GPR84 is a recently de-orphanized member of the G-protein coupled receptor (GPCR) family recognizing medium chain fatty acids, and has been suggested to play important roles in inflammation. Due to the lack of potent and selective GPR84 ligands, the basic knowledge related to GPR84 functions is very limited. In this study, we have characterized the GPR84 activation profile and regulation mechanism in human phagocytes, using two recently developed small molecules that specifically target GPR84 agonistically (ZQ16) and antagonistically (GLPG1205), respectively. Compared to our earlier characterization of the short chain fatty acid receptor FFA2R which is functionally expressed in neutrophils but not in monocytes, GPR84 is expressed in both cell types and in monocyte-derived macrophages. In neutrophils, the GPR84 agonist had an activation profile very similar to that of FFA2R. The GPR84-mediated superoxide release was low in naïve cells, but the response could be significantly primed by TNFα and by the actin cytoskeleton disrupting agent Latrunculin A. Similar to that of FFA2R, a desensitization mechanism bypassing the actin cytoskeleton was utilized by GPR84. All ZQ16-mediated cellular responses were sensitive to GLPG1205, confirming the GPR84-dependency. Finally, our data of in vivo transmigrated tissue neutrophils indicate that both GPR84 and FFA2R are involved in neutrophil recruitment processes in vivo.In summary, we show functional similarities but also some important differences between GPR84 and FFA2R in human phagocytes, thus providing some mechanistic insights into GPR84 regulation in blood neutrophils and cells recruited to an aseptic inflammatory site in vivo. 相似文献

Sjögren-Larsson syndrome (SLS) is an autosomal recessive disorder characterized by congenital ichthyosis, spastic di- or tetraplegia, and mental retardation. SLS has been reported to occur in many populations but the highest incidence is in the north of Sweden. The gene causing SLS encodes a fatty aldehyde dehydrogenase (FALDH). In the present study, a point mutation in exon 7 of the FALDH gene was found in SLS patients of northern Swedish origin. The mutation consists of a C-to-T exchange at nucleotide position 943 in the cDNA. As a consequence, a highly conserved proline is replaced by a serine. The mutation was found in 49 out of 58 affected chromosomes and could be the most widely spread SLS mutation in the world. 相似文献

In this study, the three-dimensional stabilizing capabilities of the AO-Internal Fixator (IF) and the new Universal Spine System (USS) were investigated. Both devices were tested without and with the cross-link system (IF, IFC, USS, USSC). To determine biomechanical characteristics, a human thoracolumbar spine instability model with resection of the vertebral body Th12 was created. The vertebral body was replaced by a spacer and transpedicular posterior stabilization was performed from Th11 to L1. All devices reduced the range of motion (ROM) significantly compared to the values of the intact specimen. In flexion the IFC showed the highest reduction of ROM (85% of intact), followed by the USSC, USS and IF (79% of intact). In extension the ROM was restored again most by the IFC (52% of intact), followed by the USSC, IF and USS (44% of intact). In lateral bending stability was provided by the USSC (right 78% and left 81% of intact), followed in right lateral bending by the IF, IFC and USS and in left lateral bending by the USS, IF and IFC. In axial rotation the ROM was reduced primary by the IFC (right 51% and left 46% of intact), followed in right axial rotation by the USS, USSC and IF, in left axial rotation by the USSC, USS and IF. Additional stability by crosslinking has been provided in the IF and the USS in flexion and extension, in the USS in lateral bending and in the IF in axial rotation nonsignificantly. The neutral zone (NZ) was reduced by posterior instrumentation in flexion/extension and right/left lateral bending significantly. In axial rotation only the USSC decreased the NZ below intact levels. The study showed no statistical significant differences in the stabilizing capabilities of the USS compared to the IF. For both implants the cross-link system increased stability in the chosen instability model insignificantly only. 相似文献

Loading conditions physiologically approximating those acting on the normal masticatory system were incorporated into a new mandibular load simulator. Separate tension wires attached to each ramus of the mandible simulated the resultant force vectors of the masticatory musculature. The muscle insertion points were chosen in accordance with the anatomical situation, and the maximum in vivo forces acting on the joint. In a first application, the stability of a 2.4 mm LC-WDCP was compared with that of a 2.7 mm EDCP in plastic mandible models. It was found that under largely physiological loading, the 2.4 mm LC-EDCP exerted a stabilizing effect similar to that of a 2.7 mm EDCP. Although of smaller dimensions, the 2.4 mm LC-EDCP appears to enable an osteosynthesis of similar stability in the treatment of fractures of the mandibular angle. 相似文献

The mycosporine-like amino acid (MAA), porphyra-334 (lambda(max) = 334 nm; epsilon = 42,300 M(-1) cm(-1)), was isolated from the aquatic cyanobacterium Aphanizomenon flos-aquae (AFA) and its structure was verified by spectroscopic methods. The UVA absorption properties of the crude methanolic extract were determined against two commercial sun care products in terms of mean critical wavelength, mean UVA/UVB ratios and UVA protection category (Boots the Chemists, Ltd.). Chlorite-oxidized oxypolysaccharides are polyacetal carboxylic acids. They inhibited the cytopathic effect of vesicular stomatitis virus in mouse embryo cell cultures challenged at low input multiplicity. After intraperitoneal injection of these compounds in mice, interferon appeared in the circulation. The compounds also protected mice against lethal mengovirus infection and against the development of experimental pox lesions on the tail. Chlorite-oxidized oxyamylose was antiviral only when at least 64% of the glucopyranose units were oxidized, an observation which suggested a correlation between charge density and antiviral effect. The antiviral activity was also influenced by the molecular weight, as demonstrated by the fact that chlorite-oxidized dextrans which had a high intrinsic viscosity were more active than those with low intrinsic viscosity. 相似文献

The importance of inorganic ions for the heat stability of thermophilic bacteria was investigated. Cells of Bacillus stearothermophilus, strain 1503, were incubated at elevated temperatures in various media and the number of surviving organisms was determined at suitable intervals. The bacteria rapidly died at temperatures ordinarily employed for their cultivation if the surrounding medium lacked calcium ions. Besides calcium ions, potassium and phosphate ions and glucose, or some other energy source, seemed to be required for the heat stability of the cells. A chemically defined stabilizing medium with these components was developed for the above-mentioned strain. When any component of this medium was excluded, the heat resistance of this organism was lost. This medium had a stabilizing effect also on the cells of three other strains of B. stearothermophilus. These requirements suggest that the heat stability of thermophilic bacteria is attributable to an active transport of calcium ions from the environment into the cells. 相似文献
