Separation and visualization of apolipoprotein B species by sodium dodecyl sulfate-agarose gel electrophoresis and immunoblotting

A method for separation and visualization of the different apolipoprotein B species using 0.2% sodium dodecyl sulfate-1.5% agarose gel electrophoresis and immunoblotting is described. The method is capable of demonstrating the different forms of apolipoprotein B (apo B) in plasma volumes of 10-50 microliters without prior ultracentrifugation. After ultracentrifugation of samples, estimation of the ratio between apo B 48 and apo B 100 is possible by scanning of Coomassie-stained gels or immunoblots.     

Age-related dynamics of thalamic-cortical evoked potentials in the rabbit in the early period of life

Data have been obtained on development of evoked potentials in the sensorimotor cortex to electrical stimulation of the thalamic ventroposterolateral nucleus (VPL) in rabbits in early ontogeny. In 3-5 days rabbits, under four times increase of threshold electric stimulation of VPL the thalamocortical response (TCR) is presented by a positive-negative potential with a long latency and minimum amplitude parameters. Second and third TCR positive components to increasing of threshold value of electric stimulation 4 times, are differentiated from 7-8 days age. Age dynamics of TCR amplitude-temporal parameters is characterized by a shortening of latency and an increase of oscillations amplitude, most expressed at 2-3 weeks of postnatal life. TCR of one month rabbit to increased threshold electrical stimulation of VPL is presented by short-latency positive-negative oscillation with a positive phase consisting of three components with successively increasing amplitudes.     

Characterization of the region on protein L7/L12 involved in binding to ribosomal particles

Tryptic digestion of reductively methylated protein L7/L12 yields a large tryptic fragment, which comprises amino acids 1-59. At the most, two molecules of this fragment can bind to a 50-S ribosomal particle, deprived of protein L7/L12. Besides, binding of each single 1-59 fragment competes with binding of one dimeric L7/L12 molecule. Molecular weight studies on the fragment reveal a monomeric structure. Digestion of the 1-59 fragment with carboxypeptidase Y leads to the formation of a 1-55 fragment. The binding characteristics of the latter fragment are similar to those of the 1-59 fragment. The results suggest that a monomeric stretch of L7/L12, comprising the first 55 amino acids, is sufficient for attaching L7/L12 to the ribosome.     

Use of an artificial antigen simulating the Salmonella O-determinant 4 for determining antibodies to Salmonella group B O-antigen in immunoenzyme analysis

The use of the artificial antigen abequosylmannoside copolymer with acrylamide in the enzyme immunoassay for the determination of antibodies in the sera of salmonellosis patients has enhanced the specificity of the serological diagnosis of group B salmonellosis in comparison with the use of the natural antiren, S. typhimurium lipopolysaccharide.     

Spore age and coverslip placement affects germination and post-germination development of Colletotrichum dematium var circinans

Spore suspensions from young (10–14 da; young spores) and old (4 mo; old spores) colonies of PColletotrichum dematium var circinans were placed on slides. Coverslips were left off, placed on in the normal manner, or supported on shims. Slides were placed in moist chambers and incubated in light or dark for up to 48 hrs. Germination and post-germination development were studied. Shimming had some beneficial effect on germination, especially for old spores in dark. In general, more germ-tubes and appressoria were produced on spores under shims than spores with other coverslip treatments. By 48 hrs more old spores under shims germinated, and greater numbers of germ-tubes and appressoria were produced than on other old spores under different coverslip treatments. However, numbers produced were lower than those predicted for comparably treated young spores. Spore age, incubation regime, and placement of coverslips did not affect germ-tube initiation. For all treatments more germ-tubes were initiated from spore tops than bottoms or tips. Fewer germ-tubes were initiated from spore centers than other locations on tops and bottoms, and from both tips than one tip. Approximately 26 % of all appressoria were produced sessile. A higher percentage of sessile appressoria were produced on old spores (80 %) than on young spores (20%).
