A biosynthetic labeled peptide structurally related to the thymic peptide ubiquitin was first identified fortuitously in bovine pars intermedia cells in regard to its partial NH2 terminal amino acid sequence (Met 1, Leu 8, 15 and Lys 6, 11, 27, 29, 33) after a protein segment data bank search. A peptide with the same behavior on carboxymethylcellulose chromatography and polyacrylamide gel electrophoresis has been purified after labeling experiments in two areas of rat brain, hypothalamus and striatum, and in a mouse and a human ACTH-secreting pituitary tumors. It represents about 1 to 10% of the total labeled proteins in the various experiments. Its identity with the above mentioned bovine pituitary peptide was confirmed by microsequence analysis with respect to Met 1, Lys 6, 11 in hypothalmus, Met 1 in striatum, and Lys 6, 11, 27, 29, 33 in the two pituitary tumors. The availability of standard purified ubiquitin allowed us to show that labeled and cold peptides have the same electrophoretic mobility and elution volume on Sephadex G-50 chromatography this further confirms their identity. Possible interests of such a biosynthetic characterization of a ubiquitin-related peptide are discussed, particularly in view of the structural relationship of ubiquitin to the non histone component of nuclear protein A-24, and as a test of tissue viability and biosynthetic efficiency in our in vitro biosynthetic systems.  相似文献

In this paper, we present the amino-terminal sequence of rat tonin, an endopeptidase responsible for the conversion of angiotensinogen, the tetradecapeptide renin substrate, or angiotensin I to angiotensin II. It is shown that isoleucine and proline occupy the amino- and carboxy-terminal residues respectively. The N-terminal sequence analysis permitted the identification of 34 out of the first 40 residues of the single polypeptide chain composed of 272 amino acids. These results showed an extensive homology with the sequence of many serine proteases of the trypsin-chymotrypsin family. This information, coupled with the slow inhibition of tonin by diisopropylfluorophosphate, classified this enzyme as a selective endopeptidase of the active serine protease family.  相似文献

The rate of muscle cell fusion increases between 28 °C and 40 °C by a factor of 15 to 20. The formal activation energy of the fusion process changes abruptly at about 35 °C. This change is discussed in terms of a phase transition of the membrane lipids at 35 °C. In the presence of cholesterol or dipalmitoyllecithin the fusion rate decreases markedly. Increasing the temperature reverses the effects of cholesterol and dipalmitoyllecithin. These results are discussed in terms of interactions between membrane lipids.  相似文献

The interaction of Rh2(II)(acetate)4, cis-[Rh(III)(en)2Cl2] Cl (en = ethylenediamine) and [Rh(III) (NH3)5Cl]Cl2 with calf thymus DNA has been studied at various r values [formula; see text] and interaction times. Electronic spectra, melting and cooling curves and sedimentation data indicate no interaction of the acetate complex with DNA, except in the case of a high r value and long interaction time. The other two complexes have been found to interact with the phosphate groups, thus stabilizing the macromolecule.  相似文献

Climate change has profound ecological effects, yet our understanding of how trophic interactions among species are affected by climate change is still patchy. The sympatric Atlantic haddock and cod are co‐occurring across the North Atlantic. They compete for food at younger stages and thereafter the former is preyed by the latter. Climate change might affect the interaction and coexistence of these two species. Particularly, the increase in sea temperature (ST) has been shown to affect distribution, population growth and trophic interactions in marine systems. We used 33‐year long time series of haddock and cod abundances estimates from two data sources (acoustic and trawl survey) to analyse the dynamic effect of climate on the coexistence of these two sympatric species in the Arcto‐Boreal Barents Sea. Using a Bayesian state‐space threshold model, we demonstrated that long‐term climate variation, as expressed by changes of ST, affected species demography through different influences on density‐independent processes. The interaction between cod and haddock has shifted in the last two decades due to an increase in ST, altering the equilibrium abundances and the dynamics of the system. During warm years (ST over ca. 4°C), the increase in the cod abundance negatively affected haddock abundance while it did not during cold years. This change in interactions therefore changed the equilibrium population size with a higher population size during warm years. Our analyses show that long‐term climate change in the Arcto‐Boreal system can generate differences in the equilibrium conditions of species assemblages.  相似文献

Availability of shelter is an important component of habitat selection for animals as it can influence survival (protection against harsh physical conditions and predation) and growth (energy acquisition and expenditure). Few studies address the effect of shelter on metabolic expenditures associated with non-mechanical tasks (excluding station holding or movement). The main goal of this study was to investigate the influence of shelter use on metabolic traits of smallmouth bass (Micropterus dolomieu) from two populations (Kiamika River and Lake Long). Respirometry experiments on smallmouth bass were conducted to measure standard metabolic rate (SMR), resting metabolic rate (RMR), aerobic scope (AS), recovery time (RT) and excess post-exercise oxygen consumption (EPOC) in the presence or absence of shelter. The presence of shelter did not affect most metabolic traits, except for RMR, which was reduced in the presence of shelter for Lake Long fish. The results of this study also show that larger fish had lower SMR in the presence of shelter than when it was absent. When accounting for social hierarchy, there were no differences in most metabolic traits in dominant or subordinate fish in the presence or absence of shelter, except for RT, which was significantly lower in the presence of shelter for dominant fish. These results do not support the existence of an unequivocal relationship between individual metabolic traits and the presence of shelter. If physiological motives may influence the use of shelter, sheltering in itself might not have important consequences on energy expenditures required for non-mechanical tasks.  相似文献

Habitat fragmentation is an increasing threat to wildlife species across the globe and it has been predicted that future biodiversity will decrease rapidly without the intervention of scientifically-based management. In this study we have applied a landscape genetics approach to suggest a network design that will maintain connectivity among populations of the endangered mountain Nyala (Tragelaphus buxtoni) in the fragmented highlands of Ethiopia. DNA was obtained non-invasively from 328 individuals and genetic population structure and gene flow were estimated using 12 microsatellite markers. In addition, a 475-bp segment of the mitochondrial control region was sequenced for 132 individuals. Potential dispersal corridors were determined from least-cost path analysis based on a habitat suitability map. The genetic data indicated limited gene flow between the sampled mountain Nyala populations of the Bale Massif and the Arsi Massif. The genetic differentiation observed among five sampling areas of the Bale Massif followed a pattern of isolation by distance. We detected no impact of habitat resistance on the gene flow. In the future, however, the current expanding human population in the highlands of Ethiopia may reduce the current mountain Nyala habitat and further limit migration. Hence, maintaining habitat connectivity and facilitating survival of stepping-stone populations will be important for the future conservation of the species. The approach used here may also be useful for the study and conservation of other wildlife species inhabiting areas of increasing human encroachment.  相似文献

Primates inhabiting human-modified landscapes often exploit matrix habitat to supplement their diet with cultivated foods, at times resulting in economic losses and conflict with local people. Understanding human-nonhuman primate interactions and the attitudes and perceptions of local people towards crop feeding species are crucial to designing effective species-based management plans. Over a 12-month period, we used scan sampling to study the consumption of cultivated foods and matrix use patterns by two habituated groups of Bale monkeys (Chlorocebus djamdjamensis), Ethiopian-endemic bamboo specialists, in two forest fragments (Kokosa and Afursa) set amidst human settlements and farmland in the southern Ethiopian Highlands. Further, we conducted interviews with local people to document their attitudes and perceptions towards Bale monkeys at the two sites. We found that Bale monkeys at Kokosa, a more degraded habitat by most measures, consumed significantly more cultivated foods than their counterparts at Afursa. Moreover, Bale monkeys at Kokosa spent significantly more time in the matrix than in the forest habitat, while monkeys at Afursa spent significantly less time in the matrix than in the forest habitat. Not surprisingly, local people displayed a more negative attitude towards monkeys inhabiting Kokosa than those inhabiting Afursa. The differences in Bale monkey cultivated food consumption and matrix use patterns—as well as in local people's attitudes and perceptions towards Bale monkeys—between Kokosa and Afursa are probably associated with differences in habitat structure, degree of habitat alteration, and land-use practices between the sites. We conclude that to ensure long-term coexistence between Bale monkeys and local people in human-modified landscapes, it is vital to incorporate nearby matrix habitats into management plans and to work closely with local communities to develop effective nonlethal crop protection strategies, thereby reducing the likelihood of negative interactions between Bale monkeys and humans.  相似文献

Nearly fledged Cory's Shearwater Calonectris diomedea chicks at Selvagem Grande occasionally desert their nest sites during the night, but generally return before dawn. The temporary use of other nest sites is rare and lasts less than two days. Almost all chicks from Selvagem Grande return to the island for their first breeding attempt; very few breed at islands outside the archipelago. On Selvagem Grande, almost half the males return to their native colony whereas more than 90% of females recruit to other colonies. Inter-colony movements sometimes exceed 1 km, but, if they return to their native colony, both sexes settle within 20 m of their natal site. From the second breeding attempt onwards, most birds of both sexes are faithful to the nest site. On average, less than 20% of birds desert their former nest site each year, with a shift of about 4 m only. Most nest shifts follow a breeding failure or a temporary respite from breeding, and are accompanied by a switch in partners.

Le poussin en fin de croissance du Puffin cendré Calonectris diomedea de Selvagem Grande déserte parfois son nid pendant la nuit, mais y revient généralement avant le jour. L'utilisation temporaire d'un autre site de nid est rare et dure moins de deux jours. Les jeunes adultes nés à Selvagem Grande reviennent presque toujours nicher sur l'île et extrêmement rarement en dehors de l'archipel. A Selvagem Grande, près de la moitié des mâles reviennent à leur colonie de naissance, mais moins de 10% des femelles. Le déplacement est parfois supérieur à 1 km mais, s'ils reviennent à leur colonie natale, les deux sexes s'établissent à moins de 20 m du nid où ils sont nés. A partir de leur deuxième reproduction, les deux sexes sont fidèles à leur nid. Moins de 20% des oiseaux changent de nid d'une année à l'autre, avec un déplacement de 4 m seulement, le plus souvent à la suite d'un échec ou d'une interruption temporaire de la reproduction, ce déplacement étant le plus souvent accompagne d'un changement de partenaire.  相似文献
