Physiological processes of terrestrial plants regulate the land–atmosphere exchange of carbon, water, and energy, yet few studies have explored the acclimation responses of mature boreal conifer trees to climate change. Here we explored the acclimation responses of photosynthesis, respiration, and stomatal conductance to elevated temperature and/or CO2 concentration ([CO2]) in a 3‐year field experiment with mature boreal Norway spruce. We found that elevated [CO2] decreased photosynthetic carboxylation capacity (?23% at 25 °C) and increased shoot respiration (+64% at 15 °C), while warming had no significant effects. Shoot respiration, but not photosynthetic capacity, exhibited seasonal acclimation. Stomatal conductance at light saturation and a vapour pressure deficit of 1 kPa was unaffected by elevated [CO2] but significantly decreased (?27%) by warming, and the ratio of intercellular to ambient [CO2] was enhanced (+17%) by elevated [CO2] and decreased (?12%) by warming. Many of these responses differ from those typically observed in temperate tree species. Our results show that long‐term physiological acclimation dampens the initial stimulation of plant net carbon assimilation to elevated [CO2], and of plant water use to warming. Models that do not account for these responses may thus overestimate the impacts of climate change on future boreal vegetation–atmosphere interactions. 相似文献

A putative advantage of allopolyploidy is the possibility of differential selection of duplicated (homeologous) genes originating from two different progenitor genomes. In this note we explore this hypothesis using a high throughput, SNP-specific microarray technology applied to seed trichomes (cotton) harvested from three developmental time points in wild and modern accessions of two independently domesticated cotton species, Gossypium hirsutum and G. barbadense. We show that homeolog expression ratios are dynamic both developmentally and over the several-thousand-year period encompassed by domestication and crop improvement, and that domestication increased the modulation of homeologous gene expression. In both species, D-genome expression was preferentially enhanced under human selection pressure, but for nonoverlapping sets of genes for the two independent domestication events. Our data suggest that human selection may have operated on different components of the fiber developmental genetic program in G. hirsutum and G. barbadense, leading to convergent rather than parallel genetic alterations and resulting morphology. 相似文献

Algae have been explored for renewable energy, nutraceuticals, and value-added products. However, low lipid yield is a significant impediment to its commercial viability. Genetic engineering can improve the fatty acid profile of algae without compromising its growth. This study introduced the diacylglycerol acyltransferase (BnDGAT) gene from Brassica napus into Chlorella sorokiniana-I, a fast-growing and thermotolerant natural strain isolated from wastewater, which increased its intracellular lipid accumulation. Hygromycin-resistant cells were selected, and enhanced green florescence protein fluorescence was used to distinguish pure transgenic cell lines from mixed cultures. Compared to the wild type, BnDGAT expression in transgenic C. sorokiniana-I caused a threefold increase in non-polar lipid and a twofold increase in polyunsaturated fatty acids. Nile red staining reaffirmed the presence of higher intracellular lipid bodies in transgenic cells. There was a substantial alteration in the fatty acid profile of transgenic alga expressing BnDGAT. The non-essential omega 9 (C18: 1) fatty acid decreased (5%–7% from 18%), while alpha-linolenic acid, an essential omega 3 fatty acid (C18: 3), was increased (23%–24% from 11%). This study substantiates a valuable strategy for enhancing essential omega-3 fatty acids and neutral lipids to improve its nutritional value for animal feed. The increased lipid productivity should reduce the cost of producing fatty acid methyl esters (FAME). Improved FAME quality should address the clouding issues in cold regions. 相似文献

Withdrawal of progestational support for pregnancy is part of the final common pathways for parturition, but the role of nuclear progesterone receptor (PGR) isoforms in this process is not known. To determine if the PGR-B isoform participates in cervical remodeling at term, cervices were obtained from mice lacking PGR-B (PGR-BKO) and from wild-type (WT) controls before or after birth. PGR-BKO mice gave birth to viable pups at the same time as WT controls during the early morning of Day 19 postbreeding. Morphological analyses indicated that by the day before birth, cervices from PGR-BKO and WT mice had increased in size, with fewer cell nuclei/area as well as diminished collagen content and structure, as evidenced by optical density of picrosirius red-stained sections, compared to cervices from nonpregnant mice. Moreover, increased numbers of resident macrophages, but not neutrophils, were found in the prepartum cervix of PGR-BKO compared to nonpregnant mice, parallel to findings in WT mice. These results suggest that PGR-B does not contribute to the growth or degradation of the extracellular matrix or proinflammatory processes associated with recruitment of macrophages in the cervix leading up to birth. Rather, other receptors may contribute to the progesterone-dependent mechanism that promotes remodeling of the cervix during pregnancy and in the proinflammatory process associated with ripening before parturition. 相似文献

The hypervariable D-loop region of mitochondrial DNA (mtDNA) was amplified with the polymerase chain reaction using total horse DNA samples. Analysis of single strand conformation polymorphism (SSCP) of denatured amplification products was carried out by native polyacrylamide (8%) gel electrophoresis followed by silver staining. As many as 15 distinct SSCP variants were revealed when screening a total of 78 maternally unrelated horses representing five different breeds. All breeds showed a high degree of polymorphism and the estimated probability (PImt ) that two maternally unrelated individuals have, by chance, identical SSCP variants varied between 0.14 and 0.30. We detected no heteroplasmy or deviations from strict and stable maternal inheritance when examining four maternal lineages, each represented by six to eight horses, separated by up to five generations from a common ancestral mare. The study establishes a simple screening method for detecting equine mtDNA types, which can be applied for tracing maternal genealogies and for association studies. 相似文献
