, TH59, displays high specificity (K(d) = 0.5 nM) for tandem vertebrate (TTAGGG) and insect telomeric repeats. The high affinity and specificity of these compounds were achieved by bidentate binding of two flexibly linked DNA-binding moieties. Epifluorescence microscopy studies show that fluorescent derivatives of TH52B and TH59 stain insect or vertebrate telomeres of chromosomes and nuclei sharply. Importantly, the telomere-specific polyamide signals of HeLa chromosomes co-localize with the immunofluorescence signals of the telomere-binding protein TRF1. Our results demonstrate that telomere-specific compounds allow rapid estimation of relative telomere length. The insect-specific compound TH52 was shown to be incorporated rapidly into growing Sf9 cells, underlining the potential of these compounds for telomere biology and possibly human medicine. 相似文献

Bdelloids inhabit many different environments. The entire taxon is an order belonging to the phylum Rotifera. In spite of its wide distribution, it has a very uniform morphology, suggesting that natural selection has had almost no effect on its morphological characteristics.This paper reviews the geographical and ecological distributions of bdelloids and their ability to tolerate different conditions is discussed. Two characteristics account for the wide distribution of bdelloids, parthenogenetic reproduction and their ability to withstand unfavorable conditions through anhydrobiosis. The former is an apomictic thelytoky which may ensure genetic homogeneity within clones. However, evidence which suggests variability among and within parthenogenetic clones is discussed. Some recent experimental evidence indicates that anhydrobiosis does not affect the life history of the surviving individuals. The effects of anhydrobiosis on bdelloid populations are considered. 相似文献

A 1,6-dichlorohexane-degrading strain of Acinetobacter sp. was isolated from activated sludge. The organism could grow with and quantitatively release halide from 1,6-dichlorohexane, 1,9-dichlorononane, 1-chloropentane, 1-chlorobutane, 1-bromopentane, ethylbromide, and 1-iodopropane. Crude extracts contained an inducible novel dehalogenase that liberated halide from the above compounds and also from 1,3-dichloropropane, 1,2-dibromoethane, and 2-bromoethanol. The latter two compounds were toxic suicide substrates for the organism at concentrations of 10 and 5 microM, respectively. Mutants resistant to 1,2-dibromoethane (3 mM) lacked dehalogenase activity and did not utilize haloalkanes for growth. Mutants resistant to both 1,2-dibromoethane (3 mM) and 2-bromoethanol (30 mM) could no longer oxidize or utilize alcohols and were capable of hydrolytic dehalogenation of 1,2-dibromoethane to ethylene glycol. 相似文献

Dermatan sulphate proteoglycans have been extracted from bovine lung with 2.0 M CaCl2 and isolated using CsCl density gradient centrifugation, DEAE ion-exchange chromatography, gel chromatography and preparative sodium dodecyl sulphate-polyacrylamide gel electrophoresis. Ultrastructurally these proteoglycans are specifically associated with collagen fibrils. Dermatan sulphate (Mr 15.10(3)-35.10(3), with a strong prevalence for the higher Mr) is link via an O-glycosidic bond to a protein core, which is rich in Asx, Glx and Leu. Of the total uronic acid, 91% is iduronic acid. A part of the glucuronic acid residues is located near the protein core and a large cluster of disaccharides is devoid of glucuronic acid residues. An inhibition enzyme immunoassay has been developed to quantitate the proteoglycan. A model for the interaction between dermatan sulphate proteoglycans and collagen fibrils is proposed. 相似文献

Periodate oxidation of terminalN-acetyl- andN-glycoloylneuraminic acid residues in the mucins from edible bird nest substance and pig submandibular gland, respectively, can be carried out under conditions which exclusively give rise to the formation of the C-7 analogues of these sialic acids. In contrast, the C-8 compounds can be obtained in a maximum yield of about 40%. Under identical conditions,N-glycoloylneuraminic acid is oxidized about 1.5 times faster than theN-acetylated derivative. After release of the sialic acids by acid hydrolysis, the characterization of the oxidation products was carried out by TLC, by GLC and GLC-MS of the corresponding pertrimethylsilyl derivatives, and by 500-MHz1H-NMR spectroscopy. We isolated two strains of an unnamed Fusobacterium species from human intestinal microflora, which stereospecifically transformed bile acid 3-sulfates into C-3-unsubstituted, ring A-unsaturated bile acids. Both 3 alpha- and 3 beta-sulfates of 5 beta-bile acids were metabolized to delta 3-5 beta-cholenoic acids; 3 beta-sulfates of 5 alpha-bile acids were converted into a mixture of delta 2-5 alpha-bile acids and 3 alpha-hydroxy-5 alpha-bile acids, whereas 3 alpha-sulfates of 5 alpha-bile acids were left intact. Unsulfated bile acids were not transformed into unsaturated derivatives. Earlier studies have indicated that interior (physical and/or chemical) properties of a plant may be responsible for feeding-site selection by the greenhouse whitefly, Trialeurodes vaporariorum (Westw.). In order to study the process of feeding-site selection further, stylet-penetration activities and the pathway followed by the stylets in host-plant tissue were investigated using a DC electrical recording method and transmission electron microscopy (TEM). Penetrating whiteflies attached to a gold wire were included in an electrical circuit to record electrical penetration graphs (EPGs). Seven EPG patterns have been distinguished, five of which could be correlated with components of the stylet-penetration process: 1) one with penetration of the leaf surface, 2) one with intercellular penetration and salivary-sheath secretion, 3) one with sieve element penetration and ingestion, 4) one with short penetration of a cell, and 5) one with xylem penetration. The stylet pathway is almost completely intercellular before the phloem is reached and in contrast to aphids, brief symplast punctures are very rare. In general, it takes T. vaporariorum more than half an hour from the start of a penetration to reach a sieve element. Rejection of feeding sites occurs within a few minutes of penetration by adult whiteflies, a time span in which stylets are presumed to penetrate just beyond the epidermis. Properties of the apoplast close to the leaf surface seem therefore to play a major role in feeding-site selection.
Résumé De précédentes études ont montré que les propriétés internes (physiques et/ou chimiques) d'une plante peuvent induire la sélection du site de nutrition de la mouche blanche de serres, Trialeurodes vaporariorum (Westw.). Afin d'étudier plus avant le processus de sélection du site de nutrition, les activités de piqûre et le chemin suivi par le stylet dans les tissus de la plante-hôte ont été étudiés par une méthode d'enregistrement électrique en courant continu ainsi que par microscopie à transmission d'électrons (TEM). Des aleurodes en activité de piqûre attachées à un fil d'or ont été incluses dans un circuit électrique pour enregistrer des graphes de pénétration électriques (EPG). Sept motifs d'EPG ont été distingués, dont cinq peuvent être corrélés aux composantes du processus de pénétration du stylet: 1) un avec pénétration de la surface foliaire, 2) un avec pénétration interecellulaire et sécrétion d'une gaine de salive, 3) un avec pénétration du phloème, 4) un avec courte pénétration d'une cellule, et 5) un avec pénétration du xylème. Le parcours du stylet est presque entièrement intercellulaire avant que le phloème soit atteint. Contrairement aux pucerons, les ponctions brèves dans le symplasme sont rares. Généralement, T. vaporariorum met plus d'une demi-heure, à partir du début d'une piqûre, pour atteindre un vaisseau. Le rejet des sites de nutrition par les aleurodes adultes se passe quelques minutes après la pénétration du stylet; pendant ce laps de temps, le stylet pénétrerait juste sous l'épiderme. A strain of Pseudomonas putida was isolated that was able to degrade 2-chloroethanol. The degradation proceeded via 2-chloroacetaldehyde and chloroacetate to glycolate. In crude extracts the enzymes for this degradation pathway could be detected. All enzymes proved to be inducible. The dehalogenase that catalyzed the dehalogenation of chloroacetate to glycolate was further characterized. It consisted of a single polypeptide chain with a molecular mass of 28 kDa. After induction the dehalogenase was expressed at a high level. In a mutant resistant to high concentrations of 2-chloroethanol the dehalogenase was no longer expressed. The mechanism of resistance seemed to be due to the inability to convert chloroacetate and export of this compound out of the cell.Non-standard abbreviations CEO

We describe a 1132 bp sequence of the cyanelle genome of Cyanophora paradoxa containing the rpl3 gene. This gene, which is not chloroplast encoded in plants, is the first of a long cyanelle ribosomal operon whose organization resembles that of the S10 operon of E. coli. We have shown that the rpl3 gene is transcribed in cyanelles as a 7500 nucleotide precursor and that the 5'-end of the mRNA starts approximately 90 nucleotides upstream from the initiation codon. However, no typical procaryotic promoter could be found for this gene. We have detected, using anti E. coli L3 antibodies, the cyanelle L3 protein in cyanelle extracts and in E. coli cells transformed with the cyanelle rpl3 gene. 相似文献
