Grape pomace: A novel substrate for microbial production of citric acid

Summary Grape pomace was used as substrate for microbial production of citric acid. Of the five cultures examined,Aspergillus niger NRRL 567 was found to produce the greatest amount of citric acid from grape pomace in the presence of methanol at a concentration of 3% (vol/wt). The yield was 60% based on the amount of fermentable sugar consumed.     

In vitro growth and differentiation of human kidney tubular cells on a basement membrane substrate

Summary Kidney cortical tubular cells, mainly proximal tubular cells, isolated from human kidney and grown either on a basement membrane substrate in chemically defined medium or on plastic in serum-supplemented medium, had substantial proliferative potential and could be propagated for more than 10 generations or 8 passages before senescence. Basement membrane produced on a plastic substrate by the HR-9 endodermal cell line could replace serum supplementation in promoting tubular cell growth. Tubular cells grown on an HR-9 basement membrane substrate exhibited stable epithelial morphology over an extended period of time; in the presence of 5% serum they differentiated into organized structures such as hemicysts and cell cords. Cells grown on plastic failed to differentiate and gradually degenerated. Tubular cells on HR-9 basement membrane were characterized by densely packed microvilli, abundant rough endoplasmic reticulum and free polysomes, basal cell membrane interdigitations, a well-developed endocytotic apparatus, and conspicuous junctional complexes—all features of the proximal tubular cell. Compared with cells on plastic substrate, there were higher levels of the brush border enzymes γ-glutamyl transpeptidase,l-leucine aminopeptidase, and alkaline phosphatase in cells maintained on an HR-9 basement membrane substrate, further supporting the conclusion that a basement membrane substrate promoted differentiation of tubular cells. These data and morphological observations indicate that a basement membrane substrate can promote growth and both functional and morphologic differentiation of human kidney tubular cells. Influences of medium consistency and shoot density on in vitro shoot proliferation of Populus alba × P. grandidentata

The in vitro shoot proliferation of Populus alba × P. grandidentata was affected by the medium consistency and shoot density, but not by three sizes of vessels. After 4 weeks of culture, the fresh weight and number of shoots per explant on liquid medium were significantly greater than those on agar-solidified medium. In particular, 3.2 shoots, 7 mm or longer per explant, were produced on liquid medium compared with 1.6 shoots per explant or agar-solidified medium. The fresh weight per explant after 4 weeks of culture on liquid medium and agar-solidified medium were 0.68 and 0.25 g, respectively. Increasing the number of shoots per vessel slowed the growth of the explants as measured by fresh weight and the number of shoots produced. There was little difference in the number of shoots produced between vessels with 1 or 2 shoots per vessel, but there were many fewer shoots produced when 3 shoots were placed in each vessel.Journal Paper No. J-11977 of the Iowa Agriculture and Home Economics Experiment Station, Ames, Iowa. 宁夏同心中新世铲齿象化石

本文记述了宁夏同心地区中中新世哺乳动物群中铲齿象化石——同心铲齿象 (Platybelodon tongxinensis) (原定为同心嵌齿象 (Gomphotherium tongxinensis). 通过与国内外已现的有关铲齿象对比,确认其与国内已发现的葛氏种关系最近,可能属同一枝系.同心种与葛氏种相比,是性质相对原始分布层位较低的我国第二个铲齿象种.文章初步讨论了其 M_3 的某些变异性状.并结合现有的其它标本初步阐述了铲齿象类下门齿的两种结构类型,原始型的同心层状结构和衍生型的齿质柱状结构.建议根据这两种结构仍将铲齿象类划分为 Platybelodontinae 和 Amebelodontinae.     

Typing of Nontypable Staphylococci by Lysogeny

Strains of coagulase-positive staphylococci which were nontypable with the routine typing set of phages could be typed by lysogeny with phage-propagating strains as indicators and with ultraviolet induction. About 10% of the strains could be typed without induction. About 36% of them could be typed by this method when ultraviolet irradiation was used as an inducing agent. The phage groups from which the majority of the nontypable staphylococci originated were easily identified by this method of typing.     

Tubular Transport Maxima of PAH and Diodrast Measured Individually in the Aglomerular Kidney of Lophius, and Simultaneously as Competitors Under Conditions of Equimolar Loading

The maximal tubular transfer rates (Tm) of both p-aminohippurate (PAH) and diodrast (3,5-diiodo-4-pyridone-N-acetic acid or iodopyracet) were found to be fixed and reproducible when measured separately in Lophius (goosefish) under standard laboratory conditions. Expressed on a molar basis Tm_PAH was four times Tm_D. However, when these transport competitors were presented simultaneously in equimolar concentrations with the plasma levels of each sufficiently high enough to saturate the carrier system, the relative rates of excretion were reversed with the diodrast transfer rate then four times that of PAH. The combined rate of excretion was far below Tm_PAH alone, and roughly equal to Tm_D. Interaction with a common carrier was indicated by the gradations in degree of inhibition which resulted when plasma concentration ratios of diodrast to PAH were extended from 0.1 to 3.2, and PAH transfer rates expressed as percentage of Tm_PAH were correspondingly depressed from 17 to 1.0 per cent respectively. These observations again point up the inverse relationship between transfer rate and competitive effectiveness which exists for members of a series of substances actively transported by a common mechanism. It appears that carrier affinity and dissociation characteristics may be quite different for various compounds in a series, and also that these parameters may vary significantly from species to species.     

Islet-cell Carcinoma (Zollinger—Ellison Syndrome) with Fulminating Adrenocortical Hyperfunction and Hypokalemia

The production of ACTH-like material by tumours arising in non-endocrine tissue may initiate severe adrenocortical hyperfunction. The pathogenesis and clinical and laboratory features of Cushing''s syndrome associated with such tumours are characteristic. The autonomous production by the tumour of ACTH-like material cannot be suppressed by exogenous corticoids. The onset of clinical symptoms is rapid; muscle wasting, general weakness, thirst and peripheral edema predominate, and the classical signs of Cushing''s syndrome may be absent. High levels of plasma 17-hydroxycorticosteroids and urinary 17-hydroxycorticosteroids and 17-ketosteroids, usually with normal levels of urinary aldosterone, commonly occur. Hypokalemic alkalosis unresponsive to replacement therapy may cause death. In the case reported herein, the intriguing possibility exists that two hormone-like substances were produced by the primary growth and its metastases: one, ACTH-like, to account for the adrenal hyperplasia and Cushing''s syndrome; and another, gastrin-like, giving rise to the ulcerogenic diathesis.     

Production of citric acid by immobilized dried reactivatedAspergillus niger

Summary Immobilization followed by drying was fried as a new technique for obtaining small beads which might be more suitable for industrial fermentations and for the storage of the immobilized microorganisms.Aspergillus niger NRRL 2270 was used to produce citric acid in free, immobilized, and immobilized dried reactivated (IDR) forms. The productivity based on the bead volume used increased several folds with the use of IDR cells although the absolute level of citric acid did not increase.
