Propensity scoring (PS) is an established tool to account for measured confounding in non-randomized studies. These methods are sensitive to missing values, which are a common problem in observational data. The combination of multiple imputation of missing values and different propensity scoring techniques is addressed in this work. For a sample of lymph node-positive vulvar cancer patients, we re-analyze associations between the application of radiotherapy and disease-related and non-related survival. Inverse-probability-of-treatment-weighting (IPTW) and PS stratification are applied after multiple imputation by chained equation (MICE). Methodological issues are described in detail. Interpretation of the results and methodological limitations are discussed. 相似文献

Campylobacter jejuni and Campylobacter coli are zoonotic pathogens once considered asaccharolytic, but are now known to encode pathways for glucose and fucose uptake/metabolism. For C. jejuni, strains with the fuc locus possess a competitive advantage in animal colonization models. We demonstrate that this locus is present in > 50% of genome‐sequenced strains and is prevalent in livestock‐associated isolates of both species. To better understand how these campylobacters sense nutrient availability, we examined biofilm formation and chemotaxis to fucose. C. jejuni NCTC11168 forms less biofilms in the presence of fucose, although its fucose permease mutant (fucP) shows no change. In a newly developed chemotaxis assay, both wild‐type and the fucP mutant are chemotactic towards fucose. C. jejuni 81‐176 naturally lacks the fuc locus and is unable to swim towards fucose. Transfer of the NCTC11168 locus into 81‐176 activated fucose uptake and chemotaxis. Fucose chemotaxis also correlated with possession of the pathway for C. jejuni RM1221 (fuc+) and 81116 (fuc‐). Systematic mutation of the NCTC11168 locus revealed that Cj0485 is necessary for fucose metabolism and chemotaxis. This study suggests that components for fucose chemotaxis are encoded within the fuc locus, but downstream signals only in fuc + strains, are involved in coordinating fucose availability with biofilm development. 相似文献

A rhizobacterial population of 2430 Pseudomonas isolates, originating from one maize hybrid and from its parents, was screened for auxins production. Four hundred and twelve isolates were found to be auxin producers (aia+), and 27 of them were also part of a previously described PhlD+ sub-population. Interestingly, most part of the aia(+)-PhlD+ isolates came from the hybrid. This finding indicates that heterosis allows an increased colonisation by multi-beneficial PGPR strains. Furthermore, results on the abundance and genetic diversity of aia+ isolates gave evidence that maize root colonisation by aia+ Pseudomonas is an inherited trait regulated by heterosis. In fact, two times more aia+ isolates were obtained from the rhizosphere of the hybrid than from the rhizospheres of the parents, and an amplified rDNA restriction analysis showed that the hybrid increases the genetic diversity of aia+ populations when compared to its parents. 相似文献

In Lake Lucerne, Switzerland, the predaceous cladocerans Leptodora kindti and Bythotrephes longimanus segregate along spatial and temporal dimensions. In spring (April–May/June), Bythotrephes longimanus occurs below 0–20 m, while Leptodora is absent. In summer and early autumn (July–September/October), when Leptodora dominates during daytime in the 0–20 m depth, Bythotrephes longimanus also lives in deeper zones. Food competition and fish predation pressure may be the cause of differences in ecology of Leptodora and Bythotrephes acquired during evolution. Due to its transparency and tolerance of higher temperature, Leptodora could avoid fish predation and, therefore, competes with Bythotrephes longimanus successfully. In addition, the differences between the two species may account for the spatial and temporal niche segregation in oligotrophic Swiss Lakes. But spatial niche segregation is less important in mesotrophic lakes with high prey density than in oligotrophic lakes with low prey density. In small, eutrophic lakes importance of temporal niche segregation also decreases, and Bythotrephes is seldom or not present. The preference of Bythotrephes to live in deeper water to avoid fish predation during summer may be the cause of its difficulties to establish itself in small and eutrophic lakes with high prey densities, where the hypolimnion is missing or anoxic.In the spring, Bythotrephes exhibits r-strategy (smaller body size and a higher fecundity), the female is already fertile after the first molt. In the summer, a K-strategy prevails (larger body length and lower fecundity than in the spring), and female Bythotrephes are fertile only after the second molt. Shortage of prey (biomass of Bosmina and Daphniadecreased after June especially in the surface layers) and the maximum fish predation pressure in summer may change the life strategy of Bythotrephes: while fecundity decreases from generation to generation, body length increases. Enhanced prey densities (e.g. during mesotrophic conditions in L. Lucerne) lead to larger individuals in summer and autumn. 相似文献

Approximately 150 ant species are facultatively or obligately queenless whereby mated workers assume the role of the queen.
In many of these species a reproductive dominance hierarchy is established by way of aggressive interactions. Top-ranking
workers, which are typically the most fecund, acquire a characteristic cuticular hydrocarbon profile. We studied the temporal
dynamics of this chemical change and associated interplay with observed aggressive interactions in an experimentally orphaned
colony of the facultatively queenless ant Gnamptogenys striatula. Our observations and chemical analyses demonstrate that chemical fertility signals played a major role in the establishment
of a dominance hierarchy and aggression settled dominance relationships only when ants had identical hydrocarbon profiles.
Moreover, individuals with a higher potential fertility, in this experiment reflected in a higher ovariole number, are shown

Background
Roux-en-Y gastric bypass (RYGB) surgery is associated with weight loss, improved insulin sensitivity and glucose homeostasis, and a reduction in co-morbidities such as diabetes and coronary heart disease. To generate further insight into the numerous metabolic adaptations associated with RYGB surgery, we profiled serum metabolites before and after gastric bypass surgery and integrated metabolite changes with clinical data.Methodology and Principal Findings
Serum metabolites were detected by gas and liquid chromatography-coupled mass spectrometry before, and 3 and 6 months after RYGB in morbidly obese female subjects (n = 14; BMI = 46.2±1.7). Subjects showed decreases in weight-related parameters and improvements in insulin sensitivity post surgery. The abundance of 48% (83 of 172) of the measured metabolites changed significantly within the first 3 months post RYGB (p<0.05), including sphingosines, unsaturated fatty acids, and branched chain amino acids. Dividing subjects into obese (n = 9) and obese/diabetic (n = 5) groups identified 8 metabolites that differed consistently at all time points and whose serum levels changed following RYGB: asparagine, lysophosphatidylcholine (C18:2), nervonic (C24:1) acid, p-Cresol sulfate, lactate, lycopene, glucose, and mannose. Changes in the aforementioned metabolites were integrated with clinical data for body mass index (BMI) and estimates for insulin resistance (HOMA-IR). Of these, nervonic acid was significantly and negatively correlated with HOMA-IR (p = 0.001, R = −0.55).Conclusions
Global metabolite profiling in morbidly obese subjects after RYGB has provided new information regarding the considerable metabolic alterations associated with this surgical procedure. Insulin increases muscle and fat cell glucose uptake by inducing the translocation of glucose transporter GLUT4 from intracellular compartments to the plasma membrane. Here, we have demonstrated that in 3T3-L1 adipocytes, DMSO at concentrations higher than 7.5% augmented cell surface GLUT4 levels in the absence and presence of insulin, but that at lower concentrations, DMSO only enhanced GLUT4 levels in insulin-stimulated cells. At a 5% concentration, DMSO also increased cell surface levels of the transferrin receptor and GLUT1. Glucose uptake experiments indicated that while DMSO enhanced cell surface glucose transporter levels, it also inhibited glucose transporter activity. Our studies further demonstrated that DMSO did not sensitize the adipocytes for insulin and that its effect on GLUT4 was readily reversible (t1/2∼12 min) and maintained in insulin-resistant adipocytes. An enhancement of insulin-induced GLUT4 translocation was not observed in 3T3-L1 preadipocytes and L6 myotubes, indicating cell specificity. DMSO did not enhance insulin signaling nor exocytosis of GLUT4 vesicles, but inhibited GLUT4 internalization. While other chemical chaperones (glycerol and 4-phenyl butyric acid) also acutely enhanced insulin-induced GLUT4 translocation, these effects were not mediated via changes in GLUT4 endocytosis. We conclude that DMSO is the first molecule to be described that instantaneously enhances insulin-induced increases in cell surface GLUT4 levels in adipocytes, at least in part through a reduction in GLUT4 endocytosis. 相似文献
