Bovine oocytes were matured in culture with and without cumulus cells. The proportion of oocytes matured to metaphase-II 24 h after culture was not different between those matured with and without cumulus cells. When cultured oocytes were inseminated in vitro, high proportions (84 to 87%) of oocytes were penetrated, with no difference between those matured with and without cumulus cells. However, the proportion of oocytes penetrated at the male pronuclear stage was significantly higher in cumulus-intact than in cumulus-free oocytes (90 vs 31%). The proportion of oocytes cleaved beyond the 8 approximately 32-cell stage was also significantly higher in cumulus-intact than in cumulus-free oocytes. It is concluded that the cumulus cells may have an important function for normal cytoplasmic maturation of bovine oocytes in vitro.  相似文献

We describe a new scaffold-free three-dimensional (3D) cell culture model using cholesteryl ester based lyotropic liquid crystal (LC) substrates. Keratinocytes were deposited randomly on the LC surface where they self-assembled into 3D microtissues or keratinospheroids. The cell density required to form spheroids was optimized. We investigated cell viability using dead/live cell assays. The adhesion characteristics of cells within the microtissues were determined using histological sectioning and immunofluorescence staining. Fourier transform infrared spectroscopy (FTIR) was used to characterize the biochemistry of the keratinospheroids. We found that both cells and microtissues could migrate on the LC surface. The viability study indicated approximately 80% viability of cells in the microtissues up to 20 days of culture. Strong intercellular adhesion was observed in the stratification of the multi-layered microspheroids using field emission-scanning electron microscopy (FE-SEM) and histochemical staining. The cytoskeleton and vinculins of the cells in the microtissues were expressed diffusely, but the microtissues were enriched with lipids and nucleic acids, which indicates close resemblance to the conditions in vivo. The basic 3D culture model based on LC may be used for cell and microtissue migration studies in response to cytochemical treatment.  相似文献

Several GTPases are required for ribosome biogenesis and assembly. We recently identified rice (Oryza sativa) nuclear/nucleolar GTPase 2 (OsNug2), a YlqF/YawG family GTPase, as having a role in pre-60S ribosomal subunit maturation. To investigate the potential factors involved in regulating OsNug2 function, yeast two-hybrid screens were performed using OsNug2 as bait. Rice serine/threonine kinase 1 (OsSTK1) was identified as a candidate interacting protein. OsSTK1 appeared to interact with OsNug2 both in vitro and in vivo. OsSTK1 was found to have no effect on the GTP-binding activity of OsNug2; however, the presence of recombinant OsSTK1 in OsNug2 assay reaction mixtures increased OsNug2 GTPase activity. A kinase assay showed that OsSTK1 had weak autophosphorylation activity and strongly phosphorylated serine 209 of OsNug2. Using yeast complementation testing, we identified a GAL::OsNug2(S209N) mutation-harboring yeast strain that exhibited a growth-defective phenotype on galactose medium at 39 °C, which was divergent from that of a yeast strain harboring GAL::OsNug2. The intrinsic GTPase activity of OsNug2(S209N), which was found to be similar to that of OsNug2, was not fully enhanced upon weak binding of OsSTK1. Our findings indicate that OsSTK1 functions as a positive regulator of OsNug2 by enhancing OsNug2 GTPase activity. In addition, phosphorylation of OsNug2 serine 209 is essential for its complete function in biological functional pathway.  相似文献

GnRH antagonists can suppress luteinizing hormone and follicle‐stimulating hormone (FSH), with less initial stimulatory effect and lower risk of ovarian hyperstimulation syndrome. The effects of GnRH antagonists on embryonic implantation remain controversial. To evaluate the effects of GnRH antagonists, endometrial tissues were biopsied from 12 women with intracytoplasmic sperm injection treatment, in which four subjects undergoing controlled ovulation stimulation with rFSH and GnRH antagonist, four subjects with a GnRH agonist long protocol, and four natural cycle controls. After iTRAQ quantification analysis, 24 proteins showed differential expression between natural cycle and agonist treatment group and 39 proteins between natural cycle and antagonist treatment group. A total of seven proteins demonstrated differential expression only in antagonist treatment group. Bioinformatic analysis implied these proteins can function in cell processes including angiogenesis, cell proliferation, apoptosis, cell migration, and immune response. Furthermore, GnRH antagonist suppressed the function of GNAS and ANPEP, which were important for endometrial functions. Immunohistochemical staining showed that ANPEP was mainly localized in the human endometrial stroma, while ACO2, CDC5L, and GNAS were mainly localized in the glands. This study could provide insights into the effect of GnRH antagonists on the endometrium, and help optimize the embryo implantation and improve the success rate for GnRH antagonist protocol.  相似文献

Herpes simplex virus-1 is a large double-stranded DNA virus that is self-sufficient in a number of genome transactions. Hence, the virus encodes its own DNA replication apparatus and is capable of mediating recombination reactions. We recently reported that the catalytic subunit of the HSV-1 DNA polymerase (UL30) exhibits apurinic/apyrimidinic and 5′-deoxyribose phosphate lyase activities that are integral to base excision repair. Base excision repair is required to maintain genome stability as a means to counter the accumulation of unusual bases and to protect from the loss of DNA bases. Here we have reconstituted a system with purified HSV-1 and human proteins that perform all the steps of uracil DNA glycosylase-initiated base excision repair. In this system nucleotide incorporation is dependent on the HSV-1 uracil DNA glycosylase (UL2), human AP endonuclease, and the HSV-1 DNA polymerase. Completion of base excision repair can be mediated by T4 DNA ligase as well as human DNA ligase I or ligase IIIα-XRCC1 complex. Of these, ligase IIIα-XRCC1 is the most efficient. Moreover, ligase IIIα-XRCC1 confers specificity onto the reaction in as much as it allows ligation to occur in the presence of the HSV-1 DNA polymerase processivity factor (UL42) and prevents base excision repair from occurring with heterologous DNA polymerases. Completion of base excision repair in this system is also dependent on the incorporation of the correct nucleotide. These findings demonstrate that the HSV-1 proteins in combination with cellular factors that are not encoded by the virus are capable of performing base excision repair. These results have implications on the role of base excision repair in viral genome maintenance during lytic replication and reactivation from latency.Herpes simplex virus-1 (HSV-1)2 is a large double-stranded DNA virus with a genome of ∼152 kilobase pairs (for reviews, see Refs. 1 and 2). HSV-1 switches between lytic replication in epithelial cells and a state of latency in sensory neurons during which there is no detectable DNA replication (1). Viral DNA replication is mediated by seven essential virus-encoded factors (35). Of these, two encode subunits of the viral replicase (for review, see Refs. 6 and 7). The catalytic subunit (UL30) exhibits DNA polymerase (Pol), 3′-5′ proofreading exonuclease, and RNase H activities (811). UL30 exists as a heterodimer with the UL42 protein that confers a high degree of processivity on the Pol (1117).Viral DNA replication is accompanied by vigorous recombination that leads to the formation of large networks of viral DNA replication intermediates (18). The HSV-1 single-strand DNA-binding protein (ICP8) has been shown to play a major role in mediating these recombination reactions (1921). One role for the high frequency of recombination is to restart DNA replication at sites of fork collapse. Further mechanisms that contribute to genome maintenance are processes that survey and repair damage to the DNA to ensure the availability of a robust replication template. In this regard base excision repair (BER) is essential to remove unusual bases from the DNA and to repair apurinic/apyrimidinic (AP) sites resulting from spontaneous base loss (for review, see Ref. 22). With respect to HSV-1, a recent study showed that viral DNA from infected cultured fibroblasts contains a steady state of 2.8–5.9 AP sites per viral genome equivalent (23). Because AP sites are non-instructional, the failure to repair such sites would terminate viral replication. Indeed, UL30 cannot replicate beyond a model AP site (tetrahydrofuran residue) (23), indicating that the virus must enable a process to repair such lesions. In this regard HSV-1 possesses several enzymes that would safeguard from the accumulation of unusual bases, specifically uracil, and base loss. Hence, HSV-1 encodes a uracil DNA glycosylase (UDG) (UL2) as well as a dUTPase to reduce the pool of dUTP and prevent misincorporation by the viral Pol (24, 25). Moreover, we recently showed that the catalytic subunit of the viral Pol (UL30) exhibits AP and 5′-deoxyribose phosphate (dRP) lyase activities (26). The presence of a virus-encoded UDG and DNA lyase indicates that HSV-1 has the capacity to perform integral steps of BER, specifically for the removal of uracil. Indeed, the excision of uracil may be important for viral replication. Hence, it has been shown that uracil substitutions in the viral origins of replication alters their recognition by the viral initiator protein (27). Moreover, whereas UL2 may be dispensable for viral replication in fibroblast (24), UL2 mutants exhibit reduced neurovirulence and a decreased frequency of reactivation from latency (28). Thus, UDG action in HSV-1 may be important for viral reactivation after quiescence in neuronal cells during which the genome may accumulate uracil as a result of spontaneous deamination of cytosine. In another herpesvirus, cytomegalovirus, the viral UDG was shown to be required for the transition to late-phase DNA replication (29, 30). Consequently, it is possible that BER plays a significant role in various aspects of the herpesvirus life cycle.In mammalian single-nucleotide BER initiated by monofunctional DNA glycosylases, the resulting AP sites are incised hydrolytically at the 5′ side by AP endonuclease (APE), generating a 3′-OH. This is followed by template-directed incorporation of one nucleotide by Pol β to generate a 5′-dRP flap (22, 31, 32). The 5′-dRP residue is subsequently removed by the 5′-dRP lyase activity of Pol β to leave a nick with a 3′-OH and 5′-phosphate that is ligated by DNA ligase I or the physiologically more relevant ligase IIIα-XRCC1 complex (for review, see Refs. 33 and 34). Here we show that the HSV-1 UDG (UL2) and Pol (UL30) cooperate with human APE and human ligase IIIα-XRCC1 complex to perform BER in vitro. This finding has implications on the role of BER in viral genome maintenance during lytic replication and in the emergence of the virus from neuronal latency.  相似文献

Polyhydroxyalkanoates (PHAs) are biodegradable polymers produced by microbes to overcome environmental stress. Commercial production of PHAs is limited by the high cost of production compared to conventional plastics. Another hindrance is the brittle nature and low strength of polyhydroxybutyrate (PHB), the most widely studied PHA. The needs are to produce PHAs, which have better elastomeric properties suitable for biomedical applications, preferably from inexpensive renewable sources to reduce cost. Certain unique properties of Bacillus subtilis such as lack of the toxic lipo-polysaccharides, expression of self-lysing genes on completion of PHA biosynthetic process – for easy and timely recovery, usage of biowastes as feed enable it to compete as potential candidate for commercial production of PHA.  相似文献

The identification of a three dimensional constitutive model is useful for describing the complex mechanical behavior of a nonlinear and anisotropic biological tissue such as the esophagus. The inflation tests at the fixed axial extension of 1, 1.125, and 1.25 were conducted on the muscle and mucosa layer of a porcine esophagus separately and the pressure-radius-axial force was recorded. The experimental data were fitted with the constitutive model to obtain the structure-related parameters, including the collagen amount and fiber orientation. Results showed that a bilinear strain energy function (SEF) with four parameters could fit the inflation data at an individual extension very well while a six-parameter model had to be used to capture the inflation behaviors at all three extensions simultaneously. It was found that the collagen distribution was axial preferred in both layers and the mucosa contained more collagen, which were in agreement with the findings through a pair of uniaxial tensile test in our previous study. The model was expected to be used for the prediction of stress distribution within the esophageal wall under the physiological state and provide some useful information in the clinical studies of the esophageal diseases.  相似文献
