Eleven geographical isolates of the entomopathogenic fungus Nomuraea rileyi (Farlow) Samson of Helicoverpa armigera (Hubner)/Spodoptera litura (Fabricius) origin were studied for efficacy against the two host insects. Laboratory bioassays at a concentration of 2 x 10(8) conidia ml(-1) indicated that N. rileyi isolates of S. litura origin were better in terms of time taken for mycosis and mortality in both the test larvae: S. litura (77-80% mortality in 7 days) and H. armigera (79-85% in 8 days). Among the isolates of S. litura origin, geographical isolates from Hyderabad and Karimnagar were superior in terms of high percent kill as well as 100% germination of conidia within 48 h. Fastest germination was observed with Karimnagar isolate followed by Hyderabad isolate. Conidial yield was highest on barley-carrot extract-yeast extract medium. However in terms of material cost, barley-yeast extract medium was the lowest. The Karimnagar isolate of S. litura origin gave the highest conidial yield on barley-yeast extract medium. Chitinolytic enzyme profiles of different isolates revealed polymorphism in all the isolates from S. litura origin. Overall among the parameters studied the best traits were found in the Karimnagar isolate of S. litura origin. 相似文献

The effect of concentration of hydrophilic (hydroxypropyl methylcellulose [HPMC]) and hydrophobic polymers (hydrogenated castor
oil [HCO], ethylcellulose) on the release rate of tramadol was studied. Hydrophilic matrix tablets were prepared by wet granulation
technique, while hydrophobic (wax) matrix tablets were prepared by melt granulation technique and in vitro dissolution studies
were performed using United States Pharmacopeia (USP) apparatus type II. Hydrophobic matrix tablets resulted in sustained
in vitro drug release (>20 hours) as compared with hydrophilic matrix tablets (<14 hours). The presence of ethylcellulose
in either of the matrix systems prolonged the release rate of the drug. Tablets prepared by combination of hydrophilic and
hydrophobic polymers failed to prolong the drug release beyond 12 hours. The effect of ethylcellulose coating (Surelease)
and the presence of lactose and HPMC in the coating composition on the drug release was also investigated. Hydrophobic matrix
tablets prepared using HCO were found to be best suited for modulating the delivery of the highly water-soluble drug, tramadol

Melioidosis is an emerging, potentially fatal disease caused by Burkholderia pseudomallei, which requires prolonged antibiotic treatment to prevent disease relapse. However, difficulties in laboratory diagnosis of melioidosis may delay treatment and affect disease outcomes. Isolation of B. pseudomallei from clinical specimens has been improved with the use of selective media. However, even with positive cultures, identification of B. pseudomallei can be difficult in clinical microbiology laboratories, especially in non-endemic areas where clinical suspicion is low. Commercial identification systems may fail to distinguish between B. pseudomallei and closely related species such as Burkholderia thailandensis. Genotypic identification of suspected isolates can be achieved by sequencing of gene targets such as groEL which offer higher discriminative power than 16S rRNA. Specific PCR-based identification of B. pseudomallei has also been developed using B. pseudomallei-specific gene targets such as Type III secretion system and Tat-domain protein. Matrix-assisted laser desorption ionization time-of-flight mass spectrometry, a revolutionary technique for pathogen identification, has been shown to be potentially useful for rapid identification of B. pseudomallei, although existing databases require optimization by adding reference spectra for B. pseudomallei. Despite these advances in bacterial identification, diagnostic problems encountered in culture-negative cases remain largely unresolved. Although various serological tests have been developed, they are generally unstandardized “in house” assays and have low sensitivities and specificities. Although specific PCR assays have been applied to direct clinical and environmental specimens, the sensitivities for diagnosis remain to be evaluated. Metabolomics is an uprising tool for studying infectious diseases and may offer a novel approach for exploring potential diagnostic biomarkers. The metabolomics profiles of B. pseudomallei culture supernatants can be potentially distinguished from those of related bacterial species including B. thailandensis. Further studies using bacterial cultures and direct patient samples are required to evaluate the potential of metabolomics for improving diagnosis of melioidosis. 相似文献
