Bacillithiol is a glucosamine‐derived antioxidant found in several pathogenic Gram‐positive bacteria. The compound is involved in maintaining the appropriate redox state within the cell as well as detoxifying foreign agents like the antibiotic fosfomycin. Bacillithiol is produced via the action of three enzymes, including BshA, a retaining GT‐B glycosyltransferase that utilizes UDP‐N‐acetylglucosamine and l ‐malate to produce N‐acetylglucosaminyl‐malate. Recent studies suggest that retaining GT‐B glycosyltransferases like BshA utilize a substrate‐assisted mechanism that goes through an SNi‐like transition state. In a previous study, we relied on X‐ray crystallography as well as computational simulations to hypothesize the manner in which substrates would bind the enzyme, but several questions about substrate binding and the role of one of the amino acid residues persisted. Another study demonstrated that BshA might be subject to feedback inhibition by bacillithiol, but this phenomenon was not analyzed further to determine the exact mechanism of inhibition. Here we present X‐ray crystallographic structures and steady‐state kinetics results that help elucidate both of these issues. Our ligand‐bound crystal structures demonstrate that the active site provides an appropriate steric and geometric arrangement of ligands to facilitate the substrate‐assisted mechanism. Finally, we show that bacillithiol is competitive for UDP‐N‐acetylglucosamine with a Ki value near 120–130 μM and likely binds within the BshA active site, suggesting that bacillithiol modulates BshA activity via feedback inhibition. The work presented here furthers our understanding of bacillithiol metabolism and can aid in the development of inhibitors to counteract resistance to antibiotics such as fosfomycin.  相似文献

Modifications of the Illinois River and associated tributaries have resulted in altered hydrologic cycles and persistent river‐floodplain connections during the growing season that frequently impede the establishment of hydrophytic vegetation and have reduced value for migratory waterfowl and other waterbirds. To help guide floodplain restoration, we compared energetic carrying capacity for waterfowl in two wetland complexes along the Illinois River under different management regimes during 2012–2015. The south pool of Chautauqua National Wildlife Refuge (CNWR) was seasonally flooded due to a partial river connection and managed for moist‐soil vegetation. Emiquon Preserve was hydrologically isolated from the Illinois River by a high‐elevation levee and managed as a semipermanently flooded emergent marsh. Semipermanent emergent marsh management at Emiquon Preserve produced 5,495 energetic use‐days (EUD)/ha for waterfowl and other waterbirds across wetland cover types and years, and seasonal moist‐soil management at CNWR produced 6,199 EUD/ha in one of 4 years. At Emiquon Preserve, the aquatic bed cover type produced 9,660 EUD/ha, followed by 5,261 EUD/ha in moist‐soil, 1,398 EUD/ha in persistent emergent, 1,185 EUD/ha in hemi‐marsh, and 12 EUD/ha in open water cover types. At CNWR, the annual grass and sedge cover type produced 7,031 EUD/ha, followed by 5,618 EUD/ha in annual broadleaf and 1,305 EUD/ha in perennial grass cover types. Restoration of floodplain wetlands in isolation from frequent flood pulses during the growing season can produce hemi‐marsh and aquatic bed vegetation communities that provide high‐quality habitat for waterfowl and which have been mostly eliminated from large river systems in the Midwest, U.S.A.  相似文献

Few comparisons exist between vertical accretion (VA) and carbon accumulation rates (CARs) in restored versus historic (i.e. reference) marshes. Here, we compare these processes in a formerly diked, sparsely vegetated, restored salt marsh (Six Gill Slough, SG), whose surface is subsided relative to the tidal frame, to an adjacent, relatively pristine, historic salt marsh (Animal Slough, AS). Six sediment cores were collected at both AS and SG approximately 6 years after restoration. Cores were analyzed for bulk density (BD), % loss of ignition, % organic carbon, and 210Pb. We found that sharp changes in BD in surface layers of SG cores were highly reliable markers for the onset of restoration. The mean VA since restoration at SG (0.79 [SD = 0.29] cm/year) was approximately twice that of AS (0.41 [SD = 0.16] cm/year). In comparison, the VA at AS over 50 years was 0.30 (SD = 0.09) cm/year. VA consisted almost entirely of inorganic sediment at SG whereas at AS it was approximately 55%. Mean CARs at SG were somewhat greater than at AS, but the difference was not significant due to high variability (SG: 81–210 g C m?2 year?1; AS: 115–168 g C m?2 year?1). The mean CAR at AS over the past 50 years was 118 (SD = 23) g C m?2 year?1. This study demonstrates that a sparsely vegetated, restored salt marsh can quickly begin to accumulate carbon and that historic and restored marshes can have similar CARs despite highly divergent formation processes.  相似文献

This is the first X-ray crystal structure of the monomeric form of sulfite reductase (SiR) flavoprotein (SiRFP-60) that shows the relationship between its major domains in an extended position not seen before in any homologous diflavin reductases. Small angle neutron scattering confirms this novel domain orientation also occurs in solution. Activity measurements of SiR and SiRFP variants allow us to propose a novel mechanism for electron transfer from the SiRFP reductase subunit to its oxidase metalloenzyme partner that, together, make up the SiR holoenzyme. Specifically, we propose that SiR performs its 6-electron reduction via intramolecular or intermolecular electron transfer. Our model explains both the significance of the stoichiometric mismatch between reductase and oxidase subunits in the holoenzyme and how SiR can handle such a large volume electron reduction reaction that is at the heart of the sulfur bio-geo cycle.  相似文献

The cosmopolitan nonindigenous sea anemone, Diadumene lineata, has been recorded from the Pacific coast of Canada since 1859. Despite its long historical presence in the region, the factors influencing the distribution of this species in British Columbia, Canada, are poorly understood, as are the factors that may restrict its local abundance. We conducted large‐scale surveys of D. lineata during the winter and summer of 2016, coupled with higher frequency surveys at one focal site. Furthermore, we used laboratory and field experiments to determine the potential importance of a large‐scale driver, salinity, and a small‐scale driver, microhabitat within boulders, in setting distributional limits at various spatial scales. Our results confirm that D. lineata is widely distributed within the Salish Sea. This broad distribution may be facilitated by post‐introduction dispersal; we observed anemones attached to floating substrata, and a few mature specimens contained eggs, suggesting that reproduction is perhaps not limited to asexual fission. We found that D. lineata was abundant throughout the year, although absent from the lowest salinity sites. This corresponded with our laboratory experiments, in which survival of individuals of D. lineata declined at salinities below 10 psu, and no survivors were observed at or below 2 psu. Manipulative experiments in the field demonstrated that survival was high under rocks where D. lineata is commonly found, but was reduced on the tops of rocks, highlighting the importance of microhabitat refugia in buffering temperature and reducing desiccation. A better understanding of the factors that limit the broad‐scale and local distribution of this anemone are an important step in determining potential ecological impacts of this organism and how distributions may shift into the future.  相似文献

House flies are global pests and notoriously difficult to control. Essential oils of vetiver, cinnamon, and lavender and their blends were tested for toxic and repellent effects against larval and adult flies. All of the oils had moderate toxicity for eggs. Mortality of 2nd instar larvae was 57–78% in dipping assays, 38–100% in contact assays, and 94–100% in treated media. Lavender was less effective (38% mortality) than the others (91–100%) in contact bioassays. Oil blends were not more effective against larvae than individual oils. Vetiver and cinnamon oils were strongly repellent (84 and 78%, respectively) for larvae in treated media. None of the oils were repellent for adult house flies in olfactometer assays, but testing of additional products demonstrated significant repellency for neem oil, p‐menthane‐3,8‐diol (PMD), and vanillin. Contact/fumigant toxicity of vetiver, cinnamon, and lavender oils was 100%, significantly higher than mortality from sunflower oil (67%). Blends of oils were not more effective against adults than the individual oils, but blends diluted with sunflower oil were as effective as the individual oils. Essentials oils of vetiver and cinnamon may have potential for fly management in situations where conventional insecticides cannot be used.  相似文献

Despite our growing understanding of the impacts of invasive plants on ecosystem structure and function, important gaps remain, including whether native and exotic species respond differently to plant invasion. This would elucidate basic ecological interactions and inform management. We performed a meta‐analytic review of the effects of invasive plants on native and exotic resident animals. We found that invasive plants reduced the abundance of native, but not exotic, animals. This varied by animal phyla, with invasive plants reducing the abundance of native annelids and chordates, but not mollusks or arthropods. We found dissimilar impacts among “wet” and “dry” ecosystems, but not among animal trophic levels. Additionally, the impact of invasive plants increased over time, but this did not vary with animal nativity. Our review found that no studies considered resident nativity differences, and most did not identify animals to species. We call for more rigorous studies of invaded community impacts across taxa, and most importantly, explicit consideration of resident biogeographic origin. We provide an important first insight into how native and exotic species respond differently to invasion, the consequences of which may facilitate cascading trophic disruptions further exacerbating global change consequences to ecosystem structure and function.  相似文献
