Recent data suggest that oxygen free radicals are implicated in the pathogenesis of ischemic injury. This study evaluates the effects of allopurinol, a xanthine oxidase (XO) inhibitor, on malonaldehyde generation, free sulfhydryl levels, oxygen consumption, and water contents of rat gastrocnemius muscles of female Sprague-Dawley rats subjected to tourniquet shock and after hind-limb reperfusion. Serum lactic dehydrogenase isozyme patterns after ligature release were also examined. Our results show that the four muscle parameters were not altered during 5 hr of ischemia, but that on hind-limb reperfusion, malonaldehyde production, SH levels, O2 consumption, and water contents were significantly altered in the control animals, but not in those pretreated with allopurinol. LDH serum patterns of the untreated animals showed the presence of all five isoforms; these were much less evident in the drug-protected rats. Our data suggest that following ischemia, the affected muscles are unable to recover their normal function when reperfusion is resumed. The subsequent damage is probably due to the generation of cytotoxic superoxide radicals formed during the XO-catalyzed transformation of hypoxanthine to uric acid on tissue reoxygenation. The severity of tissue damage is related to the duration of the ischemic episode possibly due to hypoxanthine accumulation during ischemia. 相似文献

In this work we establish the chromosomal composition of a wild-type, one astaxanthin and two -carotene overproducer strains of the red yeast Phaffia rhodozyma. The method used has been pulsed field gel electrophoresis, which has determined 9 DNA chromosomal bands in the yeast genome. The two largest bands are triplets and two other bands, VI and VIII, seem to be doublets. The size of the chromosomal bands varies between 0.35 and 2.5 Mb, suggesting a genome size of 25 Mb. The technique used, complemented with hybridization assays using specific DNA probes, provides direct information about the genomic organization of P. rhodozyma. We have also cloned and located in chromosomal bands different DNA sequences that code for the translation elongation factor 1 alpha (ef-1), a 7.6 kb BamHI fragment of repetitive DNA (possibly rDNA) and a randomly chosen fragment (named locus R2). Additionally, we have detected a chromosomal length polymorphism between wild-type strains and mutant strains affecting carotenogenesis obtained in our laboratory. 相似文献

Molecular phylogenetics at the population/species interface in cave spiders of the southern Appalachians (Araneae:Nesticidae:Nesticus) 总被引:5,自引:0,他引:5
This paper focuses on the relationship between population genetic structure
and speciation mechanisms in a monophyletic species group of Appalachian
cave spiders (Nesticus). Using mtDNA sequence data gathered from 256
individuals, I analyzed patterns of genetic variation within and between
populations for three pairs of closely related sister species. Each
sister-pair comparison involves taxa with differing distributional and
ecological attributes; if these ecological attributes are reflected in
basic demographic differences, then speciation might proceed differently
across these sister taxa comparisons. Both frequency-based and gene tree
analyses reveal that the genetic structure of the Nesticus species studied
is characterized by similar and essentially complete population
subdivision, regardless of differences in general ecology. These findings
contrast with results of prior genetic studies of cave-dwelling arthropods
that have typically revealed variation in population structure
corresponding to differences in general ecology. Species fragmentation
through both extrinsic and intrinsic evolutionary forces has resulted in
discrete, perhaps independent, populations within morphologically defined
species. Large sequence divergence values observed between populations
suggest that this independence may extend well into the past. These
patterns of mtDNA genealogical structure and divergence imply that species
as morphological lineages are currently more inclusive than basal
evolutionary or phylogenetic units, a suggestion that has important
implications for the study of speciation mechanisms.

This study compares the segregation of the ABO and Rh systems between malformed newborns and a control group with two purposes: (1) to evaluate the participation of genetic factors associated with these blood groups in the production of congenital malformations, and (2) to prove, indirectly, the existence of reproductive losses associated with congenital malformations. The newborns and their mothers were typed for ABO and Rh groups. Gene frequencies were similar in malformed and control newborns. In the female malformed newborns, an excess of O-B pairs and a deficit of B-B pairs in the ABO system, and an excess of Rh(-)-Rh- pairs in the Rh system were found. 相似文献

Clinical and experimental studies have shown that long-term carnitine deficiency is often associated with cardiomyopathy and ischemic failure. The present study was designed to determine whether cardiac dysfunction is seen in an experimental model of short-terrm carnitine deficiency. Carnitine deficiency was induced in Sprague-Dawley rats by supplementing the drinking water with sodium pivalate for a period of 2 weeks. This resulted in a 25% depletion of total myocardial carnitine content. When isolated working hearts from these animals were paced and subjected to increments in left atrial filling pressure, there were no differences in mechanical function compared with control hearts. Following no-flow ischemia, however, recovery of cardiac output and relaxation parameters was depressed in hearts from pivalate-treated animals. Under these conditions, L-carnitine prevented the depressions of function from occurring. Our results show that short-term carnitine deficiency is not associated with cardiac dysfunction under normoxic conditions. However, hearts from pivalate-treated animals are more susceptible to ischemic injury and thus may prove to be useful for the study of metabolic and functional aspects of carnitine deficiency. 相似文献
