The histones of the endosymbiont nucleus of the binucleate dinoflagellate Peridinium balticum were characterized by amino acid analysis and peptide mapping, and compared to calf thymus histones. Using these and various other criteria we have identified two H1-like histones as well as the highly conserved histones H3 and H4. A 13,000 dalton component in sodium dodecyl sulphate (SDS) gels can be separated into two components in Triton-containing gels. We suggest that these histones (HPb1 and HPb2) correspond to the vertebrate histones H2A and H2B, respectively. 相似文献

The case of a young man with the plasma cell type of Castleman's disease is described. Beyond the well known systemic manifestations the coagulation tests showed a decrease in the activity of factors V, VIII and X due to the presence of lupus-like circulating anticoagulants. After surgical excision of the mediastinal mass both hematochemical pathological data and the coagulation defect disappeared. It is suggested that the lupus-like anticoagulant was secreted by the tumor. 相似文献

The histone-like protein from Crypthecodinium cohnii (HCc) was examined in regard to its amino acid composition and the peptide pattern resulting from protease digestion. A revised amino acid composition indicated a higher lysine and arginine content and a lower glycine content than that determined previously. Comparative peptide mapping of HCc with HTa, a histone-like protein from Thermoplasma acidophilum, and with a histone-like protein from the dinoflagellate Gyrodinium dorsum showed significant differences in the peptide patterns produced. 相似文献

The sex-linked mutation fs(1)42 was induced by ethyl methane sulfonate. It has no effect on either the external morphology or longevity of adult hemizygotes or homozygotes. Heterozygotes and hemizygotes are fertile, but homozygotes are sterile. Egg chamber development proceeds through stages 8, and thereafter chambers degenerate. Dying follicle cells are seen in chambers at all positions in the ovarioles. Profollicle cells also die within germaria, and clusters of sister cystocytes take longer than normal to receive their coverings of follicle cells. Egg chambers in the vitellarium contain only about 60% the normal number of follicle cells, these generally have greater lateral dimensions, and their nuclei and nucleoli are also larger than normal. The follicular envelope of mutant chambers often contains gaps through which cystocytes send cytoplasmic projections. Abnormalities seen in development of the fs(1)42 oocyte are likely to be due to its envelope of defective follicle cells. 相似文献

The results of a combined experimental and theoretical study of the electronic circular dichroism spectrum of (?)–[9] (2,5)Pyridinophane are presented. Progressive angiogenesis and endothelial differentiation in the chick chorioallantoic membrane (CAM) serve to accommodate oxygen demands of the growing embryo. The present study evaluated CAM microvascular endothelial permselectivity during the most rapid phase of angiogenesis (day 10) and after initiation of endothelial cytodifferentiation (day 14). Chick embryos were incubated using established shell-less culture techniques tor intravital and ultrastructural observations. Systemic microinjections of FITC-dextrans (40, 70 and 150 KDa) provided an index of endothelial permselectivity after 2.5 min and 10 min perfusions. Ultrastructural examinations of the same dextran probes served to detect small intermittent foci within the perivascular interstitium. Although minor variations of dextran particle distributions around specific segments of the microcirculation were observed ultrastructurally, perivascular accumulation was not sufficient to elicit a detectable fluorescent signal. Thus, substantial accumulation of the graded-dextran series in the perivascular intcrstitium was not detected. Morphometric analyses of the precapillary, capillary, and postcapillary microvascular segments served to demonstrate a continuous endothelium which displayed cytoplasmic attenuation at day 14. Plasmalemmal vesicles were few and uniform within the microvascular units at day 10. A three-fold increase in vesicle densities characterized the precapillary endothelia at day 14. Average widths of the endothelial junctional clefts were homogeneous within the segmental microvascular endothelia at both days 10 and 14. Junctional cleft lengths were also homogeneous, except the significantly longer capillary endothelial clefts observed at day 10. These results are consistent with the concept that, despite certain differences in segmental vesicle densities and junctional cleft lengths, neovascularization of the CAM is achieved without excessive macromolecular efflux across the microvascular endothelia. 相似文献

The circular dichroic properties of H-Gly-Phe-(Gly)n-Trp-Gly-OH (II, n = 0,1,2) and of related simpler peptides, such as H-Phe-Gly-OH, H-Gly-Phe-OH, H-Gly-Phe-Gly-OH, H-Phe-Trp-OH, H-Phe-Trp-Gly-OH, and H-Gly-Phe-Trp-OH in water and trifluoroethanol solutions are investigated. Peptides containing only one phenylalanyl residue show markedly different near-uv dichroic signals depending on whether this residue is in the N-terminal position or not. The possible origin of this feature is discussed. The study of the oligopeptides II (n = 0,1,2) shows that no strong intramolecular interaction between the two aromatic rings is present. However, the dichroic properties of II (n = 0) are clearly anomalous, and the analysis of H-Gly-Phe-Trp-OH, H-Phe-Trp-Gly-OH, and of H-Phe-Trp-OH at different pH's, confirms that the presence of two adjacent aromatic residues may bring about chiroptical properties which indicate a restriction in the conformational equilibrium of the molecule. The limits, and the possible generalization of this finding, are discussed. 相似文献
