Laboratory and field assessment of molluscicidal activity of B-2 against Oncomelania hupensis, the vector snail of schistosomiasis in China

Molluscicidal activity of B-2 (sodium 2,5-dichloro-4-bromophenol; called as Phebrol and registered in WHO as OMS 3012) was evaluated in a laboratory and the field trials were performed in two different localities in Yueyang city, China, for control of Oncomelania hupensis. B-2 was effective against O. hupensis both in the laboratory and in the field. A dosage of 50 g/m2 in 10% granular form or 20 ml/m2 in 25% liquid form of B-2 would be recommendable as a standard mollusciciding dose for control of O. hupensis.     

Congenital Postero-Lateral Diaphragmatic Hernia in Children

Congenital diaphragmatic hernia must be seriously considered in the differential diagnosis of acute respiratory distress in neonates as well as in older infants who present with chronic progressive gastrointestinal and respiratory symptoms. An x-ray film of the chest is mandatory and is almost always diagnostic. Immediate nasogastric decompression of the stomach, with simultaneous fluid and electrolyte replacement and prompt surgical intervention offers the patient the best chance of survival.     

α-Tocopherol is an effective Phase II enzyme inducer: protective effects on acrolein-induced oxidative stress and mitochondrial dysfunction in human retinal pigment epithelial cells

Vitamin E has long been identified as a major lipid-soluble chain-breaking antioxidant in mammals. α-Tocopherol is a vitamin E component and the major form in the human body. We propose that, besides its direct chain-breaking antioxidant activity, α-tocopherol may exert an indirect antioxidant activity by enhancing the cell's antioxidant system as a Phase II enzyme inducer. We investigated α-tocopherol's inducing effect on Phase II enzymes and its protective effect on acrolein-induced toxicity in a human retinal pigment epithelial (RPE) cell line, ARPE-19. Acrolein, a major component of cigarette smoke and also a product of lipid peroxidation, at 75 μmol/L over 24 h, caused significant loss of ARPE-19 cell viability, increased oxidative damage, decreased antioxidant defense, inactivation of the Keap1/Nrf2 pathway, and mitochondrial dysfunction. ARPE-19 cells have been used as a model of smoking- and age-related macular degeneration. Pretreatment with α-tocopherol activated the Keap1/Nrf2 pathway by increasing Nrf2 expression and inducing its translocation to the nucleus. Consequently, the expression and/or activity of the following Phase II enzymes increased: glutamate cysteine ligase, NAD(P)H:quinone oxidoreductase 1, heme-oxygenase 1, glutathione S-transferase and superoxide dismutase; total antioxidant capacity and glutathione also increased. This antioxidant defense enhancement protected ARPE-19 cells from an acrolein-induced decrease in cell viability, lowered reactive oxygen species and protein oxidation levels, and improved mitochondrial function. These results suggest that α-tocopherol protects ARPE-19 cells from acrolein-induced cellular toxicity, not only as a chain-breaking antioxidant, but also as a Phase II enzyme inducer.     

微生物异源合成咖啡酸及其酯类衍生物研究进展

咖啡酸及其酯类衍生物如绿原酸、迷迭香酸和咖啡酸苯乙酯等具有天然抗氧化、抗肿瘤、抗病毒和抗炎等重要的药理活性,具有广阔的药用开发前景。从天然药物中提取或者化学合成咖啡酸及其酯类衍生物,存在含量低、提取效率不高、催化成本高昂以及环境污染等问题。随着咖啡酸及其酯类衍生物合成途径解析和合成生物学的快速发展,微生物异源合成咖啡酸及其酯类衍生物的研究已逐渐展开。对微生物异源合成咖啡酸及其酯类衍生物合成途径的最新进展以及代谢工程策略进行了综述,并讨论了目前存在的问题和未来的发展趋势。

Mechanism of indoleamine 2, 3-dioxygenase inhibiting cardiac allograft rejection in mice

Indoleamine 2, 3-dioxygenase (IDO)-mediated regulation of tryptophan metabolism plays an important role in immune tolerance in transplantation, but it has not been elucidated which mechanism specifically induces the occurrence of immune tolerance. Our study revealed that IDO exerts immunosuppressive effects through two pathways in mouse heart transplantation, ‘tryptophan depletion’ and ‘tryptophan metabolite accumulation’. The synergism between IDO⁺DC and TC (tryptophan catabolic products) has stronger inhibitory effects on T lymphocyte proliferation and mouse heart transplant rejection than the two intervention factors alone, and significantly prolong the survival time of donor-derived transplanted skin. This work demonstrates that the combination of IDO⁺DC and TC can induce immune tolerance to a greater extent, and reduce the rejection of transplanted organs.     

Novel mutations in ADAMTS13 CUB domains cause abnormal pre-mRNA splicing and defective secretion of ADAMTS13

Hereditary thrombotic thrombocytopenic purpura (TTP) is an autosomal recessive thrombosis disorder, caused by loss-of-function mutations in ADAMTS13. Mutations in the CUB domains of ADAMTS13 are rare, and the exact mechanisms through which these mutations result in the development of TTP have not yet been fully elucidated. In this study, we identified two novel mutations in the CUB domains in a TTP family with an acceptor splice-site mutation (c.3569−1, G>A, intron 25) and a point missense mutation (c.3923, G>A, exon 28), resulting in a glycine to aspartic acid substitution (p.G1308D). In vitro splicing analysis revealed that the intronic mutation resulted in abnormal pre-mRNA splicing, and an in vitro expression assay revealed that the missense mutation significantly impaired ADAMTS13 secretion. Although both the patient and her brother displayed significantly reduced ADAMTS13 activity and increased levels of ultra-large VWF (ULVWF) multimers in plasma, only the female developed acute episodes of TTP. Our findings indicate the importance of the CUB domains for the protein stability and extracellular secretion of ADAMTS13.
