Contributions to the Biomechanics of Plants. I. Stabilities of Plant Stems with Strengthening Elements of Different Cross-Sections Against Weight and Wind Forces*

Biophysical considerations allow estimates of the mechanical stresses on self-bearing vertical stems of plants. Even at moderate wind velocities the stresses induced by aerodynamic forces dominate over those induced by the own weight. Using polar coordinates, analytical expressions of cross-sectional area and axial second moment of area for centrisymmetric structures with symmetries threefold or higher are derived. Calculating the relative section modulus for various (centrisymmetric) arrangements of stabilizing structures leads to an estimate of the “mechanical effectivity” of these structures. If for plant stems, seen as composite materials, the second moments of area and the elastic moduli are known, the contribution of the different tissues to mechanical stability can be determined quantitatively. The mechanical design of early “vascular” land plants and of stems of (fossil) trees and lianas in different ontogenetic stages can be assessed.

In vitro biosynthesis of gamma-lipotropic hormone.

Sheep gamma-lipotropic hormone (gamma-LPH) is a pituitary polypeptide made of 58 amino acids and is formed of the first 58 residues of beta-lipotropic hormone (beta-LPH). The C-terminal portion (41-58) of gamma-LPH is identical with the structure of beta-melanophore-stimulating hormone (beta-MSH). We hypothetized in 1967 that beta-LPH could be the biological precursor of beta-MSH and that gamma-LPH could be an intermediate compound. We demonstrated in 1974 that beta-LPH is actively synthesized in the bovine pituitaries. We now studied the biosynthesis of gamma-LPH by monitoring the incorporation of radioactive amino acids in beef pituitary slices. We separated gamma-LPH from the other radioactive proteins with a method previously described. We characterized the radioactive proteins by ion-exchange chromatography, gel filtration and polyacrylamide gel electrophoresis. Our results show that radioactive gamma-LPH was actively synthesized. This gamma-LPH has all the chemical characteristics of nonradioactive gamma-LPH. However, in the conditions used, we were unable to demonstrate biosynthesis of beta-MSH. These results suggest that gamma-LPH is biosynthesized more slowly than beta-LPH and that the conversion into beta-MSH, if it exists, is a slow or subactive process in the species studied.     

Specific cleavage of beta-LPH and ACTH by tonin: release of an opiate-like peptide beta-LPH (61-78).

Tonin, a rat enzyme capable of cleaving angiotensinogen, the tetradecapeptide renin substrate and angiotensin I directly to antiotensin II is also shown to cleave beta-lipotropin into beta-LPH 1–50, 1–51, 51–60, 52–60, 61–78 and 79–91, thereby selectively releasing the opiate-like segment beta-LPH 61–78. Its action on ACTH was similar, releasing ACTH 1–8, 1–7, 3–8, 3–7 and 9–39. In both situations the cleavages are of a selective tryptic-chymotryptic type at specific arginine, phenylalanine residues. Comparison of the tonin cleavage with those of trypsin, trypsin in combination with citraconylation of the lysine residues of beta-LPH is made. The data presented show that tonin does not cleave Met-enkephalin and can be used as an enzyme to study the presence of endorphin-like sequences in polypeptides.
