Autism spectrum disorders (ASD) and schizophrenia are neurodevelopmental disorders for which recent evidence indicates an important etiologic role for rare copy number variants (CNVs) and suggests common genetic mechanisms. We performed cytogenomic array analysis in a discovery sample of patients with neurodevelopmental disorders referred for clinical testing. We detected a recurrent 1.4 Mb deletion at 17q12, which harbors HNF1B, the gene responsible for renal cysts and diabetes syndrome (RCAD), in 18/15,749 patients, including several with ASD, but 0/4,519 controls. We identified additional shared phenotypic features among nine patients available for clinical assessment, including macrocephaly, characteristic facial features, renal anomalies, and neurocognitive impairments. In a large follow-up sample, the same deletion was identified in 2/1,182 ASD/neurocognitive impairment and in 4/6,340 schizophrenia patients, but in 0/47,929 controls (corrected p = 7.37 × 10−5). These data demonstrate that deletion 17q12 is a recurrent, pathogenic CNV that confers a very high risk for ASD and schizophrenia and show that one or more of the 15 genes in the deleted interval is dosage sensitive and essential for normal brain development and function. In addition, the phenotypic features of patients with this CNV are consistent with a contiguous gene syndrome that extends beyond RCAD, which is caused by HNF1B mutations only.  相似文献

Background  

Extraintestinal pathogenic E. coli (ExPEC) represent a phylogenetically diverse group of bacteria which are implicated in a large range of infections in humans and animals. Although subgroups of different ExPEC pathotypes, including uropathogenic, newborn meningitis causing, and avian pathogenic E. coli (APEC) share a number of virulence features, there still might be factors specifically contributing to the pathogenesis of a certain subset of strains or a distinct pathotype. Thus, we made use of suppression subtractive hybridization and compared APEC strain IMT5155 (O2:K1:H5; sequence type complex 95) with human uropathogenic E. coli strain CFT073 (O6:K2:H5; sequence type complex 73) to identify factors which may complete the currently existing model of APEC pathogenicity and further elucidate the position of this avian pathoype within the whole ExPEC group.  相似文献

Plants of the fern Matteuccia struthiopteris from northern and southern populations in Norway were studied in a phytotron. Relative growth rate (RGR), growth period, and sporophyll production were measured under different photoperiod (12–24 h) day-length and temperature (9–21 °C) treatments. For the southern plants, there were no significant differences between the different light treatments, but for the northern plants there was a significant (p<0.01) linear increase in the mean maximum RGR with increasing day-length. Small, but statistically significant (p<0.01) differences were found between northern and southern plants when the mean maximum RGR-values were compared. When plants from different origin were exposed to different treatment, there were major differences between the populations in the production of sporophylls (p<0.0001). Southern plants produced in average three times more sporophylls than the northern plants, and they had also higher proportions of fertile plants. Diurnal alternating temperature treatments gave no significant (p>0.05) effect on the mean maximum RGR compared with constant temperatures, but they gave significantly higher production of sporophylls. In general, the northern plants had a higher temperature threshold (approximately 12 °C) for sporophyll production than the southern plants (approximately 9 °C). Plants exposed to 24 h with natural light were generally more often fertile than plants exposed to a shorter photoperiod. The mean maximum RGR-values and time needed to develop the fronds at the 9 °C treatment were fairly equal to those found under natural conditions close to the altitudinal distribution limit of M.struthiopteris in W Norway. In general, the investigation showed that the applied temperature and light treatments affected sporophyll production more than vegetative growth.  相似文献

A Synechococcus sp. strain PCC 7002 Fe bioreporter was constructed containing the isiAB promoter fused to the Vibrio harveyi luxAB genes. Bioreporter luminescence was characterized with respect to the free ferric ion concentration in trace metal-buffered synthetic medium. The applicability of the Fe bioreporter to assess Fe availability in the natural environment was tested by using samples collected from the Baltic Sea and from the high-nutrient, low-chlorophyll subarctic Pacific Ocean. Parallel assessment of dissolved Fe and bioreporter response confirmed that direct chemical measurements of dissolved Fe should not be considered alone when assessing Fe availability to phytoplankton.  相似文献

Nutrients are imported into developing legume seeds by mass flow through the phloem, and reach developing embryos following secretion from their symplasmically isolated coats. To sustain homeostasis of seed coat water relations, phloem-delivered nutrients and water must exit seed coats at rates commensurate with those of import through the phloem. In this context, coats of developing French bean seeds were screened for expression of aquaporin genes resulting in cloning PvPIP1;1, PvPIP2;2 and PvPIP2;3. These genes were differentially expressed in all vegetative organs, but exhibited their strongest expression in seed coats. In seed coats, expression was localized to cells of the nutrient-unloading pathway. Transport properties of the PvPIPs were characterized by expression in Xenopus oocytes. Only PvPIP2;3 showed significant water channel activity (Pos = 150-200 microm s(-1)) even when the plasma membrane intrinsic proteins (PIPs) were co-expressed in various combinations. Permeability increases to glycerol, methylamine and urea were not detected in oocytes expressing PvPIPs. Transport active aquaporins in native plasma membranes of seed coats were demonstrated by measuring rates of osmotic shrinkage of membrane vesicles in the presence and absence of mercuric chloride and silver nitrate. The functional significance of aquaporins in nutrient and water transport in developing seeds is discussed.  相似文献

Causes for rarity in plants are poorly understood. Graptophyllum reticulatum is an endangered endemic species, and it has three close relatives with different conservation status: the vulnerable G. ilicifolium, the rare G. excelsum, and the common G. spinigerum. Applied to the chlorophyll a fluorescence transient of leaves, the JIP test provides a Performance Index (PI) which quantifies the main steps in photosystem II (PSII) photochemistry including light energy absorption, excitation energy trapping, and conversion of excitation energy into electron flow. The PI is calculated from three components which depend on the reaction center density, the trapping efficiency, and the electron transport efficiency. PI was measured in the natural habitats of the four species and under artificially imposed environmental stresses in the glasshouse to determine whether conservation status was related to stress resilience. The results showed that soil type is unlikely to restrict the endangered G. reticulatum, vulnerable G. ilicifolium, or rare G. excelsum because PI was similar in plants grown in diverse soils in the glasshouse. Photoinhibition is likely to restrict the endangered G. reticulatum to shade habitats because PI was significantly reduced when plants were exposed to more than 15% ambient light in controlled experiments. Water availability may determine the location and distribution of the vulnerable G. ilicifolium and common G. spinigerum because PI was reduced more than 60% when plants were exposed to water stress. While the characteristics of their natural habitats correspond to and explain the physiological responses, there was no obvious relationship between conservation status and environmental resilience. PI can be used to monitor vigor and health of populations of plants in the natural habitat. In cultivation experiments PI responds to key environmental variables that affect the distribution of species with conservation significance.  相似文献

We have identified thioredoxins (Trx) of Malassezia sympodialis, a yeast involved in the pathogenesis of atopic eczema, and of Aspergillus fumigatus, a fungus involved in pulmonary complications, as novel IgE-binding proteins. We show that these Trx, including the human enzyme, represent cross-reactive structures recognized by serum IgE from individuals sensitized to M. sympodialis Trx. Moreover, all three proteins were able to elicit immediate-type allergic skin reactions in sensitized individuals, indicating a humoral immune response based on molecular mimicry. To analyze structural elements involved in these reactions, the three-dimensional structure of M. sympodialis Trx (Mala s 13) has been determined at 1.4-A resolution by x-ray diffraction analysis. The structure was solved by molecular replacement and refined to a crystallographic R factor of 14.0% and a free R factor of 16.8% and shows the typical Trx fold. Mala s 13 shares 45% sequence identity with human Trx and superposition of the solved Mala s 13 structure with those of human Trx reveals a high similarity with a root mean square deviation of 1.11 A for all Calpha atoms. In a detailed analysis of the molecular surface in combination with sequence alignment, we identified conserved solvent-exposed amino acids scattered over the surface in both structures which cluster to patches, thus forming putative conformational B cell epitopes potentially involved in IgE-mediated cross- and autoreactivity.  相似文献

Phosphatidic acid (PA) is a minor but important phospholipid that, through specific interactions with proteins, plays a central role in several key cellular processes. The simple yet unique structure of PA, carrying just a phosphomonoester head group, suggests an important role for interactions with the positively charged essential residues in these proteins. We analyzed by solid-state magic angle spinning 31P NMR and molecular dynamics simulations the interaction of low concentrations of PA in model membranes with positively charged side chains of membrane-interacting peptides. Surprisingly, lysine and arginine residues increase the charge of PA, predominantly by forming hydrogen bonds with the phosphate of PA, thereby stabilizing the protein-lipid interaction. Our results demonstrate that this electrostatic/hydrogen bond switch turns the phosphate of PA into an effective and preferred docking site for lysine and arginine residues. In combination with the special packing properties of PA, PA may well be nature's preferred membrane lipid for interfacial insertion of positively charged membrane protein domains.  相似文献
