The structural basis for the coupling of ATP binding and hydrolysis to chaperone activity remains a central question in Hsp90 biology. By analogy to MutL, ATP binding to Hsp90 is thought to promote intramolecular N-terminal dimerization, yielding a molecular clamp functioning in substrate protein activation. Though observed in studies with recombinant domains, whether such quaternary states are present in native Hsp90s is unknown. In this study, native subunit interactions in GRP94, the endoplasmic reticulum Hsp90, were analyzed using chemical cross-linking in conjunction with tandem mass spectrometry. We report the identification of two distinct intermolecular interaction sites. Consistent with previous studies, one site comprises the C-terminal dimerization domain. The remaining site represents a novel intermolecular contact between the N-terminal and middle (M) domains of opposing subunits. This N+M domain interaction was present in the nucleotide-empty, ADP-, ATP-, or geldanamycin-bound states and could be selectively disrupted upon addition of synthetic geldanamycin dimers. These results identify a compact, intertwined quaternary conformation of native GRP94 and suggest that intersubunit N+M interactions are integral to the structural biology of Hsp90.  相似文献

It has been predicted that elevated atmospheric CO2 will increase enzyme activity as a result of CO2-induced carbon entering the soil. The objective of this study was to investigate the effects of elevated atmospheric CO2 on soil enzyme activities under a rice/wheat rotation. This experiment was conducted in Wuxi, Jiangsu, China as part of the China FACE (Free Air Carbon Dioxide Enrichment) Project. Two atmospheric CO2 concentrations (580±60) and (380±40) μmol·mol-1) and three N application treatments (low-150, normal-250 and high-350 kg N·hm-2) were included. Soil samples (0-10 cm) were collected for analysis of β-glucosidase, invertase, urease, acid phosphates and β-glucosaminidase activities. The results revealed that with elevated atmospheric CO2 β-glucosidase activity significantly decreased (P < 0.05) at low N application rates; had no significant effect with a normal N application rate; and significantly increased (P < 0.05) with a high N application rate. For urease activity, at low and normal N application rates (but not high N application rate), elevated atmospheric CO2 significantly increased (P < 0.05) it. With acid phosphatase elevated atmospheric CO2 only had significant higher effects (P < 0.05) at high N application rates. Under different CO2 concentration, effects of N fertilization are also different. Soil β-glucosidase activity at ambient CO2 concentration decreased with N fertilization, while it increased at elevated CO2 concentration. In addition, invertase and acid phosphatase activities at elevated CO2 concentration, significantly increased (P < 0.05) with N treatments, but there was no effect with the ambient CO2 concentration. For urease activity, at ambient CO2 concentration, N fertilization increased it significantly (P < 0.05), whereas at elevated CO2 concentration it was not significant. Additionally, with β-glucosaminidase activity, there were no significant effects from N application. In general, then, elevated atmospheric CO2 increased soil enzyme activity, which may be attributed to the following two factors: (1) elevated atmospheric CO2 led to more plant biomass in the soil, which in turn stimulated soil microbial biomass and activity; and (2) elevated atmospheric CO2 increased plant photosynthesis, thereby increasing plant-derived soil enzymes.  相似文献

利用噬菌体展示技术已选出了多条与靶结合的肽. 然而,即使是体内直接筛选得到的,肽与肿瘤或靶器官的体内结合并不理想. 为了更好地理解噬菌体在体内的循环, 通过MAG3 99mTc标记噬菌体肽库,研究了肽库在体内分布. 体内分布实验结果显示,99mTc-噬菌体主要分布在肝和脾中,而心脏、肌肉、脑和胰腺这些器官或组织中的分布非常低. 99mTc-噬菌体在胃、肠和骨中的累积,随着时间延长在不断升高,其他器官中的吸收则在不断降低. 从5 min到30 min,99mTc-噬菌体在血中清除迅速. 当噬菌体在体内循环足够长的时间后,一些噬菌体颗粒可以穿透血管进入并内化在器官或组织中. 总之,为了筛选具有高特异性和亲和性的肽,应该根据靶器官和筛选部位的不同,在筛选前确定合适的噬菌体在体内的循环时间.  相似文献

Coronaviruses are the causative agents of respiratory disease in humans and animals, including severe acute respiratory syndrome. Fusion of coronaviruses is generally thought to occur at neutral pH, although there is also evidence for a role of acidic endosomes during entry of a variety of coronaviruses. Therefore, the molecular basis of coronavirus fusion during entry into host cells remains incompletely defined. Here, we examined coronavirus-cell fusion and entry employing the avian coronavirus infectious bronchitis virus (IBV). Virus entry into cells was inhibited by acidotropic bases and by other inhibitors of pH-dependent endocytosis. We carried out fluorescence-dequenching fusion assays of R18-labeled virions and show that for IBV, coronavirus-cell fusion occurs in a low-pH-dependent manner, with a half-maximal rate of fusion occurring at pH 5.5. Fusion was reduced, but still occurred, at lower temperatures (20 degrees C). We observed no effect of inhibitors of endosomal proteases on the fusion event. These data are the first direct measure of virus-cell fusion for any coronavirus and demonstrate that the coronavirus IBV employs a direct, low-pH-dependent virus-cell fusion activation reaction. We further show that IBV was not inactivated, and fusion was unaffected, by prior exposure to pH 5.0 buffer. Virions also showed evidence of reversible conformational changes in their surface proteins, indicating that aspects of the fusion reaction may be reversible in nature.  相似文献

Mucolipidosis type IV is a genetic lysosomal storage disease associated with degenerative processes in the brain, eye, and other tissues. Mucolipidosis type IV results from mutations in the gene MCOLN1, which codes for the TRP family ion channel, mucolipin 1. The connection between lysosomal dysfunction and degenerative processes in mucolipidosis type IV is unclear. Here we report that mucolipidosis type IV and several unrelated lysosomal storage diseases are associated with significant mitochondrial fragmentation and decreased mitochondrial Ca2+ buffering efficiency. The mitochondrial alterations observed in these lysosomal storage diseases are reproduced in control cells by treatment with lysosomal inhibitors and with the autophagy inhibitor 3-methyladenine. This suggests that inefficient autophagolysosomal recycling of mitochondria generates fragmented, effete mitochondria in mucolipidosis. Mitochondria accumulate that cannot properly buffer calcium fluxes in the cell. A decrease in mitochondrial Ca2+ buffering capacity in cells affected by these lysosomal storage diseases is associated with increased sensitivity to apoptosis induced by Ca2+-mobilizing agonists and executed via a caspase-8-dependent pathway. Deficient Ca2+ homeostasis may represent a common mechanism of degenerative cell death in several lysosomal storage diseases.  相似文献

海南黑脸琵鹭的越冬行为分析   总被引:9,自引:1,他引:8  
黑脸琵鹭(Plataleaminor)是全球濒危鸟类之一。海南北黎湾是继我国香港和台湾之后发现的黑脸琵鹭第三个重要的越冬地。为了深入了解黑脸琵鹭在该地的保护状况,以及影响其种群越冬的致危因素,作者于2004年1–3月,采用瞬时扫描和焦点动物观察法对北黎湾黑脸琵鹭的越冬行为进行了观察。结果表明:(1)在黑脸琵鹭的各种行为中,休息行为所占的比例最大,为69.72%;其次是站立行为,为10.42%;护理行为位居第3,占10.36%;位移、取食和社会行为在白天所占的比例相对较低。(2)白天黑脸琵鹭休息行为有明显的高峰和低谷,8:00–17:00主要以休息为主,而在17:00以后,休息行为明显减少,护理、取食和站立行为有所增加。位移和社会行为发生率较低,呈间歇性变化。黑脸琵鹭各种行为之间在持续时间上有显著差异,其中以休息行为持续时间最长,其次是取食行为,而行走行为持续时间最短。(3)黑脸琵鹭的取食频次在各时间段也有显著的差异,在16:00以后取食的频次较高。(4)黑脸琵鹭的取食行为受潮汐的影响,过高和过低的水位都将影响其取食活动。最后针对性地提出了黑脸琵鹭及其越冬地的保护和管理计划。  相似文献
