To provide a definitive linkage map for multiple sclerosis, we have genotyped the Illumina BeadArray linkage mapping panel (version 4) in a data set of 730 multiplex families of Northern European descent. After the application of stringent quality thresholds, data from 4,506 markers in 2,692 individuals were included in the analysis. Multipoint nonparametric linkage analysis revealed highly significant linkage in the major histocompatibility complex (MHC) on chromosome 6p21 (maximum LOD score [MLS] 11.66) and suggestive linkage on chromosomes 17q23 (MLS 2.45) and 5q33 (MLS 2.18). This set of markers achieved a mean information extraction of 79.3% across the genome, with a Mendelian inconsistency rate of only 0.002%. Stratification based on carriage of the multiple sclerosis–associated DRB1*1501 allele failed to identify any other region of linkage with genomewide significance. However, ordered-subset analysis suggested that there may be an additional locus on chromosome 19p13 that acts independent of the main MHC locus. These data illustrate the substantial increase in power that can be achieved with use of the latest tools emerging from the Human Genome Project and indicate that future attempts to systematically identify susceptibility genes for multiple sclerosis will have to involve large sample sizes and an association-based methodology. 相似文献

Three new species of Neotibilis Grazia & Barcellos are described from Brazilian Amazon region and Ecuador, based on general and genital morphology: Neotibilis (Neotibilis) claviformis sp. nov., Neotibilis (Neotibilis) ecuadorensis sp. nov. and Neotibilis (Neotibilis) manauara sp. nov. Illustrations and a key to species of the type subgenus are given. The range of Neotibilis is expanded, with new records for N. (N.) biguttata (Walker)--Brazil (Amazonas) and N. Dengue is hyperendemic in Brazil, with outbreaks affecting all regions. Previous studies identified geographical barriers to dengue transmission in Brazil, beyond which certain areas, such as South Brazil and the Amazon rainforest, were relatively protected from outbreaks. Recent data shows these barriers are being eroded. In this study, we explore the drivers of this expansion and identify the current limits to the dengue transmission zone. We used a spatio-temporal additive model to explore the associations between dengue outbreaks and temperature suitability, urbanisation, and connectivity to the Brazilian urban network. The model was applied to a binary outbreak indicator, assuming the official threshold value of 300 cases per 100,000 residents, for Brazil’s municipalities between 2001 and 2020. We found a nonlinear relationship between higher levels of connectivity to the Brazilian urban network and the odds of an outbreak, with lower odds in metropoles compared to regional capitals. The number of months per year with suitable temperature conditions for Aedes mosquitoes was positively associated with the dengue outbreak occurrence. Temperature suitability explained most interannual and spatial variation in South Brazil, confirming this geographical barrier is influenced by lower seasonal temperatures. Municipalities that had experienced an outbreak previously had double the odds of subsequent outbreaks. We identified geographical barriers to dengue transmission in South Brazil, western Amazon, and along the northern coast of Brazil. Although a southern barrier still exists, it has shifted south, and the Amazon no longer has a clear boundary. Few areas of Brazil remain protected from dengue outbreaks. Communities living on the edge of previous barriers are particularly susceptible to future outbreaks as they lack immunity. Bumblebees are widely distributed across the world and have great economic and ecological importance as pollinators in the forest as well as in agriculture. The insect midgut consists of three cell types, which play various important roles in digestion, absorption, and hormone production. The present study characterized the anterior and posterior midgut regions of the bumblebee, Bombus morio. The digestive, regenerative and endocrine cells in the midgut showed regional differences in their number, nuclear size, as well as the size of the striated border. Ultrastructurally, the digestive cells contained many mitochondria and long microvilli; however, in the anterior midgut region, these cells showed dilated basal labyrinths with a few openings for the hemocoel, whereas the labyrinths of the basal posterior region remained inverse characteristics. Thus, the characterization of the midgut of B. morio supported an ecto-endoperitrophic circulation, contributing to a better understanding of the digestive process in this bee. 相似文献

Background
The HIV-Brazil Cohort Study was established to analyze the effectiveness of combination antiretroviral therapy (cART) and the impact of this treatment on morbidity, quality of life (QOL) and mortality. The study design, patients’ profiles and characteristics of cART initiation between 2003 and 2010 were described.Methodology/Principal Findings
Since 2003, the HIV-Brazil Cohort has been following HIV-infected adults receiving cART at 26 public health care facilities, using routine clinical care data and self-reported QOL questionnaires. When not otherwise available, data are obtained from national information systems. The main outcomes of interest are diseases related or unrelated to HIV; suppression of viral replication; adverse events; virological, clinical and immunological failures; changes in the cART; and mortality. For the 5,061 patients who started cART between 2003 and 2010, the median follow-up time was 4.1 years (IQR 2.2–5.9 years) with an 83.4% retention rate. Patient profiles were characterized by a predominance of men (male/female ratio 1.7∶1), with a mean age of 36.9 years (SD 9.9 years); 55.2% had been infected with HIV via heterosexual contact. The majority of patients (53.4%) initiated cART with a CD4+ T-cell count ≤200 cells/mm3. The medications most often used in the various treatment regimens were efavirenz (59.7%) and lopinavir/ritonavir (18.2%). The proportion of individuals achieving viral suppression within the first 12 months of cART use was 77.4% (95% CI 76.1–78.6). Nearly half (45.4%) of the patients presented HIV-related clinical manifestations after starting cART, and the AIDS mortality rate was 13.9 per 1,000 person-years.Conclusions/Significance
Results from cART use in the daily practice of health services remain relatively unknown in low- and middle-income countries, and studies with the characteristics of the HIV-Brazil Cohort contribute to minimizing these shortcomings, given its scope and patient profile, which is similar to that of the AIDS epidemic in the country. 相似文献18.
Iron (Fe) minerals play an important role in carbon (C) and nutrient dynamics in redox fluctuating soils. We explored how the frequency of redox oscillations influence Fe reduction rates and C content in Puerto Rican soils. We hypothesized that iron reduction rates would be faster during short oscillation periods than in longer oscillation periods. Surface soils from an upland valley in a humid tropical forest were exposed to systematic redox oscillations over 49 days. The oxidation events were triggered by the introduction of air (21% O2), maintaining the time ratio under oxic or anoxic conditions at 1:6 (τox/τanox). After pre-conditioning the soil to fluctuating redox conditions for 1 month, we imposed 280- and 70-h (or 11.67- and 2.5-day) redox oscillations, measuring FeII every few days. We found that by the end of the experiment, Fe reduction rates were higher in the short oscillation period (τox = 10 h, τanox = 60 h) than in the long oscillation period (τox = 40 h, τanox = 240 h). Carbon and nitrogen loss however was similar for both treatments. These results suggest the characteristics of redox fluctuations can alter rates of Fe reduction and potentially influence ecosystem processes that depend on iron behavior. 相似文献

Contamination of soils and groundwater by chlorobenzene and benzene is a common problem at industrial sites worldwide. Since chemical remediation techniques are rarely completely effective, remnants of these contaminants often persist at levels that can still influence ecosystem health. We evaluated the potential of Pinus taeda and Eucalyptus urograndis to accelerate the removal of these compounds from sand/water systems using a completely randomized block greenhouse experiment with a no-plant control. At 2-day intervals, we added a solution containing both chlorobenzene and benzene with the same concentration of 50 mg L?1 (25 mg pot?1), and we monitored leachate concentrations daily. The planted treatments showed greater decrease of contaminants over time. In the absence of plants, the contaminant mass decreased 50–60% during each 2-day cycle; whereas, in the planted treatments the contaminant mass decreased 91–98%. At the end of the experiment the plant roots, leaves, and the sand-substrate each contained less than 1 mg kg?1 of contaminants, which is ~1% of the total contaminant mass added. In addition, we observed no tree mortality even at concentrations exceeding the aqueous solubility limit of both compounds. Our results suggest both trees are good candidates for remediating chlorobenzene and benzene in soils and groundwater. 相似文献
