Background

Polysaccharides, one of the active ingredients in herbal medicine, are proved to enhance innate immunity against infections. The aim of this study is to explore the immunoregulatory ability of polysaccharides from Rhynchosia minima root in vitro and in vivo.

Methods

Polysaccharide fractions of R. minima root were obtained by chromatographic column. The content of NO was measured by spectrophotometry. The levels of cytokines (tumor necrosis factor-α, TNF-α; interleukin-6, IL-6; and monocyte chemoattractant protein-1, MCP-1) were determined by enzyme-linked immuno-sorbent assay (ELISA) kits. The translocation of p65 into the nucleus was imaged by confocal microscopy. The mRNA expression of TNF-α, IL-6, and MCP-1 was determined by quantitative real-time PCR. T-lymphocyte subgroups of spleen from immunosuppressive mouse were evaluated by flow cytometry.

Results

PRM3 remarkably enhanced the phagocytic ability of macrophages and promoted the release of NO and the secretion of cytokines (TNF-α, IL-6, and MCP-1) from macrophages. Simultaneously, PRM3 potently activated NF-κB signaling pathway via Toll-like receptor 4 (TLR4). In addition, PRM3 obviously increased the levels of serum cytokines, markedly up-regulated the percentages of CD3+ and CD4+ T lymphocytes and the CD4+/CD8+ ratio of splenocytes, and effectively attenuated cyclophosphamide induced immunosuppression in mice.

Conclusions

PRM3 profoundly enhanced the immune function in vitro and in vivo through TLR4-NF-κB pathway and is a promising candidate of immunopotentiator which could be applied in functional foods or drugs.

General significance

This study reported a polysaccharide PRM3 from R. minima root exhibited potent immunoenhancing activity and significantly alleviated cyclophosphamide-induced immunosuppression through TLR4-NF-κB pathway.  相似文献

The 67th Discussion Forum on Life Cycle Assessment (LCA), organised by partners of the European project RELIEF (RELIability of product Environmental Footprints), focused on methods for better understanding the impacts of land use linked to agricultural value chains. The first session of the forum was dedicated to methods that help in retrospective tracking of land use within complex supply chains. Novel approaches were presented for the integration of increasingly available spatially located land use data into LCA. The second session focused on forward-looking projections of land use change and included emerging, predictive methods for the modelling of land change. The third session considered impact assessment methods related to the use of land and their application together with land change modelling approaches. Discussions throughout the day centred on opportunities and challenges arising from integrating spatially located land use information into Life Cycle Assessment. Increasing amounts of spatially located land use data are becoming available and this could potentially increase the robustness and specificity of Life Cycle Assessment. However, the use of such data can be computationally expensive and requires the development of skills (i.e. use of geographical information systems (GIS) and model coding) within the LCA community. Land change modelling and ecosystem service modelling are associated with considerable uncertainty which must be communicated appropriately to stakeholders and decision-makers when interpreting results from an LCA. The new approaches were found to challenge aspects of the traditional LCA approach—particularly the division between the life cycle inventory and impact assessment and the assumption of linearity between scale and impacts when deriving characterisation factors. Lysinibacillus sphaericus has great application potential not only in the biocontrol of mosquitoes but also in the bioremediation of toxic metals. Phages contribute to the genetic diversity and niche adaptation of bacteria, playing essential roles in their life cycle, but may also cause economic damage for industrially important bacteria through phage contamination during fermentation. In this study, the L. sphaericus phage vB_LspM-01, which belongs to the Myoviridae family, was isolated and characterized. Results showed that vB_LspM-01 could specifically infect most tested L. sphaericus isolates but was not active against isolates belonging to other species. Furthermore, phage-born endolysin exhibited a broader antimicrobial spectrum than the host range of the phage. The vB_LspM-01 genome had no obvious similarity with that of its host, and ca. 22.6% of putative ORFs could not get a match with the public databases. Phylogenic analysis based on the putative terminase large subunit showed high similarity with the phages identified with pac-type headful packaging. The vB_LspM-01 encoding genes were only detected in a tiny percentage of L. sphaericus C3-41 individual cells in the wild population, whereas they showed much higher frequency in the resistant population grown within the plaques; however, the phage genes could not be stably inherited during host cell division. Additionally, the vB_LspM-01 encoding genes were only detected in the host population during the logarithmic growth phase. The mitomycin C induction helped the propagation and lysogeny-lysis switch of vB_LspM-01. The study demonstrated that vB_LspM-01 can be present in a pseudolysogenic state in L. sphaericus C3-41 populations.

Length‐weight relationships were described for five fish species belonging to three families and four genera from the Daninghe River, one of primary tributaries of the upper Yangtze River, China. A total of 1894 specimens were collected from 2014 to 2016 by using electro‐fishing equipment. Length–weight relationships for five species were recorded previously in FishBase and new maximum total lengths for five species were also presented in this paper. The results will be useful for fishery research, management and conservation in the upper Yangtze River Basin.  相似文献

The disordered phases of LiCB11H12 and NaCB11H12 possess superb superionic conductivities that make them suitable as solid electrolytes. In these materials, cation diffusion correlates with high orientational mobilities of the CB11H12? anions; however, the precise relationship has yet to be demonstrated. In this work, ab initio molecular dynamics and quasielastic neutron scattering are combined to probe anion reorientations and their mechanistic connection to cation mobility over a range of timescales and temperatures. It is found that anions do not rotate freely, but rather transition rapidly between orientations defined by the cation sublattice symmetry. The symmetry‐breaking carbon atom in CB11H12? also plays a critical role by perturbing the energy landscape along the instantaneous orientation of the anion dipole, which couples fluctuations in the cation probability density directly to the anion motion. Anion reorientation rates exceed 3 × 1010 s?1, suggesting the underlying energy landscape fluctuates dynamically on diffusion‐relevant timescales. Furthermore, carbon is found to modify the orientational preferences of the anions and aid rotational mobility, creating additional symmetry incompatibilities that inhibit ordering. The results suggest that synergy between the anion reorientational dynamics and the carbon‐modified cation–anion interaction accounts for the higher ionic conductivity in CB11H12? salts compared with B12H122?.  相似文献

A new small‐molecule acceptor (2,9‐bis(2‐methylene‐(3(1,1‐dicyanomethylene)benz[f]indanone))7,12‐dihydro‐(4,4,10,10‐tetrakis(4‐hexylphenyl)‐5,11‐diocthylthieno[3′,2′:4,5]cyclopenta[1,2‐b]thieno[2″,3″:3′,4′]cyclopenta[1′,2′:4,5]thieno[2,3‐f][1]benzothiophene) (NNBDT) based on naphthyl‐fused indanone ending units is reported. This molecule shows a narrow optical bandgap of 1.43 eV and effective absorption in the range of 700–870 nm. The devices based on poly[(2,6‐(4,8‐bis(5‐(2‐ethylhexyl)thiophen‐2‐yl)‐benzo[1,2‐b:4,5‐b′]dithiophene))‐alt‐(5,5‐(1′,3′‐di‐2‐thienyl‐5′,7′‐bis(2‐ethylhexyl)benzo[1′,2′‐c:4′,5′‐c′]dithiophene‐4,8‐dione))] (PBDB‐T):NNBDT yield a power conversion efficiency of 11.7% with a low energy loss of 0.55 eV and a high fill factor (FF) of 71.7%. Another acceptor (2,9‐bis(2‐methylene‐(3(1,1‐dicyanomethylene)benz[f]indanone))7,12‐dihydro‐4,4,7,7,12,12‐hexaoctyl‐4H‐cyclopenta[2″,1″:5,6;3″,4″:5′,6′]diindeno[1,2‐b:1′,2′‐b′]dithiophene (FDNCTF) is introduced as the third component to fabricate ternary devices. The two acceptors (NNBDT and FDNCTF) possess complementary absorption, same molecular orientation, and well‐miscible behavior. It is found that there exists a nonradiative energy transfer process from FDNCTF to NNBDT. The fullerene‐free ternary cells based on PBDB‐T:NNBDT:FDNCTF achieve a high efficiency of 12.8% with an improved short circuit current near 20 mA cm?2 in contrast to the binary devices. The result represents the best performance for fullerene‐free ternary solar cells reported to date and highlights the potential of ternary solar cells.  相似文献

Hard carbon has long been considered the leading candidate for anode materials of Na‐ion batteries. Intensive research efforts have been carried out in the search of suitable carbon structure for an improved storage capability. Herein, an anode based on multishelled hollow carbon nanospheres, which are able to deliver an outstanding electrochemical performance with an extraordinary reversible capacity of 360 mAh g?1 at 30 mA g?1, is designed. An interesting dependence of the electrochemical properties on the multishelled structural features is identified: with an increase in the shell number of the model carbon materials, the sloping capacity in the charge/discharge curve remains almost unchanged while the plateau capacity continuously increases, suggesting an adsorption‐filling Na‐storage mechanism for the multishelled hollow hard carbon materials. The findings not only provide new perspective in the structural design of high‐performance anode materials, but also shed light on the complicated mechanism behind Na‐storage by hard carbon.  相似文献

Gram-positive, aerobic, non-motile, pale-yellow, and rodshaped bacterium, designated as Gsoil 188T, was isolated from the soil of a ginseng field in Pocheon, South Korea. A phylogenetic analysis based on 16S rRNA gene sequence comparison revealed that the strain formed a distinct lineage within the genus Brevibacterium and was most closely related to B. epidermidis NBRC 14811T (98.4%), B. sediminis FXJ8.269T (98.2%), B. avium NCFB 3055T (98.1%), and B. oceani BBH7T (98.1%), while it shared less than 98.1% identity with the other species of this genus. The DNA G + C content was 68.1 mol%. The predominant quinone was MK-8(H2). The major fatty acids were anteiso-C15:0 and anteiso-C17:0. The cell wall peptidoglycan of strain Gsoil 188T contained meso-diaminopimelic acid. The major polar lipids were phosphatidylglycerol, diphosphatidylglycerol, and an unidentified aminolipid. The physiological and biochemical characteristics, low DNA-DNA relatedness values, and taxonomic analysis allowed the differentiation of strain Gsoil 188T from the other recognized species of the genus Brevibacterium. Therefore, strain Gsoil 188T represents a novel species of the genus Brevibacterium, for which the name Brevibacterium anseongense sp. nov. is proposed, with the type strain Gsoil 188T (= KACC 19439T = LMG 30331T).  相似文献
