Adiponectin is associated with obesity and insulin resistance. To date, there has been no genome-wide association study (GWAS) of adiponectin levels in Asians. Here we present a GWAS of a cohort of Korean volunteers. A total of 4,001 subjects were genotyped by using a genome-wide marker panel in a two-stage design (979 subjects initially and 3,022 in a second stage). Another 2,304 subjects were used for follow-up replication studies with selected markers. In the discovery phase, the top SNP associated with mean log adiponectin was rs3865188 in CDH13 on chromosome 16 (p = 1.69 × 10(-15) in the initial sample, p = 6.58 × 10(-39) in the second genome-wide sample, and p = 2.12 × 10(-32) in the replication sample). The meta-analysis p value for rs3865188 in all 6,305 individuals was 2.82 × 10(-83). The association of rs3865188 with high-molecular-weight adiponectin (p = 7.36 × 10(-58)) was even stronger in the third sample. A reporter assay that evaluated the effects of a CDH13 promoter SNP in complete linkage disequilibrium with rs3865188 revealed that the major allele increased expression 2.2-fold. This study clearly shows that genetic variants in CDH13 influence adiponectin levels in Korean adults.  相似文献

Respiratory chain complex I (NADH:ubiquinone oxidoreductase) deficiency is one of the most frequent causes of mitochondrial disease in humans. The activity of this complex can be confidently measured in most tissue samples, but not in cultured skin fibroblasts or circulating lymphocytes. Highly contaminating non-mitochondrial NADH-quinone oxidoreductase activity in fibroblasts and the limited access of substrates to complex I in lymphocytes hinder its measurement in permeabilized cells. Complex I assay in these cells requires the isolation of mitochondria, which in turn necessitates large quantities of cells and is not feasible when studying circulating lymphocytes. Here we report a simple method to measure complex I activity in a minute amount of either cell type. The procedure strongly reduces contaminating NADH:quinone oxidoreductase activity and permits measuring high rates of rotenone-sensitive complex I activity thanks to effective cell permeabilization.  相似文献

The oriental armyworm Mythimna separata is an agricultural insect pest in Eastern Asia. Mythimna separata moths have a high phototactic response to green (520 nm) light. The biological characteristics of insects living under light of a specific wavelength at night can change and, accordingly, Juvenile hormone (JH) levels may be influenced by this light. The present study evaluates changes in the total JH levels at different developmental stages (larvae, pupae and adults) of M. separata reared under green light with different exposure periods at night (or dark period). The results show that, when the exposure time per day of the green light at night is extended, the JH levels in the final‐instar larvae (22 days) and older age pupae (8 days) are significantly reduced, and the JH levels in earlier age pupae (4 days) and adults (3, 6 and 9 days) are significantly increased, compared with groups not exposed to green light. Additionally, the JH level of male moths significantly differs from that of the female moths. We suggest that the JH level of M. separata insects could be regulated by the green light at night (or dark period). The findings of the present study will help to explain the relationship between the light environment and biological characteristics in nocturnal moths.  相似文献

The appropriate regulation of the actin cytoskeleton is essential for cell movement, changes in cell shape, and formation of membrane protrusions like lamellipodia and filopodia. Moreover, several regulatory proteins affecting actin dynamics have been identified in the motile regions of cells. Here, we provide evidence for the involvement of SPIN90 in the regulation of actin cytoskeleton and actin comet tail formation. SPIN90 was distributed throughout the cytoplasm in COS-7 cells, but exposing the cells to platelet-derived growth factor (PDGF) caused a redistribution of SPIN90 to the cell cortex and the formation of lamellipodia (or membrane ruffles), both of which were dramatically inhibited in SPIN90-knockdown cells. In addition, the binding of the C terminus of SPIN90 with both the Arp2/3 complex (actin-related proteins Arp 2 and Arp 3) and G-actin activates the former, leading to actin polymerization in vitro. And when coexpressed with phosphatidylinositol 4-phosphate 5 kinase, SPIN90 was observed within actin comet tails. Taken these findings suggest that SPIN90 participates in reorganization of the actin cytoskeleton and in actin-based cell motility.  相似文献

1. Whether the ingestion of excess phenylalanine (Phe) or tyrosine (Tyr) in diets alters the release of cholecystokinin (CCK) into the blood was investigated in chickens. A single meal of control, Phe excess, or Tyr excess diets was given through a stomach tube, followed by the CCK determination with specific CCK-8 antibody. 2. Plasma CCK level increased significantly at 30 min after feeding the diets: about 2.5 times the basal level in the control group and about 3 times the basal level in both of the experimental groups. 3. The Phe excess diet significantly delayed crop emptying rates compared to the control diet, while the Tyr diet on the contrary enhanced the emptying rate. 4. Enhanced plasma CCK levels and delayed crop emptying by Phe excess diet are suggested to be associated with depressed food intake.  相似文献
