Gene amplified in squamous cell carcinoma (SCC) 1 (GASC1), also known as KDM4C/JMJD2C, encodes a histone demethylase that specifically demethylates lysine residues (H3K9, H3K36, and H1.4K26) and plays a crucial role in the regulation of gene expression as well as in heterochromatin formation. GASC1 is located at human chromosome 9p23–24, where frequent genomic amplification is observed in human esophageal cancer, and its aberrant expression is detected in a variety of human cancers, such as breast, colon, and prostate. Therefore, it is highly likely that GASC1 contributes to the genesis and/or development of cancer. However, there is a lack of direct evidence of GASC1 having an oncogenic function. In this study, we aimed to clarify the role of GASC1 in the skin SCC carcinogenesis. For this purpose, we generated Gasc1-heterozygous mice (Gasc1 +/−) with reduced expression of Gasc1. On the basis of our results, Gasc1 +/− mice displayed a significantly lower incidence and multiplicity of both benign and malignant tumors induced by the two-stage skin carcinogenesis protocol than wild-type mice. In addition, the volume of carcinoma was significantly lower in Gasc1 +/ mice. Consistent with these observations, knocking down of Gasc1 resulted in reduced cell viability of SCC cells in vitro. Our findings clearly demonstrated that GASC1 has an oncogenic role in skin carcinogenesis.

The involvement of the human CD4 molecule in T cell alloresponses to transfected HLA-DQ and -DR Ag was investigated with antibodies to defined CD4 epitopes. Anti-CD4 reagents inhibited T cell proliferation in a dose-dependent manner, and affected responses to HLA-DQ and -DR products equally well. As previously observed for conventional alloresponses, saturating concentrations of CD4 antibodies were required (and sufficient) for substantial blocking of T cell responses to transfected HLA-DQ and -DR products, and antibodies to a wide range of CD4 epitopes were inhibitory. In contrast to these results on class II-dependent T cell proliferation, MHC-independent T cell activation (via CD3 antibodies) was largely resistant to inhibition with the same dose range of CD4 mAb (provided that CD3 and CD4 reagents could not compete for the same class of FcR). This observation validates our conclusion that despite the substantial sequence differences between HLA-DQ and -DR heterodimers, the participation of CD4 epitopes in T cell responses to these molecules is conserved.  相似文献

The muscular system of the tarsier was first described by Burmeister (1846), who noted that brachial extensors (triceps complex) have six heads. The first three heads, respectively, correspond to the long, lateral and medial heads of the triceps brachii muscle in man. The fourth head is the anconaeus and the fifth is the dorsoepitrochlearis. Schultz (1984) divided the sixth head into two different parts (preaxial and postaxial) from the viewpoint of nerve supply. The present study found that the whole sixth head is innervated by the ulnar nerve, and we propose that it is recognized as the proximal and distal heads of the (preaxial) epitrochleoanconaeus muscle. The proximal head may have developed specially in the tarsier in addition to the distal head observed in other prosimians. It is thought to support the extension of the elbow joint and contribute to the tarsier's effective locomotion.  相似文献

Amino acid sequences of cytotoxin-like basic proteins (CLBPs), purified from the venoms of Formosan cobra (Naja naja atra) and Indian cobra (Naja naja), were reinvestigated. The determined sequences differed from those reported previously by Takechi et al. [(1985) Biochem. Int. 11, 795-802; (1987) Biochem. Int. 14, 145-152]. The sequence of CLBPs at residues 25-30 was found to be hydrophilic as compared with those of cytotoxins. The difference in the hydrophobicity of this region might be responsible for the difference in their cytotoxic activities.  相似文献
