Summary When the bI4 RNA maturase, encoded by the fourth intron of the mitochondrial cytochrome b gene of Saccharomyces cerevisiae, was expressed in Escherichia coli, formation of intra-chromosomal Lac+ recombinants was stimulated threefold. This hyper-rec phenotype was recA as well as recBCD dependent. The most active form of the bI4 maturase stimulated homologous recombination whereas splicing deficient mutants of bI4 maturase were either deficient in or unable to stimulate homologous recombination.  相似文献

Lymphocytes from rodents cultured on syngeneic fibroblasts become cytotoxic against syngeneic but not against allogeneic target cells. We investigated whether known antigens are involved in the phenomenon and the data indicate that H-2 antigens must be shared between sensitizing fibroblasts and responder lymphocytes to generate autocytotoxic cells. Furthermore, the cytotoxicity of autosensitized lymphocytes is restricted to target cells identical with respect to theK and/orI regions. F1 hybrid lymphocytes cultured on parental fibroblasts develop cytotoxicity towards sensitizing cells. In contrast, parental lymphocytes cultured on F1 hybrid fibroblasts will not damage the F1 cells, although they are cytotoxic against both syngeneic and allogeneic parental cells. In addition, parental or F1 hybrid lymphocytes cultured on parental fibroblasts are not cytotoxic against F1 hybrid target cells. Fibroblasts heterozygous for theK end only, are also resistant to the cytotoxic action of such lymphocytes. Thus it seems that H-2 antigens, specifically theK end, antigens have a significant role in the phenomenon of autosensitization.  相似文献

Among a number of mutants unable to utilize non-fermentable carbon substrates, scoring for membrane ATPase and for ATP-driven transhydrogenase activity permitted to distinguish two phenotypes: (A) mutants lacking ATPase and ATPdriven transhydrogenase; (B) one mutant with an ATPase which behaved according to several criteria as released into solution instead of being membrane bound, a.o it exhibited no ATP-driven transhydrogenase activity. All A and B mutants exhibited a common nutritional pattern.The ATPase-deficient group, when scored for ATPase-binding sites on its membrane particles revealed three different subgroups: (1) mutants having free ATPase-binding sites, (2) mutants with ATPase-binding sites made available by the procedure which releases ATPase from wild-type membrane, and (3) mutants with no detectable ATPase-binding sites.Membranes of the mutant B with unbound ATPase also exhibited a deficiency in ATPase-binding sites, but its soluble ATPase was also found unable to bind to ATPase-binding sites of wild type membranes.The double alteration, namely abnormal or inactive ATPase and absence of ATPase-binding sites on the membrane is compatible with a single mutational defect.  相似文献

The synthesis of DNA does not proceed in a perfectly synchronous way in about 20% of the binucleate cells induced in Allium sativum L. root meristems by treatment with methyl 3 hydroxy 6 (1 H, 3 H) quinazoline dione, 2.4 (an inhibitor of cytokinesis). — Nuclear labelling after tritiated thymidine incorporation and the measurement of nuclear DNA by cytophotometry both establish that the beginning of the S phase as well as its duration may be different in the two nuclei included in a single cytoplasm. Yet the nuclear synchronism progressively asserts itself as the cell cycle goes on and, following this asynchronous synthesis, the beginning of mitosis is synchronous. The restoration of such a coordination points to the existence in the cytoplasm of separable factors inducing DNA synthesis and mitosis.  相似文献

The binding of [6,7-(3)H]oestradiol-17beta to subcellular fractions of the hypothalamus and the cerebellum of the rabbit was studied in vitro. Uptake of steroid was higher in hypothalamic nuclei than in cerebellar nuclei. Lower binding was observed in other fractions of both tissues. After dialysis of the fractions, hypothalamic nuclei retained a high percentage of oestradiol whereas cerebellar nuclei lost most of the bound steroid. Supernatant fractions of both tissues retained a significant proportion of label after dialysis and after gel filtration on Sephadex G-200. No specific binding was observed in these fractions when subjected to sucrose-density-gradient centrifugation. Purification of nuclei followed by incubation with labelled oestradiol in the absence of the supernatant fraction resulted in loss of binding of steroid by hypothalamic nuclei. Incubation of the purified hypothalamic nuclei with supernatant fraction maintained the binding specificity of hormone retention.  相似文献

Summary Mutants of Escherichia coli K12 carrying exc mutations inducing the release of the plasmid pBR322-encoded -lactamase (EC 3.5.2.6) into the extracellular medium were analysed and compared with previously described excretory mutants carrying lky mutations associated with the release of alkaline phosphatase and to tolA and tolB mutants, originally described as tolerant towards various colicins. The exc, lky and tol mutations mapped near the gal operon at min 16.5 of the E. coli linkage map. A genetic analysis presented in this paper showed that some exc and lky mutations belonged to the tolA and tolB complementation groups. Furthermore, we identified a third cistron, excC, involved in the excretion of periplasmic enzymes but distinct from the two others.  相似文献

Summary The structural modifications in the middle intestine of the trout, Salmo gairdneri, induced by transfer to seawater have been studied. During the first two days in seawater, significant distensions of the intercellular spaces are observed between the apical tight junctions and the basement membrane. These dilations are more frequent in the apical part of the intestinal folds. At the basal part of the cell, numerous lamellar processes open in the intercellular spaces. They are closely associated with elongated mitochondria, and are often mixed with small clear vesicles. After seven days in seawater, intercellular spaces are less expanded. Numerous mitochondria are observed in the apical part of the cell, and numerous myelinic bodies with dense granules lie near the nucleus. After one month in seawater, the epithelium resembles that of the freshwater controls; mitochondria are more numerous and other organelles are well developed. The most important modifications of the ultrastructure of the intestine mucosa occur during the first two days in seawater, in correlation with important physiological changes following the abrupt increase of environmental salinity.  相似文献
