STUDIES OF THE TRIAD : II. Penetration of Tracers into the Junctional Gap

Ferritin and Imferon molecules were introduced as tracers inside "skinned" muscle fibers to test which part of the triadic junction gap is freely exchangeable with the sarcoplasm. At least 50% of the T-system surface is freely accessible from the sarcoplasm. Of the remainder, 30% of the total T-system surface is covered by the junctional feet, and 20% in the center of the junction may or may not be accessible. The possibility is discussed that the triadic junction may not function as an electrical coupling.     

The effect of the plating medium on the recovery of nonsense suppressors in Saccharomyces cerevisiae

The recovery of nonsense suppressors in a strain of Saccharomyces cerevisiae carrying five ochre mutations, tr, hi, ly, ar, and ad, is affected by the plating medium. The highest frequency is observed on tryptophanless medium, while the lowest is observed on adenineless medium. Experiments showed that exogenous histidine inhibits suppressor expression and that exogenous adenine relieves this inhibition. In histidine-independent strains, mutation expression requires adenine. A model, based on the role of RNA in supersuppression and on the biosynthetic pathways of histidine and adenine, is proposed to account for the observed data. It cannot, however, account for the high frequency of suppressors on tryptophanless medium. The tentative conclusion is drawn either that mis-reading of the tryptophan nonsense codon by mutated tRNA is facilitated by the neighboring bases or that the type of acceptable amino acid is less rigorously limited in the mutated site of the tryptophan locus than in those of the other suppressible loci.This work was in part supported by the Calouste Gulbenkian Foundation and by the Medical Research Council (Grant No. G969/24/B).     

Lung volumes, chest wall configuration, and pattern of breathing in microgravity

We studied the changes in functional residual capacity (FRC), thoracoabdominal volume (Vw), and chest wall configuration in five normal subjects seated in an aircraft flying parabolic trajectories resulting in 20-s periods of microgravity. We measured vital capacity (VC), inspiratory capacity, and tidal volume by integrating airflow at the mouth and changes in rib cage and abdominal volume (delta Vrc and delta Vab, respectively, where delta Vrc + delta Vab = delta Vw) using induction plethysmography. During microgravity (0 Gz) FRC decreased by 413 +/- 70 (SE) ml and VC by 0.37 liter. The decrease in Vw did not differ from that in FRC and was entirely the result of reduction of Vab, the Vrc showing no significant change. During tidal breathing the abdominal contribution (delta Vab/delta Vw) increased from 0.39 +/- 0.08 at 1 Gz to 0.57 +/- 0.08 at 0 Gz. During brief periods of hypergravity (approximately 1.8 Gz) all changes were opposite in sign and relatively smaller. Limited data during "roller coaster" flight patterns suggested that, in contrast to configurational changes, the temporal pattern of breathing was uninfluenced by changes in Gz. We conclude that at the onset of weightlessness there are substantial changes in lung volume and thoracoabdominal configuration. Abdominal contribution to tidal excursions increases but the temporal pattern of breathing is unchanged.     

Determination of trace elements in aerosol samples by Instrumental Neutron Activation Analysis

Two nuclear techniques, Energy-Dispersive X-Ray Fluorescence Analysis (EDXRF) and Instrumental Neutron Activation Analysis (INAA), were used to analyze aerosol samples collected in the city of São Paulo, Brazil. Na, Cl, Mn, V, Al, Sm, Mo, W, La, As, Br, Sb, K, Ba, Se, Th, Cr, Rb, Ca, Fe, Ce, and Sc were determined by INAA, and Al, Si, P, S, Cl, K, Ca, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, Ga, As, Se, Br, Rb, Sr, Hg, and Pb were determined by EDXRF. A preliminary identification of the main source of the atmospheric aerosol was performed based on enrichment factor and correlation coefficient calculations.

     

Antagonism between growth and flocculation inPichia stipitis NRRL Y-7124:Influence of Ca +2 and Mg +2 ions

Summary The effects of Mg⁺² and Ca⁺² ion concentrations onPichia stipitis growth and flocculation were studied by a 2×2 factorial design. Negative interaction was detected between these ions (p<0.01): maximum growth rate was attained with high Mg⁺² (3 mM) and low Ca⁺² (0.34 mM) concentration levels, whereas the inverse ionic ratio was required to reach the maximum flocculation extent. Neither stimulatory or inhibitory effects on growth or flocculation were observed by increasing other cations (Zn⁺², Mn⁺², Co⁺², Fe⁺²) or vitamins concentration. An antagonistic relationship between growth and flocculation was concluded.     

Reduced lignin in transgenic plants containing a caffeic acidO-methyltransferase antisense gene

Lignin is a major structural polymer of secondarily thickended plant vascular tissue and fibres, imparting mechanical strength to stems and trunks and hydrophobicity to conducting vessels. Constitutive expression of a lucerne caffeic acid 3-O-methyltransferase antisense RNA in transgenic tobacco leads to a significant reduction in lignin content, particularly in the younger parts of the stems, without apparent alterations in lignin monomer composition. These observations open up the possibility of genetically manipulating plants with reduced lignin for improved processing and biomass digestibility.
