Elicitors of defence response can modulate pathways other than those related to pathogen attacks. In this paper, we demonstrate that, as with sugars, chitosan and oligogalacturonides (OGs) can repress the gibberellin signalling pathway leading to the induction of -amylase in barley embryos. These results are suggestive of a complex cross-talk between the defence, hormonal and metabolic signalling pathways.  相似文献

The human serum vitamin D-binding protein (DBP) has many physiologically important functions, ranging from transporting vitamin D3 metabolites, binding and sequestering globular actin and binding fatty acids to functioning in the immune system. Here we report the 2.3 A crystal structure of DBP in complex with 25-hydroxyvitamin D3, a vitamin D3 metabolite, which reveals the vitamin D-binding site in the N-terminal part of domain I. To more explicitly explore this, we also studied the structure of DBP in complex with a vitamin D3 analog. Comparisons with the structure of human serum albumin, another family member, reveal a similar topology but also significant differences in overall, as well as local, folding. These observed structural differences explain the unique vitamin D3-binding property of DBP.  相似文献

Abstract. We investigated the effects of dispersal limitation, diaspore density, soil chemistry, disturbance and mollusc herbivory on the abundance and distribution of the perennial forest herb Dentaria bulbifera (Brassicaceae). After experimental sowing of bulbils in originally empty patches, juveniles of Dentaria emerged in 83% of plots. The seventh year after sowing, plants still persisted and were reproducing in several plots, demonstrating that the species is dispersal‐limited. However, long‐term survival of transplants was higher at sites where Dentaria occurred naturally, and this difference increased with time after sowing. Thus, the importance of local factors may only be expressed fully several years after colonisation. Differences in soil chemistry and disturbance treatment were not associated with differences in patch suitability. Removal of molluscs, however, significantly increased recruitment, and the effects of a single molluscicide treatment persisted after four years. We conclude that dispersal is not the only limitation to the distribution of Dentaria, and that local factors, such as mollusc herbivory, are also crucial.  相似文献

Abstract— In slice preparations the exchange of dissolved substances between cells and incubation medium is delayed by diffusion through the extracellular space. The delay may seriously interfere with the study of membrane transport in terms of unidirectional fluxes across the cell membranes. A three-compartment serial model has been developed to describe exchange between slice and incubation medium. By aid of this model it is shown that the diffusion delay prevents determination of unidirectional fluxes for the two non-metabolizable glucose analogues 3-O-methylglucose and α-methyl-glucosidc. The membrane transport of the slowly transported α-methylglucoside can however be examined by aid of the model whereas the transport of 3-O-methylglucose is so rapid that it can not be examined with respect to Vmax Km and Kr. An attempt to determine these parameters will result in falsely large values which reflect extracellular diffusion and not membrane transport.  相似文献

Background

Facilitation is a promising implementation intervention, which requires theory-informed evaluation. This paper presents an exemplar of a multi-country realist process evaluation that was embedded in the first international randomised controlled trial evaluating two types of facilitation for implementing urinary continence care recommendations. We aimed to uncover what worked (and did not work), for whom, how, why and in what circumstances during the process of implementing the facilitation interventions in practice.

Methods

This realist process evaluation included theory formulation, theory testing and refining. Data were collected in 24 care home sites across four European countries. Data were collected over four time points using multiple qualitative methods: observation (372 h), interviews with staff (n?=?357), residents (n?=?152), next of kin (n?=?109) and other stakeholders (n?=?128), supplemented by facilitator activity logs. A combined inductive and deductive data analysis process focused on realist theory refinement and testing.

Results

The content and approach of the two facilitation programmes prompted variable opportunities to align and realign support with the needs and expectations of facilitators and homes. This influenced their level of confidence in fulfilling the facilitator role and ability to deliver the intervention as planned. The success of intervention implementation was largely dependent on whether sites prioritised their involvement in both the study and the facilitation programme. In contexts where the study was prioritised (including release of resources) and where managers and staff support was sustained, this prompted collective engagement (as an attitude and action). Internal facilitators’ (IF) personal characteristics and abilities, including personal and formal authority, in combination with a supportive environment prompted by managers triggered the potential for learning over time. Learning over time resulted in a sense of confidence and personal growth, and enactment of the facilitation role, which resulted in practice changes.

Conclusion

The scale and multi-country nature of this study provided a novel context to conduct one of the few trial embedded realist-informed process evaluations. In addition to providing an explanatory account of implementation processes, a conceptual platform for future facilitation research is presented. Finally, a realist-informed process evaluation framework is outlined, which could inform future research of this nature.

The high percentage of false positives generated by differential display (as high as 85%) has previously limited the potential of the method. This report describes an efficient methodology that enables false positives to be discarded prior to cloning, via reverse Northern analysis. This first step of the screening also allows the detection of putative low abundance differential clones. Following cloning, a second reverse Northern combined with partial DNA sequencing and RT-PCR detection allows isolation of all differential cDNAs including very low abundance clones. Use of the sequential screening procedure described here led to the isolation of novel tomato genes responding to the plant hormone ethylene while minimising labor and materials input.  相似文献

Exosomes are nanovesicles released by virtually all cells, which act as intercellular messengers by transfer of protein, lipid, and RNA cargo. Their quantitative efficiency, routes of cell uptake, and subcellular fate within recipient cells remain elusive. We quantitatively characterize exosome cell uptake, which saturates with dose and time and reaches near 100% transduction efficiency at picomolar concentrations. Highly reminiscent of pathogenic bacteria and viruses, exosomes are recruited as single vesicles to the cell body by surfing on filopodia as well as filopodia grabbing and pulling motions to reach endocytic hot spots at the filopodial base. After internalization, exosomes shuttle within endocytic vesicles to scan the endoplasmic reticulum before being sorted into the lysosome as their final intracellular destination. Our data quantify and explain the efficiency of exosome internalization by recipient cells, establish a new parallel between exosome and virus host cell interaction, and suggest unanticipated routes of subcellular cargo delivery.  相似文献
