极端酶具有超常的生物学稳定性,能够在极端温度、pH、压力和离子强度下表现出生物学活性,因此极端酶为生物催化和生物转化提供了良机.新的极端物种的发现、基因组序列的确定及基因工程技术的应用,加快了发现和制备新酶的进程.蛋白质工程和定向进化技术进一步改善酶的活性和特异性,促进了极端酶的工业应用.对极端酶的研究加深了人们对酶稳定性机制的理解,丰富了分子进化理论.  相似文献

Human serum albumin (HSA) is the major protein component of human plasma. To date, HSA for clinical uses is mostly produced by fractionation of human whole blood, which is accompanied by a lot of limitations. To obtain long-term bioactive albumin, we used hsa as a foreign gene and constructed a recombinant plasmid pJS700-HSA which carries a recombinant gene cotC-hsa under the control of cotC promoter. Plasmid pJS700-HSA was transformed into Bacillus subtilis by double cross-over and an amylase inactivated mutant was produced. After induction of spore formation, western blot and fluorescence immunoassay were used to monitor HSA surface expression on spores. We estimated that HSA displayed on the spore accounted for 0.135 % of the total spore proteins and about 0.023 fg HSA were exposed on the surface of each spore. Oral administration to mice with spores displaying HSA implied that the recombinant spores may have potential ability to increase the serum albumin level in vivo due to the resistant characters of spores.  相似文献

生物电化学系统能促进微生物与电极间的相互作用,从而改变微生物的代谢状态。本工作为研究运动发酵单胞菌(Zymomonas mobilis)在电环境中的代谢表现,在外接3V电源的H型电化学发酵装置中测试了其发酵效能。结果表明,相比于无电压的对照,阳极甘油产量上升24%,阴极葡萄糖消耗上升16%,产物乙醇和琥珀酸的产量也分别上升13%和8%。转录组分析表明,代谢物的显著改变归因于电环境导致的有机酸代谢、氧化还原平衡、电子传递等通路的改变。从表达差异显著的基因中挑选了代表胞内氧化还原平衡、生物膜形成和电子传递的3个基因ZMO1060(编码超氧化物歧化酶)、ZMO0401(编码二鸟苷酸磷酸二酯酶)和ZMO1819(编码固氮蛋白)进行验证,结果表明过表达ZMO1060和ZMO1819能够更显著地改变生物电化学系统中Z.mobilis的代谢。本工作为应用生物电化学系统调控微生物代谢物生产提供了参考。  相似文献

居民对农业文化遗产的认知态度影响其行为选择,并对农业文化遗产的保护及可持续发展具有重要意义。基于人文地理学及环境心理学领域的地方认同理论,选择历史认同、现实认同、情感认同及行为认同4个维度变量,通过问卷调查,以福州居民对全球重要农业文化遗产"福州茉莉花与茶文化系统"的认知及保护态度作为研究对象,并通过构建福州居民农业文化遗产认知及保护规律定量分析居民认知态度、保护行为与人口特征之间的关系。结果表明:(1)福州居民对茉莉花与茶文化系统的行为认同维度得分高于其他维度,且福州市民各维度得分均高于外来居民;(2)受访居民对农业文化遗产的地方认同与其年龄、受教育水平、收入水平及在福州居住时间长短的关系较为密切;(3)通过构建福州居民农业文化遗产认知及保护规律发现,福州居民对农业文化遗产的保护行为主要受其对该遗产项目认知态度的影响,与居民人口特征相关性较弱。拓展农业文化遗产领域的研究视角及方法,促进农业文化遗产地动态保护与可持续发展具有参考价值。  相似文献

NPM1突变基因表达抑制K562白血病细胞体外增殖和侵袭   总被引:1,自引:1,他引:1  
核仁磷酸蛋白(nucleophosmin,NPM1)突变是近年发现的在急性髓系白血病中发挥重要作用的基因改变,为探讨NPM1突变对K562白血病细胞体外增殖和侵袭能力的影响,将载体pEGFPC1-NPM1-mA转染K562细胞系,构建稳定表达NPM1突变蛋白的白血病细胞株(K562-mA)。利用细胞生长曲线观察细胞体外增殖能力;流式细胞仪检测细胞周期进程改变;细胞粘附、Transwell实验分别用以观察细胞体外粘附、迁移及侵袭能力。结果发现,NPM1突变转染后K562细胞体外增殖能力明显减弱;同时G1期细胞比例明显增高,S期细胞比例显著减低。与未处理组和空载体转染组细胞相比,K562-mA细胞体外迁移能力有所增加,但细胞粘附及侵袭能力却明显减弱。提示NPM1突变基因的表达能够抑制白血病细胞体外增殖和侵袭能力,为进一步深入探讨NPM1突变在白血病发生发展中的调控机制奠定了良好的基础。  相似文献

Arsenite (As(III)), an effective chemotherapeutic agent for the acute promyelocytic leukemia (APL) and multiple myeloma (MM), might be also a promise for the therapy of other cancers, including the solid tumors. However, the molecular bases of arsenite‐induced cytotoxicity in the tumor cells have not been fully defined. In this study, we have disclosed that arsenite effectively induces the apoptotic response in the HepG2 human hepatoma cells by triggering GADD45α induction and the subsequent activation of JNKs/AP‐1 cell death pathway. However, signaling events relating to GADD45α/JNKs/AP‐1 pathway activation have not been observed in HL7702 human diploid hepatic cells under the same arsenite exposure condition. Our results thus have illustrated the selective pro‐apoptotic role of arsenite in the hepatoma cells by activating GADD45α‐dependent cell death pathway whereas with little effect on the normal hepatic cells. The approaches to up‐regulate GADD45α levels might be helpful in improving the chemotherapeutic action of arsenite on certain solid tumors including hepatoma. J. Cell. In this study, we investigate the role of liver X receptor alpha (LXR alpha) in lipogenesis in geese in order to understand the differences in hepatic steatosis mechanisms between mammals and waterfowl. Primary goose hepatocytes were isolated and treated with the LXR alpha agonist T0901317. Triglyceride (TG) accumulation, acetyl-CoA carboxylase alpha (ACC alpha) and fatty acid synthase (FAS) activities, and gene expression levels of LXR alpha, sterol regulatory element-binding proteins-1 (SREBP-1), FAS, ACC alpha and lipoprotein lipase (LPL) were measured in primary hepatocytes. We found a dose-dependent up-regulation of TG accumulation, ACC, and FAS activities and the mRNA levels of LXR alpha, SREBP-1, FAS, ACC alpha, and LPL genes in the presence of To-901317. We also found that binding of nuclear SREBP-1 to ACC alpha SRE sequence was induced by To-901317 (P < 0.05). In conclusion, LXR alpha is involved in the induction of the lipogenic pathway through activation of SREBP-1 and its target genes in goose primary hepatocytes.  相似文献

奇异果甜蛋白(thaumatin)是迄今为止最甜的物质之一,对其研究具有很重要的意义。奇异果甜蛋白的生化性质基本清楚,基因序列和氨基酸序列都已测定。它的甜味可能是由奇异果甜蛋白上特定基团和受体结合引起的。对奇异果甜蛋白的生理功能知之甚少。近二十年来,在奇异果甜蛋白的基因工程上取得了一定进展,但仍然存在许多困难。 Abstract:Thaumatin is one of the sweetest substances known to date,it is important to study the thaumatin.The biochemical properties of thaumatin have been clarified clearly.Thaumatin had been isolated and sequenced.The mechanism of the sweetness of thaumatin may be due to the combination of some special groups and the receptors.The exact function of thaumatin is still not clear.Although gene engineering of thaumatin has been carried out for 20 years,there are still some difficulties to be solved for using in the market.  相似文献

Lysophosphatidic acid (LPA) is a bioactive lipid that serves as an extracellular signaling molecule acting through cognate G protein-coupled receptors designated LPA(1-6) that mediate a wide range of both normal and pathological effects. Previously, LPA(1), a G(αi)-coupled receptor (which also couples to other G(α) proteins) to reduce cAMP, was shown to be essential for the initiation of neuropathic pain in the partial sciatic nerve ligation (PSNL) mouse model. Subsequent gene expression studies identified LPA(5), a G(α12/13)- and G(q)-coupled receptor that increases cAMP, in a subset of dorsal root ganglion neurons and also within neurons of the spinal cord dorsal horn in a pattern complementing, yet distinct from LPA(1), suggesting its possible involvement in neuropathic pain. We therefore generated an Lpar5 null mutant by targeted deletion followed by PSNL challenge. Homozygous null mutants did not show obvious base-line phenotypic defects. However, following PSNL, LPA(5)-deficient mice were protected from developing neuropathic pain. They also showed reduced phosphorylated cAMP response element-binding protein expression within neurons of the dorsal horn despite continued up-regulation of the characteristic pain-related markers Caα(2)δ(1) and glial fibrillary acidic protein, results that were distinct from those previously observed for LPA(1) deletion. These data expand the influences of LPA signaling in neuropathic pain through a second LPA receptor subtype, LPA(5), involving a mechanistically distinct downstream signaling pathway compared with LPA(1).  相似文献

甘薯分子遗传图谱的建立对甘薯分子育种技术体系的拓展和应用具有重要意义。当前,对甘薯分子遗传图谱的研究虽然取得一定的进展,但存在着很多技术瓶颈,如作图策略应用和优化等。总结了甘薯经典和分子遗传研究进展,剖析了甘薯分子遗传图谱作图的3种方法与策略;探讨和提出了提高甘薯作图效率和质量的途径主要是:优化作图群体质量、克服偏分离、整合多群体间遗传连锁图谱和选择合适的分子标记类型;并指出染色体关联在遗传作图中的重要性,提出甘薯分子育种领域亟待加强的方面,以期为今后甘薯精密分子图谱的建立及基于分子图谱的甘薯分子育种提供新的思路。  相似文献
