Sex‐determination mechanisms vary both within and among populations of common frogs, opening opportunities to investigate the molecular pathways and ultimate causes shaping their evolution. We investigated the association between sex‐chromosome differentiation (as assayed from microsatellites) and polymorphism at the candidate sex‐determining gene Dmrt1 in two Alpine populations. Both populations harboured a diversity of X‐linked and Y‐linked Dmrt1 haplotypes. Some males had fixed male‐specific alleles at all markers (“differentiated” Y chromosomes), others only at Dmrt1 (“proto‐” Y chromosomes), while still others were genetically indistinguishable from females (undifferentiated X chromosomes). Besides these XX males, we also found rare XY females. The several Dmrt1 Y haplotypes differed in the probability of association with a differentiated Y chromosome, which we interpret as a result of differences in the masculinizing effects of alleles at the sex‐determining locus. From our results, the polymorphism in sex‐chromosome differentiation and its association with Dmrt1, previously inferred from Swedish populations, are not just idiosyncratic features of peripheral populations, but also characterize highly diverged populations in the central range. DNP (dynamic nuclear polarization)-enhanced solid-state NMR is employed to directly detect protein–DNA and protein–ATP interactions and identify the residue type establishing the intermolecular contacts. While conventional solid-state NMR can detect protein–DNA interactions in large oligomeric protein assemblies in favorable cases, it typically suffers from low signal-to-noise ratios. We show here, for the oligomeric DnaB helicase from Helicobacter pylori complexed with ADP and single-stranded DNA, that this limitation can be overcome by using DNP-enhanced spectroscopy. Interactions are established by DNP-enhanced 31P–13C polarization-transfer experiments followed by the recording of a 2D 13C–13C correlation experiment. The NMR spectra were obtained in less than 2 days and allowed the identification of residues of the motor protein involved in nucleotide binding. 相似文献

Twenty years ago, a group of French cell biologists merged two scientific clubs with the aim of bringing together researchers in the fields of Endocytosis and Exocytosis. Founded in 1997, the first annual meeting of the Exocytosis Club was held in 1998. The Endocytosis Club held quarterly meetings from its founding in 1999. The first joint annual meeting of the Exocytosis–Endocytosis Club took place in Paris in April, 2001. What started as a modest gathering of enthusiastic scientists working in the field of cell trafficking has gone from strength to strength, rapidly becoming an unmissable yearly meeting, vividly demonstrating the high quality of science performed in our community and beyond. On the occasion of the 20th meeting of our club, we want to provide historic insight into the fields of exocytosis and endocytosis, and by extension, to subcellular trafficking, highlighting how French scientists have contributed to major advances in these fields. Today, the Exocytosis–Endocytosis Club represents a vibrant and friendly community that will hold its 20th meeting at the Presqu'Ile de Giens, near Toulon in the South of France, on May 11–13, 2017. 相似文献

Two plant fossil‐bearing beds from the middle Barremian of Belgium were analysed to ascertain how experimental designs affect conclusions regarding palaeodiversity at a local scale. We analysed eight lateral samples per bed taken regularly every 3 m using an exhaustive sub‐sampling method. The Clench equation was used to evaluate the completeness of the taxonomic inventory of the samples and the sampling effort needed to obtain a reliable representation of diversity. The number of replicates needed to obtain the same representation of diversity from different nearby lateral samples of the same bed ranged from 5 to 19. Richness (S), Evenness (J) and the number of equiprobable taxa (2H’) greatly varied between samples from the same bed, even over short distances. Only one of the studied samples was representative of the taxonomic inventory of its bed. Our study shows that 1) the selection bias of the sampling area is reduced by increasing the number of lateral samples taken in a bed, enabling more reliable conclusions about local‐scale diversity; 2) intense sub‐sampling methods are needed to account for statistically independent observations of detailed lateral variation; and 3) sampling methods in palaeodiversity analyses must look for a similar degree of representativeness in samples rather than a homogeneous sample size. Using a sampling effort analysis provides evidence for the completeness of the data set, adjusting the amount of work required. Implementing the Clench equation in palaeodiversity analyses improves the performance of data acquisition in palaeoecological studies and provides a quality test of the data sets derived from them. 相似文献
