We have isolated and sequenced 30 independent clones derived from MnlI digestion of purified mouse major (gamma) satellite DNA. These clones contained between 0.9 and 1.1 gamma monomeric units derived presumably from random chromosomal sources. Individual clones showed a mean deviation from the mouse consensus satellite sequence of 3.9%, with a range of 0.9-9.1%. Cleavage of total mouse LTK cell genomic DNA with three different restriction enzymes (HindIII, BglII, BamHI) that do not cut within satellite monomers, followed by Southern and pulsed-field gel electrophoretic analyses, showed that the majority of monomers were organized into largely uninterrupted arrays that varied from a minimum of 240 kb to greater than 2000 kb in length. We suggest that the high degree of conservation of the mouse gamma-satellite sequences throughout the mouse genome results from frequent recombinational exchange between nonhomologous chromosomes. Further, this same process, facilitated by the all-acrocentric constitution of the typical mouse karyotype, and the extremely long and homologous gamma-satellite arrays, may be related to the common occurrence of Robertsonian translocation in mouse. 相似文献

An issue that continues to impact digital forensics is the increasing volume of data and the growing number of devices. One proposed method to deal with the problem of “big digital forensic data”: the volume, variety, and velocity of digital forensic data, is to reduce the volume of data at either the collection stage or the processing stage. We have developed a novel approach which significantly improves on current practice, and in this paper we outline our data volume reduction process which focuses on imaging a selection of key files and data such as: registry, documents, spreadsheets, email, internet history, communications, logs, pictures, videos, and other relevant file types. When applied to test cases, a hundredfold reduction of original media volume was observed. When applied to real world cases of an Australian Law Enforcement Agency, the data volume further reduced to a small percentage of the original media volume, whilst retaining key evidential files and data. The reduction process was applied to a range of real world cases reviewed by experienced investigators and detectives and highlighted that evidential data was present in the data reduced forensic subset files. A data reduction approach is applicable in a range of areas, including: digital forensic triage, analysis, review, intelligence analysis, presentation, and archiving. In addition, the data reduction process outlined can be applied using common digital forensic hardware and software solutions available in appropriately equipped digital forensic labs without requiring additional purchase of software or hardware. The process can be applied to a wide variety of cases, such as terrorism and organised crime investigations, and the proposed data reduction process is intended to provide a capability to rapidly process data and gain an understanding of the information and/or locate key evidence or intelligence in a timely manner. 相似文献

Afenestrata koreana n. sp. collected from roots of bamboo (Phyllostachys pubescens) in Gyeongnam Province in the southern part of the Korean peninsula is described and illustrated. Its primary differentiating characteristics are a globose to subspherical body in adults with a prominent neck and terminal cone, thick cuticle, terminal vulva, and deep vagina. Fenestra, bullae, and underbridge are absent. The anus is on the immediate posterior side of the cone. Superficial small tubercules cover all the terminal cone area. The new species differs markedly from the other two known species in the genus, specifically in having three incisures in the lateral field of juveniles and a shorter stylet length in juveniles and adults. In order to produce suitable animal model systems for the analysis of possible synergistic interaction between hepatitis B virus (HBV) or the human papillomavirus (HPV) with other viral or chemical co-factors, we have produced transgenic mice carrying HBV, HPV16 or HPV18. Transfer of 3343 micro-injected one-cell (C57/BL6 X CBA) F1 embryos into pseudo-pregnant ICR recipients produced 181 pups. Seventeen of 151 of the pups analysed were found to be transgenics. Southern blot and slot blot analysis showed that the copy number of the transgenome in the transgenic mice ranged from 1-50 copies per cell. All the transgenomes analysed appeared to be in tandem repeats without detectable sequence rearrangements. The transgenome sequences were also stable on transmission to the F3 generation. On breeding, six of the ten transgenic mice analysed showed the Mendelian mode of inheritance with integration at single or multiple loci. Of the remaining four mice, three were mosaics and one was sterile. Analysis of the gene expression of the transgenic mice is currently in progress. 相似文献

Cytogenetic methods and molecular probes derived from the centromere and short arm of chromosome 14 were used to investigate the structural properties of a chromosome 14 variant. Results of GTL, CBG, Ag-NOR, and non-banded Giemsa staining of the chromosomes suggested the complete absence of the short arm and possibly a large part of the centromere. Negative in situ hybridisation with an alpha satellite III probe confirmed the absence of the arm; the detection of normal amounts of alpha satellite DNA, however, indicated retention of the centromeric domain. The natural occurrence of a human acrocentric variant lacking a short arm was thus established. Within the detection limits of the methods used, the results demonstrate that satellite III DNA is not essential for normal centromeric activity and allow us to exclude the presence of this satellite DNA within the centromere and proximal long-arm region of human chromosome 14. 相似文献
